Claims
- 1. Method for the selection of human cell lines for the preparation of human proteins by endogenous activation of a target gene endogenously present in the cell, comprising(a) assaying a human cell line for the presence of the following features: (i) a target gene with the desired nucleic acid sequence, (ii) at least 5 population doublings within 14 days in a suspension culture, (iii) at least 5 population doublings within 14 days in a serum-free culture medium, (iv) the presence of more than 2 chromosomal copies of the target gene, and (b) selecting a cell line satisfying features (i), (ii), (iii) and (iv) as a starting cell line for the endogenous activation of the target gene.
- 2. Method according to claim 1, wherein said human cell line produces less than about 10 pg protein/106 cells/day.
- 3. Method according to claim 1, wherein said a human cell line synthesizes the cell protein with a glycosylation pattern comparable to the naturally occurring target protein.
- 4. Method according to claim 1, wherein said human cell line is free of detectable infectious contamination.
- 5. Method for the preparation of human protein by endogenous gene activation in a human cell line, comprising endogenously activating expression of a target gene in a human cell line, which has been selected by the method according to claim 1, wherein said target gene encodes a protein, and isolating said protein from said human cell line.
- 6. Method according to claim 5, wherein said human cell line furthermore satisfies at least one feature selected from the group consisting of:(iv) shows no substantial endogenous expression of the target gene; (v) synthesizes the protein with a glycosylation pattern comparable to the naturally occurring target protein; and, (vi) is free of detectable infectious contamination.
- 7. Method according to claim 5 for the preparation of a human factor selected from erythropoietin (EPO), thrombopoietin (TPO), colony stimulating factors, proteins which affect blood coagulation, interferons, interleukins, chemokines, neurotrophic factors, proteins which affect bone growth, hedgehog proteins, tumor growth factors, growth hormones, adrenocortico tropic hormone (ACTH), encephalins, endorphins, receptors and other protein-binding proteins.
- 8. Method according to claim 7 for the preparation of EPO.
- 9. The method of claim 5 further comprising culturing said selected cell line in a fermenter.
