Claims
- 1. A 1-R.sub.1 -1,2-dihydro-2-oxo-5-(3-R.sub.2 -4-R.sub.3 -phenyl)-6-R-nicotinonitrile having the formula ##STR5## or acid-addition salt thereof, where R.sub.1 is hydrogen, lower-alkyl or lower-hydroxyalkyl, R is hydrogen or lower-alkyl, R.sub.2 is hydrogen, amino or nitro and R.sub.3 is hydroxy.
- 2. A compound according to claim 1 where R.sub.2 is hydrogen and R is hydrogen, methyl or ethyl, or where R.sub.2 is nitro or amino and R is methyl or ethyl.
- 3. 1,2-Dihydro-5-(4-hydroxyphenyl)-6-methyl-2-oxonicotinonitrile.
- 4. 1,2-Dihydro-5-(4-hydroxyphenyl)-2-oxonicotinonitrile.
- 5. 1,2-Dihydro-5-(4-hydroxy-3-nitrophenyl)-6-methyl-2-oxonicotinonitrile.
- 6. 5-(3-Amino-4-hydroxyphenyl)-1,2-dihydro-6-methyl-2-oxonicotinonitrile or acid-addition salt thereof.
- 7. A cardiotonic composition for increasing cardiac contractility, said composition comprising a pharmaceutically acceptable carrier and, as the active component thereof, a cardiotonically effective amount of 1-R.sub.1 -1,2-dihydro-2-oxo-5-(3-R.sub.4 -R.sub.5 -phenyl)-6-R-nicotinonitrile of the formula ##STR6## or pharmaceutically acceptable acid-addition salt thereof, where R.sub.1 is hydrogen, lower-alkyl or lower-hydroxyalkyl, R is hydrogen or lower-alkyl, and, R.sub.4 and R.sub.5 are each hydrogen, amino or hydroxy, at least one of R.sub.4 and R.sub.5 being other than hydrogen or where R.sub.4 is nitro and R.sub.5 is hydroxy.
- 8. A composition according to claim 7 where R.sub.1 is hydrogen, R is methyl or ethyl, R.sub.4 is hydrogen, hydroxy or amino and R.sub.5 is hydroxy or hydrogen the latter only when R.sub.4 is other than hydrogen or where R.sub.4 is nitro, R.sub.5 is hydroxy, R.sub.1 is hydrogen and R is methyl or ethyl.
- 9. A composition according to claim 8 where the active component is 1,2-dihydro-5-(4-hydroxyphenyl)-6-methyl-2-oxonicotinonitrile.
- 10. The method for increasing cardiac contractility in a patient requiring such treatment which comprises administering orally or parenterally in a solid or liquid dosage form to such patient a cardiotonic composition according to claim 7.
- 11. The method according to claim 10 where R.sub.1 is hydrogen, R is methyl or ethyl, R.sub.4 is hydrogen, hydroxy or amino, and R.sub.5 is hydroxy or hydrogen the latter only when R.sub.4 is other than hydrogen or where R.sub.4 is nitro, R.sub.5 is hydroxy, R.sub.1 is hydrogen and R is methyl or ethyl.
- 12. The method according to claim 10 where the active component is 1,2-dihydro-5-(4-hydroxyphenyl)-6-methyl-2-oxonicotinonitrile.
