Claims
- 1. A method of treating allergic diseases in warm-blooded animals suffering from the same, which method comprises the systemic administration to warm blooded animals of an effective anti-allergic amount of compound of the formula: ##STR88## a pharmaceutically acceptable acid addition salt or a stereochemically isomeric form thereof, wherein:
- --A.sup.1 .dbd.A.sup.2 --A.sup.3 .dbd.A.sup.4 --represents a bivalent radical of the formula:
- --N.dbd.CH--CH.dbd.CH-- (a-1);
- --CH.dbd.N--CH.dbd.CH-- (a-2);
- --CH.dbd.CH--N.dbd.CH-- (a-3) or;
- --CH.dbd.CH--CH.dbd.N-- (a-4),
- wherein one or two hydrogen atoms in said radicals (a-1)-(a-4) may, each independently from each other, be replaced by halo, C.sub.1-6 alkyl, C.sub.1-6 alkyloxy, trifluoromethyl, or hydroxy;
- R represents hydrogen or C.sub.1-6 alkyl;
- R.sup.1 represents hydrogen, C.sub.1-10 alkyl, C.sub.3-6 cycloalkyl, Ar.sup.1, C.sub.1-6 alkyl substituted with one or two Ar.sup.1 radicals, or a radical of the formula --Alk--G--R.sup.2,
- wherein:
- Ar.sup.1 represents phenyl; phenyl substituted with 1, 2, or 3 substituents each independently selected from halo, hydroxy, nitro, cyano, trifluoromethyl, C.sub.1-6 alkyl, C.sub.1-6 alkyloxy, C.sub.1-6 alkylthio, mercapto, amino, mono- and di(C.sub.1-6 alkyl)amino, carboxyl, C.sub.1-6 alkyloxycarbonyl, and C.sub.1-6 alkylcarbonyl; thienyl; halothienyl; furanyl; C.sub.1-6 alkyl substituted furanyl; pyridinyl; pyrimidinyl; pyrazinyl; thiazolyl; imidazolyl; or imidazolyl substituted with C.sub.1-6 alkyl;
- Alk represents C.sub.1-6 alkanediyl;
- G represents O, S, or NR.sup.3, wherein R.sup.3 represents hydrogen, C.sub.1-6 alkylcarbonyl, C.sub.1-6 alkyloxycarbonyl, or Ar.sup.2 --C.sub.1-6 alkyl;
- Ar.sup.2 represents phenyl or phenyl substituted with 1, 2, or 3 substitutents each independently selected from halo, hydroxy, nitro, cyano, trifluoromethyl, C.sub.1-6 alkyl, C.sub.1-6 alkyloxy, C.sub.1-6 alkylthio, mercapto, amino, mono- and di(C.sub.1-6 alkyl)amino, carboxyl, C.sub.1-6 alkyloxycarbonyl and C.sub.1-6 alkylcarbonyl; and
- R.sup.2 represents hydrogen; C.sub.2-6 alkenyl; C.sub.2-6 alkenyl substituted with Ar.sup.2 ; C.sub.3-6 alkynyl; Ar.sup.1 ; C.sub.1-6 alkyl; or C.sub.1-6 alkyl substituted with Ar.sup.1, hydroxy, C.sub.1-6 alkyloxy, carboxyl, C.sub.1-6 alkyloxycarbonyl, Ar.sup.2 -oxycarbonyl, or Ar.sup.2 --C.sub.1-6 alkyloxycarbonyl, wherein Ar.sup.1 and Ar.sup.2 are as defined above;
- m represents a number having a value of from 1 to 4;
- n represents a number having value of from 1 to 2; and
- L represents a radical of the formula:
- --Alk--R.sup.4 (b-1);
- --Alk--Y--R.sup.5 (b-2); or
- --Alk--Z.sup.1 --(C.dbd.X)--Z.sup.2 --R.sup.6 (b-3)
- wherein:
- Alk is as defined above;
- Y represents O, S, Nr.sup.8, wherein R.sup.8 represents hydrogen, C.sub.1-6 alkyl, C.sub.1-6 alkylcarbonyl, or Ar.sup.1 -carbonyl, wherein Ar.sup.1 is as defined above;
- Z.sup.1 and Z.sup.2 each independently represent O, S, NR.sup.9, or a direct bond, wherein R.sup.9 represents hydrogen or C.sub.1-6 alkyl;
- X represents O, S, or NR.sup.10, wherein R.sup.10 represents hydrogen, C.sub.1-6 alkyl, or cyano; and
- R.sup.4, R.sup.5, and R.sup.6 represent Het,
- wherein Het represents:
- (i) an optionally substituted five- or six-membered heterocyclic ring containinged 1, 2, 3, or 4 heteroatoms selected from the group consisting of oxygen, sulfur, and nitrogen, provided that no more than two oxygens or sulfurs are present; or
- (ii) an optionally substituted five- or six-membered heterocyclic ring containing 1 2 heteroatoms selected from the group consisting of oxygen, sulfur, and nitrogen, being fused with an optionally substituted five- or six-membered ring through two ring carbon atoms or one ring carbon and one ring nitrogen atom, containing ion the remainder of the fused ring only carbon atoms; or
- (iii) an optionally substituted five- or six-membered heterocyclic ring containing 1 or 2 heteroatoms selected from the group consisting of oxygen, sulfur, and nitrogen, being fused with an optionally substituted five- or six membered heterocyclic ring through two ring carbon atoms or one ring carbon and one ring nitrogen atom, containing in the remainder of the fused ring 1 or 2 heteroatoms selected from the group consisting of oxygen, sulfur and nitrogen;
- wherein:
- Het may optionally be substituted with up to 4 substituents when Het is a monocyclic ring system, and wherein Het may optionally be substituted with up to 5 substituents when Het is a bicyclic ring system, said substituents of Het being selected from the group consisting of a bivalent radical .dbd.X; halo; isocyanato; isothiocyanato; nitro; cyano; trifluoromethyl; a radical of formula --A; a radical of formula --Y--A; or a radical of formula --A.sup.1 --C(.dbd.X)--Z.sup.2 --A;
- wherein:
- .dbd.X is as defined above;
- A represents hydrogen, Ar.sup.2, or C.sub.1-6 alkyl being optionally substituted with Ar.sup.2, C.sub.1-6 alkyloxy, Ar.sup.2 --O, hydroxy, or C.sub.1-6 alkyloxycarbonyl; and
- Y, Z.sup.1, and Z.sup.2 each have the meaning as defined above;
- provided that (i) when in the radical --Y--A, A is hydrogen, then Y is other than a direct bond, or (ii) when in the radical --Z.sup.1 --C(.dbd.X)--Z.sup.2 --A, A is hydrogen and Z.sup.1 is NR.sup.9, O, or S, then Z.sup.2 is other than O or S.
- 2. A method according to claim 1 wherein --A.sup.1 .dbd.A.sup.2 --A.sup.3 .dbd.A.sup.4 -- is a bivalent radical having a formula (a-1) through (a-4).
- 3. A method according to claim 1 wherein --A.sup.1 .dbd.A.sup.2 --A.sup.3 .dbd.A.sup.4 -- is a bivalent radical having the formula (a-5) or (a-6).
- 4. A method according to claim 1 wherein R.sup.1 is C.sub.1-6 alkyl substituted with Ar.sup.1 or wherein R.sup.1 is a radical --Alk--O--R.sup.2, said R.sup.2 being hydrogen, C.sub.1-6 alkyl, C.sub.2-6 alkenyl, C.sub.3-6 alkynyl or Ar.sup.1.
- 5. A method according to claim 4 wherein R represents hydrogen, m represents 1, and n represents 1.
- 6. A method according to claim 5 wherein R.sup.8 is hydrogen or C.sub.1-4 alkyl, X is O or S, and Z.sup.1 and Z.sup.2 are each independently NH or a direct bond.
- 7. A method according to claim 6 wherein R.sup.1 is an ethoxyethyl radical.
- 8. A compound of the formula: ##STR89## a pharmaceutically acceptable acid addition salt or a stereochemically isomeric form thereof, wherein:
- --A.sup.1 .dbd.A.sup.2 --A.sup.3 .dbd.A.sup.4 -- represents a bivalent radial of the formula:
- --N.dbd.CH--CH.dbd.CH-- (a-1);
- --CH.dbd.N--CH.dbd.CH-- (a-2);
- --CH.dbd.CH--N.dbd.CH-- (a-3); or
- --CH.dbd.CH--CH.dbd.N-- (a-4),
- wherein one or two hydrogen atoms in said radicals (a-1)-(a-4) may, each independently from each other, be replaced by halo, C.sub.1-6 alkyl, C.sub.1-6 alkyloxy, trifluoromethyl, or hydroxy;
- R represents hydrogen or C.sub.1-6 alkyl;
- R.sup.1 represents hydrogen, C.sub.1-10 alkyl, C.sub.3-6 cycloalkyl, Ar.sup.1, C.sub.1-6 alkyl substituted with one or two Ar.sup.1 radicals, or a radical of the formula --Alk--G--R.sup.2,
- wherein:
- Ar.sup.1 represents phenyl; phenyl substituted with 1, 2, or 3 substitutents each independently selected from halo, hydroxy, nitro, cyano, trifluoromethyl, C.sub.1-6 alkyl, C.sub.1-6 alkyloxy, C.sub.1-6 alkylthio, mercapto, amino, mono- and di(C.sub.1-6 alkyl)amino, carboxyl, C.sub.1-6 alkyloxycarbonyl, and C.sub.1-6 alkylcarbonyl; thienyl; halothienyl; furanyl; C.sub.1-6 alkyl substituted furanyl; pyridinyl; pyrimidinyl; pyrazinyl; thiazolyl; imidazolyl; or imidazolyl substituted with C.sub.1-6 alkyl;
- Alk represents C.sub.1-6 alkanediyl;
- G represents O, S, or NR.sup.3, wherein R.sup.3 represents hydrogen, C.sub.1-6 alkylcarbonyl, C.sub.1-6 alkyloxycarbonyl, or Ar.sup.2 --C.sub.1-6 alkyl;
- Ar.sup.2 represents phenyl or phenyl substituted with 1, 2, or 3 substitutents each independently selected from halo, hydroxy, nitro, cyano, trifluoromethyl, C.sub.1-6 alkyl, C.sub.1-6 alkyloxy, C.sub.1-6 alkylthio, mercapto, amino, mono- and di(C.sub.1-6 alkyl)amino, carboxyl, C.sub.1-6 alkyloxycarbonyl and C.sub.1-6 alkylcarbonyl; and
- R.sup.2 represents hydrogen; C.sub.2-6 alkynyl; C.sub.2-6 alkenyl substituted with Ar.sup.2 ; C.sub.3-6 alkynyl; Ar.sup.1 ; C.sub.1-6 alkyl; or C.sub.1-6 alkyl substituted with Ar.sup.1, hydroxy, C.sub.1-6 alkyloxy, carboxyl, C.sub.1-6 alkyloxycarbonyl, Ar.sup.2 -oxycarbonyl, or Ar.sup.2 --C.sub.1-6 alkyloxycarbonyl, wherein Ar.sup.1 and Ar.sup.2 are as defined above;
- m represents a number having a value of from 1 to 4;
- n represents a number having a value of from 1 to 2; and
- L represents a radical of the formula:
- --Alk--R.sup.4 (b-1);
- --Alk--Y--R.sup.5 (b-2); or
- --Alk--Z.sup.1 --(C.dbd.X)--Z.sup.2 --R.sup.6 (b-3),
- wherein:
- Alk is as defined above;
- Y represents O, S, NR.sup.8, wherein R.sup.8 represents hydrogen, C.sub.1-6 alkyl, C.sub.1-6 alkylcarbonyl, or Ar.sup.1 -carbonyl, wherein Ar.sup.1 is as defined above;
- Z.sup.1 and Z.sup.2 each independently represent O, S, NR.sup.9, or a direct bond, wherein R.sup.9 represents hydrogen or C.sub.1-6 alkyl;
- X represents O, S, or NR.sup.10, wherein R.sup.10 represents hydrogen, C.sub.1-6 alkyl, or cyano; and
- R.sup.4, R.sup.5, and R.sup.6 represent Het,
- wherein Het represents:
- (i) an optionally substituted five- or six-membered heterocyclic ring containinged 1, 2, 3, or 4 heteroatoms selected from the group consisting of oxygen, sulfur, and nitrogen, provided that no more than two oxygens or sulfurs are present; or
- (ii) an optionally substituted five- or six-membered heterocyclic ring containing 1 or 2 heteroatoms selected from the group consisting of oxygen, sulfur, and nitrogen, being fused with an optionally substituted five- or six-membered ring through two ring carbon atoms or one ring carbon and one ring nitrogen atom, containing in the remainder of the fused ring only carbon atoms; or
- (iii) an optionally substituted five- or six-membered heterocyclic ring containing 1 or 2 heteroatoms selected from the group consisting of oxygen, sulfur, and nitrogen, being fused with an optionally substituted five- or six-membered heterocyclic ring through two ring carbon atoms or one ring carbon and one ring nitrogen atom, containing in the remainder of the fused ring 1 or 2 heteroatoms selected from the group consisting of oxygen, sulfur and nitrogen;
- wherein:
- Het may optionally be substituted with up to 4 substitutents when Het is a monocyclic ring system, and wherein Het may optionally be substituted with up to 5 substituents when Het is a bicyclic ring system, said substituents of Het being selected from the group consisting of a bivalent radical .dbd.X; halo; isocyanato; isothiocyanato; nitro; cyano; trifluoromethyl; a radical of formula --A; a radical of formula --Y--A; or a radical of formula --Z.sup.1 --C(.dbd.X)--Z.sup.2 --A;
- wherein:
- .dbd.X is as defined above;
- A represents hydrogen, Ar.sup.2, or C.sub.1-6 alkyl being optionally substituted with Ar.sup.2, C.sub.1-6 alkyloxy, Ar.sup.2 --O, hydroxy, or C.sub.1-6 alkyloxycarbonyl; and
- Y, Z.sup.1, and Z.sup.2 each have the meaning as defined above;
- provided that (i) when in the radical --Y--A, A is hydrogen, then Y is other than a direct bond, or (ii) when in the radical --Z.sup.1 --C(.dbd.X)--Z.sup.2 --A, A is hydrogen and Z.sup.1 is NR.sup.9, O, or S, then Z.sup.2 is other than O or S.
- 9. A chemical compound according to claim 8 wherein R.sup.1 is C.sub.1-6 alkyl substituted with Ar.sup.1 or wherein R.sup.1 is a radical --Alk--O--R.sup.2, said R.sup.2 being hydrogen, C.sub.1-6 alkyl, C.sub.2-6 alkenyl, C.sub.3-6 alkynyl or Ar.sup.1.
- 10. A compound according to claim 9 wherein R represents hydrogen, m represents 1, and n represents 1.
- 11. A chemical compound according to claim 10 wherein R.sup.8 is hydrogen or C.sub.1-4 alkyl, X is O or S, and Z.sup.1 and Z.sup.2 are each independently NH or a direct bond.
- 12. A chemical compound according to claim 11 wherein R.sup.1 is an ethoxyethyl radical.
- 13. An anti-allergic composition comprising one or more pharmaceutical carriers and as active ingredient an anti-allergic effective amount of at least one compound as defined in claim 1.
- 14. An anti-allergic composition according to claim 13 wherein R.sup.1 is C.sub.1-6 alkyl substituted with Ar.sup.1 or wherein R.sup.1 is a radical --Alk--O--R.sup.2, said R.sup.2 being hydrogen, C.sub.1-6 alkyl, C.sub.2-6 alkenyl, C.sub.3-6 alkynyl or Ar.sup.1.
- 15. An anti-allergic composition according to claim 14 wherein R represents hydrogen, m represents 1, and n represents 1.
- 16. An anti-allergic composition according to claim 13 wherein R.sup.8 is hydrogen or C.sub.1-4 alkyl, X is O or S, and Z.sup.1 and Z.sup.2 are each independently NH or a direct bond.
- 17. An anti-allergic composition according to claim 16 wherein R.sup.1 is an ethoxyethyl radical.
- 18. A method according to claim 1 wherein the said compound is 3-(2-ethoxyethyl)-2-[[4-[2-(2-pyridinyl)ethyl]-1-piperazinyl]methyl]-3H-imidazo[4,5-b]pyridine (E)-2-butenedioate.
- 19. A compound according to claim 8 wherein the said compound is 3-(2-ethoxyethyl)-2-[[4-[2-(2-pyridinyl)ethyl]-1-piperazinyl]methyl]-3H-imidazo[4,5-b]pyridine (E)-2-butenedioate.
- 20. An anti-allergic composition according to claim 15 wherein the said compound is 3-(2-ethoxyethyl)-2-[[4-[2- (2-pyridinyl)ethyl]-1-piperazinyl]methyl]-3H-imidazo[4,5-b]pyridine (E)-2-butenedioate.
