Claims
- 1. A method for the determination of an analyte comprising the steps of:
- A. at a pH of 9 or less, contacting a sample of a liquid suspected of containing an analyte with a reducible compound of the structure CAR--R.sup.1).sub.n wherein CAR-- is a substituted or unsubstituted aromatic or quinone nucleus, R.sup.1 is a moiety which comprises a shiftable detectable species, and n is 1 or 2,
- provided said compound is capable of being reduced at said pH to release said shiftable detectable species, and
- further provided that when R.sup.1 is replaced with H, CAR--H).sub.n has an E.sub.1/2 of either at least about +100 mV when measured in water, or of at least about -650 mV when measured in acetonitrile, and
- B. detecting the detectable species released as a result of the presence of said analyte.
- 2. The method of claim 1 wherein said detectable species is a chromogen or fluorogen.
- 3. A method for the determination of an analyte comprising the steps of:
- A. at a pH of 9 or less, contacting a sample of a liquid suspected of containing an analyte with a reducible compound of the structure CAR--R.sup.1 wherein CAR-- is ##STR61## R.sup.2 and R.sup.4 are independently hydrogen, substituted or unsubstituted alkyl, substituted or unsubstituted aryl or an electron withdrawing group,
- R.sup.3 is R.sup.1, hydrogen, substituted or unsubstituted alkyl, substituted or unsubstituted aryl or an electron withdrawing group, or R.sup.3 and R.sup.4, taken together, represent the atoms necessary to complete a substituted or unsubstituted fused carbocyclic ring,
- R.sup.5 is substituted or unsubstituted alkylene of 1 or 2 carbon atoms,
- R.sup.6 is substituted or unsubstituted alkyl, substituted or unsubstituted cycloalkyl, substituted or unsubstituted heterocycle or substituted or unsubstituted aryl, provided that when FRAG is a fluorogen, R.sup.6 is methyl,
- Q is carbonyl or thiocarbonyl,
- FRAG is a shiftable detectable species which provides a detectable species when released from said reducible compound, and
- m is 0 or 1,
- provided that when R.sup.1 is replaced with H, CAR--H has an E.sub.1/2 of either at least about +100 mV when measured in water, or of at least about -650 mV when measured in acetonitrile, and
- B. detecting the detectable species released as a result of the presence of said analyte.
- 4. The method of claim 3 for the determination of a nonliving analyte in the presence of an interactive composition for said analyte.
- 5. The method of claim 3 for the determination of living cells in the presence of an electron transfer agent.
- 6. The method of claim 5 for the determination of bacterial cells.
- 7. The method of claim 5 wherein when said compound is reduced at about pH 7, at least about 50% of said FRAG moiety is released within about 30 minutes.
- 8. A method for providing a chemically or biologically useful moiety comprising, at a pH of 9 or less, reducing a reducible compound of the structure CAR--R.sup.1).sub.n wherein CAR-- is a substituted or unsubstituted aromatic or quinone nucleus, R.sup.1 is a moiety which comprises a shiftable detectable species, and n is 1 or 2,
- provided said compound is capable of being reduced at said pH to release said shiftable detectable species, and
- further provided that when R.sup.1 is replaced with H, CAR--H).sub.n has an E.sub.1/2 of either at least about +100 mV when measured in water, or at least about -650 mV when measured in acetonitrile.
