Claims
- 1. A apparatus for a glycosyltransferase-catalyzed synthesis of an oligosaccharide composition, from saccharide units and an acceptor moiety, said apparatus comprising reactor means containing a pair of glycosyltransferases and an acceptor which is one member selected from the group consisting of:
- a) an .alpha.-1,4 galactosyltransferase, a .beta.-1,3 N-acetylgalactosaminyltransferase and lactose or a lactoside;
- b) a .beta.-1,4 N-acetylgalactosaminyltransferase, a .beta.-1,3 galactosyltransferase and lactose or a lactoside;
- c) a .beta.-1,4 galactosyltransferase, a .beta.-1,3 galactosyltransferase and lactose or a lactoside;
- d) an .beta.-1,3 galactosyltransferase, a .beta.-1,3 N-acetylgalactosaminyltransferase and lactose or a lactoside;
- e) an .beta.-1 N-acetylgalactosaminyltransferase, a .beta.-1,3 galactosyltransferase and serine or threonine;
- f) a .beta.-1,6 N-acetylglucosaminyltransferase, a .beta.-1,4 galactosyltransferase and gal .beta.-1,3 galNAc .beta.-1-O-linked serine or gal .beta.-1,3 galNAc .alpha.-1-O-linked threonine;
- g) an .alpha.-1,4 N-acetylglucosaminyltransferase, a .beta.-1,4 glucuronyltransferase and heparins or heparan sulfates; and
- h) a .beta.-1,4 N-acetylgalactosaminyltransferase, a .beta.-1,3 glucuronyltransferase and chondroitin or chondroitin sulfate;
- wherein said glycosyltransferases are purified.
- 2. The apparatus of claim 1, wherein said reactor means comprises at least one reaction zone.
- 3. The apparatus of claim 1, wherein said reactor means comprises two reaction zones.
- 4. The apparatus of claim 1, wherein said reactor means comprises a plurality of reaction zones, each comprising a different one of said two glycosyltransferases.
- 5. The apparatus of claim 4, wherein means for purifying an intermediate product formed in each of said reaction zones from contaminants in a reaction mixture produced therein is situated in fluid communication in between said reaction zones.
- 6. The apparatus of claim 4, wherein at least one of said reaction zones comprises one of said glycosyltransferases immobilized onto a solid support.
- 7. The apparatus of claim 4, wherein at least one of said glycosyltransferases is immobilized onto a solid support.
- 8. The apparatus of claim 4, wherein both of said glycosyltransferases are immobilized onto a solid support.
- 9. The apparatus of claim 6 wherein an active site of said at least one of said glycosyltransferases immobilized onto said solid support is protected during immobilization.
- 10. The apparatus of claim 6 wherein an active site of each of said glycosyltransferases immobilized onto said solid support is protected during immobilization.
- 11. A apparatus for a glycosyltransferase-catalyzed synthesis of an oligosaccharide composition, from saccharide units and an acceptor moiety, said apparatus comprising reactor means containing a pair of glycosyltransferases and an acceptor which is one member selected from the group consisting of:
- a) an .alpha.-2,3 sialyltransferase, an .alpha.-1,4 fucosyltransferase and lactose or polylactosamine;
- b) an .alpha.-1,2 fucosyltransferaSe, an .alpha.-1,3 fucosyltransferase and polylactose or polylactosamine; and
- c) a .beta.-1,4 galactosyltransferase, an .alpha.-1,3 galactosyltransferase and galNAc or glcNAc;
- wherein said glycosyltransferases are purified.
- 12. The apparatus of claim 1 wherein said system comprises an .alpha.-2,3 sialyltransferase, an .alpha.-1,4 fucosyltransferase and lactose or polylactosamine.
- 13. The apparatus of claim 1 wherein said system comprises an .alpha.-1,4 galactosyltransferase, a .beta.-1,3 N-acetylgalactosaminyltransferase and lactose or a lactoside.
- 14. The apparatus of claim 1 wherein said system comprises a .beta.-1,4 N-acetylgalactosaminyltransferase, a .beta.-1,3 galactosyltransferase and lactose or a lactoside.
- 15. The apparatus of claim 1 wherein said system comprises a .beta.-1,4 galactosyltransferase, a .beta.-1,3 galactosyltransferase and lactose or a lactoside.
- 16. The apparatus of claim 1 wherein said system comprises an .alpha.-1,3 galactosyltransferase, a .beta.-1,3 N-acetylgalactosaminyltransferase and lactose or a lactoside.
- 17. The apparatus of claim 1 wherein said system comprises an .alpha.-1 N-acetylgalactosaminyltransferase, a .beta.-1,3 galactosyltransferase and serine or threonine.
- 18. The apparatus of claim 1 wherein said system comprises a .beta.-1,6 N-acetylglucosaminyltransferase, a .beta.-1,4 galactosyltransferase and gal .beta.-1,3 galNAc .alpha.-1-O-linked serine or gal .beta.-1,3 galNAc .alpha.-1-O-linked threonine.
- 19. The apparatus of claim 1 wherein said system comprises an .alpha.-1,4 N-acetylglucosaminyltransferase, a .beta.-1,4 glucuronyltransferase and heparins or heparan sulfates.
- 20. The apparatus of claim 1 wherein said system comprises a .beta.-1,4 N-acetylgalactosaminyltransferase, a .beta.-1,3 glucuronyltransferase and chondroitin or chondroitin sulfate.
- 21. The apparatus of claim 11 wherein said system comprises an .alpha.-1,2 fucosyltransferase, an .alpha.-1,3 fucosyltransferase and polylactose or polylactosamine.
- 22. The apparatus of claim 11 wherein said system comprises a .beta.-1,4 galactosyltranserase, an .alpha.-1,3 galactosyltransferase and galNAc or glcNAc.
- 23. An apparatus for a glycosyltransferasecatalyzed synthesis of an oligosaccharide composition, from saccharide units and an acceptor moiety, said apparatus comprising reactor means having inlet means, outlet means and containing a pair of glycosyltransferases and an acceptor which is one member selected from the group consisting of:
- a) an .beta.-1,4 galactosyltransferase, a .beta.-1,3 N-acetylgalactosaminyltransferase and lactose or a lactoside;
- b) a .beta.-1,4 N-acetylgalactosaminyltransferase, a .beta.-1,3 galactosyltransferase and lactose or a lactoside;
- c) a .beta.-1,4 galactosyltransferase, a .beta.-1,3 galactosyltransferase and lactose or a lactoside;
- d) an .alpha.-1,3 galactosyltransferase, a .beta.-1,3 N-acetylgalactosaminyltransferase and lactose or a lactoside;
- e) an .alpha.-1 N-acetylgalactosaminyltransferase, a .beta.-1,3 galactosyltransferase and serine or threonine;
- f) a .beta.-1,6 N-acetylglucosaminyltransferase, a .beta.-1,4 galactosyltransferase and gal .beta.-1,3 galNAc .alpha.-1-O-linked serine or gal .beta.-1,3 galNAc .alpha.-1-O-linked threonine;
- g) an .alpha.-1,4 N-acetylglucosaminyltransferase, a .beta.-1,4 glucuronyltransferase and heparins or heparan sulfates; and
- h) a .beta.-1,4 N-acetylgalactosaminyltransferase, a .beta.-1,3 glucuronyltransferase and chondroitin or chondroitin sulfate;
- wherein said glycosyltransferases are purified.
- 24. A apparatus for a glycosyltransferase-catalyzed synthesis of an oligosaccharide composition, from saccharide units and an acceptor moiety, said apparatus comprising reactor means having inlet means, outlet means and containing a pair of glycosyltransferases and an acceptor which is one member selected from the group consisting of:
- a) an .alpha.-2,3 sialyltransferase, an .alpha.-1,4 fucosyltransferase and lactose or polylactosamine;
- b) an .alpha.-1,2 fucosyltransferase, an .alpha.-1,3 fucosyltransferase and polylactose or polylactosamine; and
- c) a .beta.-1,4 galactosyltransferase, an .alpha.-1,3 galactosyltransferase and galNAc or glcNAc;
- wherein said glycosyltransferases are purified.
- 25. A apparatus for a glycosyltransferase-catalyzed synthesis of an oligosaccharide composition, from saccharide units and an acceptor moiety, said apparatus comprising reactor means containing:
- i) a pair of glycosyltransferases which are a .beta.-1,4 galactosyltransferase and an .alpha.-1,3 galactosyltransferase; and
- ii) an acceptor selected from the group consisting of GalNAc and GlcNAc,
- wherein said glycosyltransferases are purified.
