Claims
- 1. A compound of the formula ##STR42## wherein: z is 1;
- X is ##STR43## a is 1 or 2; R, R.sup.1, R.sup.3, R.sup.4, R.sup.5 and R.sup.6 are each independently hydrogen, alkyl or aralkyl;
- R.sub.n is hydrogen, alkyl, aralkyl, acetyl, propionyl, benzoyl, carbalkoxy, carbamyl, carbalkoxyalkyl, hydroxyalkyl, alkoxyalkyl or amidino;
- R.sup.5 groups on vicinal carbon atoms may together form a carbon-carbon double bond when a=2;
- R groups on vicinal carbon atoms may together form a carbon-carbon double bond when R.sub.n is hydrogen;
- R.sup.4 and R.sup.5 geminal groups may together form a spiro substituent, --(CH.sub.2).sub.d --, where d is 2 to 5;
- R.sup.5 and R.sup.6 together may form a carbon-nitrogen double bond when R.sup.3 is hydrogen and a=1;
- R.sup.2 is hydrogen or --(CH.sub.2).sub.c --Y;
- c is 1-3;
- Y is hydrogen, --OR.sub..alpha., --S--R.sub..alpha., or ##STR44## R.sub..alpha. is hydrogen, alkyl, cycloalkyl, acetyl, propionyl or benzoyl;
- R.sub..beta. is hydrogen or alkyl;
- R.sub..alpha. and R.sub..beta. together with the nitrogen to which they are attached may form a 3-7 member ring, which may also contain 0-2 additional hetero atoms, and which is selected from the group of ethyleneimine, trimethyleneamine, piperidine, piperazine, morpholine, thiomorpholine, N-methyl-piperazine, N-phenylpiperazine, N-benzylpiperazine and N-methyl-imidizolidine;
- or a pharmaceutically acceptable salt thereof.
- 2. A compound of the formula ##STR45## wherein: z is 1;
- X is ##STR46## b is 1 or 2; R, R.sup.1, R.sup.3, R.sup.4, R.sup.5 and R.sup.6 are each independently hydrogen, alkyl or aralkyl;
- R.sub.n is hydrogen, alkyl, aralkyl, acetyl, propionyl, benzoyl, carbalkoxy, carbamyl, carbalkoxyalkyl, hydroxyalkyl, alkoxyalkyl or amidino;
- R.sup.5 groups on vicinal carbon atoms may together form a carbon-carbon double bond when b=2;
- R groups on vicinal carbon atoms may together form a carbon-carbon double bond when R.sub.n is hydrogen;
- R.sup.4 and R.sup.5 geminal groups may together form a spiro substituent, --(CH.sub.2).sub.d --, where d is 2 to 5;
- R.sup.5 and R.sup.6 may together form a carbon-nitrogen double bond when R.sup.3 is hydrogen and b=1;
- R.sup.2 is hydrogen or --(CH.sub.2).sub.c --Y;
- c is 1-3;
- Y is hydrogen, --OR.sub..alpha., --S--R.sub..alpha., or ##STR47## R.sub..alpha. is hydrogen, alkyl, cycloalkyl, acetyl, propionyl or benzoyl;
- R.sub..beta. is hydrogen or alkyl;
- R.sub..alpha. and R.sub..beta. together with the nitrogen to which they are attached may form a 3-7 member ring, which may also contain 0-2 additional hetero atoms, and which is selected from the group of ethyleneimine, trimethyleneamine, piperidine, piperazine, morpholine, thiomorpholine, N-methyl-piperazine, N-phenylpiperazine, N-benzylpiperazine and N-methyl-imidizolidine;
- or a pharmaceutically acceptable salt thereof.
- 3. A compound of the formula ##STR48## wherein: z is 1;
- X is ##STR49## R, R.sup.1, R.sup.3, R.sup.4, R.sup.5 and R.sup.6 are each independently hydrogen, alkyl or aralkyl;
- R.sub.n is hydrogen, alkyl, aralkyl, acetyl, propionyl, benzoyl, carbalkoxy, carbamyl, carbalkoxyalkyl, hydroxyalkyl, alkoxyalkyl or amidino;
- R groups on vicinal carbon atoms may together form a carbon-carbon double bond when R.sub.n is hydrogen;
- R.sup.4 and R.sup.5 geminal groups may together form a spiro substituent, --(CH.sub.2).sub.d --, where d is 2 to 5;
- R.sup.5 and R.sup.6 groups may together form a carbon-nitrogen double bond when R.sup.3 is hydrogen;
- R.sup.2 is hydrogen or --(CH.sub.2).sub.c --Y;
- c is 1-3;
- Y is hydrogen, --OR.sub..alpha., --S--R.sub..alpha., or ##STR50## R.sub..alpha. is hydrogen, alkyl, cycloalkyl, acetyl, propionyl or benzoyl;
- R.sub..beta. is hydrogen or alkyl;
- R.sub..alpha. and R.sub..beta. together with the nitrogen to which they are attached may form a 3-7 member ring, which may also contain 0-2 additional hetero atoms, and which is selected from the group of ethyleneimine, trimethyleneamine, piperidine, piperazine, morpholine, thiomorpholine, N-methyl-piperazine, N-phenylpiperazine, N-benzylpiperazine and N-methyl-imidizolidine;
- or a pharmaceutically acceptable salt thereof.
- 4. A compound according to claim 1 wherein: a is 1.
- 5. A compound according to claim 4 which is 6-[6-{3,4-dihydro-2(1H)-2-oxo-quinazolinyl}]-5-(4-methyl-1-piperazinomethyl)-4,5-dihydro-3(2H)-pyridazinone.
- 6. A compound according to claim 4 which is 6-[6-{3,4-dihydro-3-methyl-2(1H)-2-oxo-quinazolinyl}]-5-(1-piperidinomethyl)-4,5-dihydro-3(2H)-pyridazinone.
- 7. A compound according to claim 4 which is 6-[6-{3,4-dihydro-3-methyl-2(1H)-2-oxo-quinazolinyl}]-5-(N,N-dimethylaminomethyl)-4,5-dihydro-3(2H)-pyridazinone.
- 8. A compound according to claim 4 which is 6-[6-{3,4-dihydro-3-methyl-2(1H)-2-oxo-quinazolinyl}]-5-(N,N-diethylaminomethyl)-4,5-dihydro-3(2H)-pyridazinone.
- 9. A compound according to claim 4 which is 6-[6-{3,4-dihydro-2(1H)-2-oxo-quinazolinyl}]-5-(N,N-dimethylaminomethyl)-4,5-dihydro-3(2H)-pyridazinone.
- 10. A compound according to claim 4 which is 6-[6-{3,4-dihydro-3-methyl-2(1H)-2-oxo-quinazolinyl}]-5-hydroxymethyl-4,5-dihydro-3(2H)-pyridazinone.
- 11. A compound according to claim 4 which is 6-[6-(3,4-dihydro-3-methyl-2(1H)-2-oxo-quinazolinyl)]-5-(4-morpholinomethyl)-4,5-dihydro-3(2H)-pyridazinone.
- 12. A compound according to claim 1 wherein: a is 2.
- 13. A compound according to claim 4 wherein: R.sup.5 and R.sup.6 together form a carbon-nitrogen bond.
- 14. A compound according to claim 12 wherein: vicinal R.sup.5 groups together form a carbon-carbon double bond.
- 15. A compound according to claim 1 wherein: R.sup.2 is --CH.sub.2 --O--R.sub..alpha. or --CH.sub.2 --S--R.sub..alpha..
- 16. A compound according to claim 1 wherein: R.sup.2 is hydrogen or --CH.sub.3.
- 17. A compound according to claim 1 wherein: R.sup.2 is ##STR51##
- 18. A compound according to claim 17 wherein: R.sup.2 is ##STR52##
- 19. A compound according to claim 17 wherein: R.sub..alpha. and R.sub..beta. are alkyl.
- 20. A compound according to claim 17 wherein: R.sub..alpha. and R.sub..beta. together with the nitrogen to which they are attached form a 3-7 member ring, which may also contain 0-2 additional hetero atoms, and which is selected from the group of ethyleneimine, trimethyleneamine, piperidine, piperazine, morpholine, thiomorpholine, N-methylpiperazine, N-phenylpiperazine, N-benzylpiperazine and N-methylimidizolidine.
- 21. A compound according to claim 2 wherein: b is 1.
- 22. A compound according to claim 2 wherein: b is 2.
- 23. A compound according to claim 21 wherein: R.sup.5 and R.sup.6 groups together form a carbon-nitrogen bond.
- 24. A compound according to claim 22 wherein: vicinal R.sup.5 groups together form a carbon-carbon double bond.
- 25. A compound according to claim 2 wherein: R.sup.2 is --CH.sub.2 --O--R.sub..alpha. or --CH.sub.2 --S--R.sub..alpha..
- 26. A compound according to claim 2 wherein: R.sup.2 is hydrogen or --CH.sub.3.
- 27. A compound according to claim 2 wherein: R.sup.2 is ##STR53##
- 28. A compound according to claim 27 wherein: R.sup.2 is ##STR54##
- 29. A compound according to claim 27 wherein: R.sub..alpha. and R.sub..beta. are alkyl.
- 30. A compound according to claim 28 wherein: R.sub..alpha. and R.sub..beta. together with the nitrogen to which they are attached form a 3-7 member ring, which may also contain 0-2 additional hetero atoms, and which is selected from the group of ethyleneimine, trimethyleneamine, piperidine, piperazine, morpholine, thiomorpholine, N-methyl-piperazine, N-phenylpiperazine, N-benzylpiperazine and N-methylimidizolidine.
- 31. A compound according to claim 3 wherein: R.sup.6 and one of the R.sup.5 groups together form a carbon-nitrogen bond.
- 32. A compound according to claim 3 wherein: R.sup.2 is --CH.sub.2 --O--R.sub..alpha. or --CH.sub.2 --S--R.sub..alpha..
- 33. A compound according to claim 3 wherein: R.sup.2 is hydrogen or --CH.sub.3.
- 34. A compound according to claim 3 wherein: R.sup.2 is ##STR55##
- 35. A compound according to claim 34 wherein: R.sup.2 is ##STR56##
- 36. A compound according to claim 35 wherein: R.sub..alpha. and R.sub..beta. are alkyl.
- 37. A compound according to claim 34 wherein: R.sub..alpha. and R.sub..beta. together with the nitrogen to which they are attached may form a 3-7 member ring, which may also contain 0-2 additional hetero atoms, and which is selected from the group of ethyleneimine, trimethyleneamine, piperidine, piperazine, morpholine, thiomorpholine, N-methyl-piperazine, N-phenyl-piperazine, N-benzylpiperazine and N-methylimidizolidine.
- 38. A compound according to claim 1 which is selected from the group consisting of
- 6-[6-(3,4-dihydro-1,3-dimethyl-2(1H)-2-oxo-quinazolinyl)]-4,5-dihydro-3(2H)-pyridazinone;
- 6-[6-(3,4-dihydro-3-methyl-2(1H)-2-oxo-quinazolinyl)]-5-(N,N-dimethylaminomethyl)-4,5-dihydro-3(2H)-pyridazinone;
- 6-[6-(3,4-dihydro-3-methyl-2(1H)-2-oxo-quinazolinyl)]-5-(4-morpholinylmethyl)-4,5-dihydro-3(2H)-pyridazinone;
- 6-[6-(3,4-dihydro-3-methyl-2(1H)-2-oxo-quinazolinyl)]-2-benzyl-4,5-dihydro-3(2H)-pyridazinone;
