Claims
- 1. A compound of formula I: ##STR25## wherein R.sup.1 is --H, --OH, --O(C.sub.1 -C.sub.4 alkyl), --OCO(C.sub.1 -C.sub.6 alkyl), --O(CO)O(C.sub.1 -C.sub.6 alkyl), --OCOAr, --O(CO)OAr, where Ar is phenyl or optionally substituted phenyl, or --OSO.sub.2 (C.sub.2 -C.sub.6 alkyl);
- R.sup.2 is C.sub.1 -C.sub.5 n-alkyl, C.sub.3 -C.sub.6 branched alkyl, C.sub.3 -C.sub.7 cycloalkyl, C.sub.3 -C.sub.6 alkenyl, C.sub.4 -C.sub.7 cycloalkenyl, or C.sub.3 -C.sub.5 alkynyl;
- n is 2 or 3; and
- R.sup.3 is 1-piperidinyl, 1-pyrrolidinyl, methyl-1-pyrrolidinyl, dimethyl-1-pyrrolidinyl, 4-morpholino, dimethylamino, diethylamino, or 1-hexamethyleneimino; or a pharmaceutically acceptable salt or solvate thereof.
- 2. A compound according to claim 1 wherein R.sup.1 is --H.
- 3. A compound according to claim 1 wherein R.sup.2 is C.sub.1 -C.sub.5 n-alkyl, C.sub.3 -C.sub.6 branched alkyl, or C.sub.3 -C.sub.7 cycloalkyl.
- 4. A compound according to claim 1 wherein R.sup.1 is --OH.
- 5. A compound according to claim 4 wherein R.sup.2 is C.sub.1 -C.sub.5 n-alkyl, C.sub.3 -C.sub.6 branched alkyl, or C.sub.3 -C.sub.7 cycloalkyl.
- 6. A compound according to claim 1 wherein R.sup.2 is cyclohexyl.
- 7. A compound according to claim 1 wherein n is two.
- 8. A compound according to claim 7 wherein R.sup.3 is pyrrolidinyl, piperidinyl, or hexamethyleneimino.
- 9. A compound according to claim 1 wherein R.sup.1 is hydrogen, R.sup.2 is cyclohexyl, R.sup.3 is piperidinyl, n is two, and said salt thereof is the hydrochloride salt.
- 10. A compound according to claim 1 wherein R.sup.1 is hydrogen, R.sup.2 is 2-propyl, R.sup.3 is piperidinyl, n is two, and said salt thereof is the hydrochloride salt.
- 11. A compound according to claim 1 wherein R.sup.1 is hydrogen, R.sup.2 is propyl, R.sup.3 is piperidinyl, n is two, and said salt thereof is the hydrochloride salt.
- 12. A pharamceutical composition comprising a compound according to claim 1 with a pharmaceutically acceptable carrier, diluent, or excipient.
- 13. A compound according to claim 1 selected from the group consisting of
- 3-[4-[2-(1-pyrrolidinyl)ethoxy]phenoxy]-2-cyclohexylbenzo[b]thiophene hydrochloride;
- 3-[4-[2-(1-hexamethyleneimino)ethoxy]phenoxy]-2-cyclohexylbenzo[b]thiophene hydrochloride;
- 3-[4-[3-(1-piperidinyl)propoxy]phenoxy]-2-cyclohexylbenzo[b]thiophene hydrochloride;
- 3-[4-[2-(1-piperidinyl)ethoxy]phenoxy]-2-cyclohexyl-6-methoxybenzo[b] thiophene hydrochloride;
- 3-[4-[2-(1-piperidinyl)ethoxy]phenoxy]-2-cyclohexyl-6-hydroxybenzo[b]thiophene;

- 14. A method of inhibiting bone loss or bone resorption which comprises administering to a patient in need thereof an effective amount of a compound of claim 1.
- 15. A method according to claim 14, wherein said bone loss or bone resorption is due to menopause or ovariectomy.
- 16. A method of lowering serum cholesterol levels which comprises administering to a patient in need thereof an effective amount of a compound of claim 1.
- 17. A method of inhibiting estrogen-dependent cancer which comprises administering to a patient in need thereof an effective amount of a compound of claim 1.
- 18. A method according to claim 17 wherein said estrogen-dependent cancer is breast cancer.
- 19. A method according to claim 17 wherein said estrogen-dependent cancer is uterine cancer.
- 20. A compound of formula II: ##STR26## wherein: R.sup.1a is --H or --OR.sup.8, where R.sup.8 is a hydroxy-protecting group;
- R.sup.4 is --OH or --H;
- R.sup.5 and R.sup.6 are, independently, --H, C.sub.1 -C.sub.4 n-alkyl, C.sub.3 -C.sub.5 branched alkyl, C.sub.2 -C.sub.5 alkenyl, C.sub.2 -C.sub.4 alkynyl, or R.sup.5 and R.sup.6 may be taken together with methylene groups or vinyl groups to form 3 to 7-membered cycloalkyl or cycloalkenyl rings; and
- R.sup.7 is --OH or --OR.sup.9, where R.sup.9 is a hydroxy-protecting group which can be selectively removed in the presence of R.sup.8 ; with the proviso that only two of R.sup.4, R.sup.5, and R.sup.6 can be hydrogen.
- 21. A compound according to claim 20 selected from the group consisting of
