Claims
- 1. A method for transforming an initial FRET image of a specimen to a processed FRET image, the initial image being represented by a set of pixels each having three color space coordinates, the method including the steps of:(a) generating a corrected FRET image from the initial image by correcting for spectral overlap by applying a transformation of the following form to obtain a corrected FRET channel image Fc: Fc=Fb−(Fdb/Ddb)×Db−(Fab/Aab)×Ab wherein Fb are pixels of an initial FRET channel image, Db are pixels of an initial donor channel image, Ab are pixels of an initial acceptor channel image, Fb are donor pixels of the initial FRET channel image, Fab are acceptor pixels of the initial FRET channel image, Ddb are donor pixels of the initial donor channel image, and Aab are acceptor pixels of the initial acceptor channel image, all after background subtraction;(b) selecting at least two image regions in the corrected FRET image, each image region including at least one pixel, wherein the image regions define at least two distinguishable spectral categories and do not substantially overlap spatially; (c) performing a color space transformation on the corrected image, based on the color space coordinates of the pixels in each of the selected image regions, to generate a processed image; and (d) evaluating the processed image for a visible indication of an improved spatial distribution of color.
- 2. The method of claim 1, including the step of further correcting the corrected FRET image for cross-talk resulting from detection of donor fluorescence through an acceptor emission channel and detection of acceptor fluorescence through a donor emission channel.
- 3. The method of claim 1, including the step of further correcting the corrected FRET image for non-zero values of a signal Ad from a donor-only specimen using an acceptor filter set, and a signal Da from an acceptor-only specimen using a donor filter set.
- 4. The method of claim 1, including the step of further correcting the corrected FRET channel image Fc by normalizing for the concentration of a donor present in the specimen.
- 5. The method of claim 1, including the step of further correcting the corrected FRET channel image Fc, so as to be approximately normalized for the concentration of a donor, by applying a correction of 1/Df, where Df is a signal generated through a donor filter set when both donors and acceptors are present in the specimen.
- 6. The method of claim 1, including the step of further correcting the corrected FRET channel image Fc by normalizing for the concentration of an acceptor present in the specimen.
- 7. The method of claim 1, including the step of further correcting the corrected FRET channel image Fc, so as to be approximately normalized for the concentration of an acceptor, by applying a correction of 1/Af, where Af is a signal generated through an acceptor filter set when both donors and acceptors are present in the specimen.
- 8. The method of claim 1, including the step of further correcting the corrected FRET channel image Fc by normalizing for the concentration of both a donor and an acceptor present in the specimen.
- 9. The method of claim 1, including the step of further correcting the corrected FRET channel image Fc, so as to be approximately normalized for the concentration of both a donor and an acceptor in the specimen, by applying a correction of 1/(Df×Af), where Df is a signal generated through a donor filter set when both donors and acceptors are present, and Af is a signal generated through an acceptor filter set when both donors and acceptors are present.
- 10. The method of claim 1, including the step of further correcting the corrected FRET image by correcting for cross-talk resulting from detection of donor fluorescence through an acceptor emission channel and detection of acceptor fluorescence through a donor emission channel, and by normalizing for the concentration of both a donor and an acceptor in the specimen.
- 11. The method of claim 1, wherein the extent of photobleachin of the initial FRET image is less than about 5%.
- 12. A computer program, residing on a computer-readable medium, for transforming an initial FRET image to a processed FRET image, the initial image being represented by a set of pixels each having three color space coordinates, the computer program comprising instructions for causing a computer to:(a) generate a corrected FRET image from an initial image by correcting for spectral overlap by applying a transformation of the form to obtain a corrected FRET channel image Fc: Fc=Fb−(Fdb/Ddb)×Db−(Fab/Aab)×Ab wherein Fb are pixels of an initial FRET channel image Db are pixels of an initial donor channel image, Ab are pixels of an initial acceptor channel image, Fdb are donor pixels of the initial FRET channel image, Fab are acceptor pixels of the initial FRET channel image, Ddb are donor pixels of the initial donor channel image and Aab are acceptor pixels of the initial acceptor channel image, all after background subtraction;(b) select at least two image regions in the corrected FRET image, each image region including at least one pixel, wherein the image regions define at least two distinguishable spectral categories and do not substantially overlap spatially; (c) perform a color space transformation on the corrected image, based on the color space coordinates of the pixels in each of the selected image regions, to generate a processed image; and (d) evaluate the processed image for a visible indication of an improved spatial distribution of color.
