Claims
- 1. A chromatographic support for use in affinity separation and heterogeneous immunoassay consisting of:
- a. a primary solid support consisting essentially of particulate resins or porous plastics, the primary solid support having a flocculating agent immobilized thereon; and
- b. a secondary support consisting essentially of microparticles having ligand or binder for the ligand affixed thereon wherein said secondary support is flocculated onto the primary support.
- 2. The chromatographic support of claim 1 wherein the flocculating agent is polyethyleneimine.
- 3. The chromatographic support of claim 1 wherein the primary support is a resin packed in a column.
- 4. The chromatographic support of claim 1 wherein the microparticles are in the range of 0.1 to 10 micrometers.
- 5. The chromatographic support of claim 4 wherein the microparticles are in the range of 1 to 5 micrometers.
- 6. The chromatographic support of claim 5 wherein the microparticles are chromium dioxide magnetic particles.
- 7. The chromatographic support of claim 5 wherein the microparticles are latex particles.
- 8. A method of performing sandwich immunoassay for detection of a target antigen in a sample comprising the steps of:
- a. forming a primary chromatographic support consisting essentially of particulate resins or porous plastics, the primary chromatographic support having a flocculating agent immobilized thereon;
- b. forming a secondary support of microparticles having first antibody to the target antigen affixed thereon;
- c. forming a reaction mixture which contains a ternary complex consisting of first antibody-antigen-second antibody by contacting the sample with said microparticles and excess labeled second antibody to the target antigen;
- d. contacting the reaction mixture of step c with said primary support to trap microparticles having said ternary complex; and
- e. measuring either bound or unbound label to determine the presence of the target antigen.
- 9. The method of claim 8 wherein the flocculating agent is polyethyleneimine.
- 10. The method of claim 9 wherein the primary support is a resin packed in a column.
- 11. The method of claim 10 wherein the microparticles are chromium dioxide magnetic particles.
- 12. The method of claim 11 wherein the target antigen is thyroid stimulating hormone.
- 13. A method of performing a competitive immunoassay comprising the steps of:
- a. forming a primary chromatographic support consisting essentially of particulate resins or porous plastics, the primary chromatographic support having a flocculating agent immobilized thereon;
- b. forming a secondary support consisting essentially of microparticles having target antigen or its analog affixed thereon;
- c. forming a reaction mixture of a sample suspected of containing a target antigen, said secondary support and a labeled antibody to the target antigen;
- d. contacting the reaction mixture of step c with said primary support to trap the microparticles having said labeled antibody captured thereon; and
- e. measuring either bound or unbound label to determine the presence of the target antigen.
- 14. The method of claim 13 wherein the flocculating agent is polyethyleneimine.
- 15. The method of claim 14 wherein the primary support is a resin packed in a column.
- 16. The method of claim 15 wherein the microparticles are chromium dioxide magnetic particles.
- 17. The method of claim 16 wherein the target antigen is digoxin and the analog attached to the magnetic particles is ouabain.
- 18. A sandwich immunoassay for the detection of a target antigen in a sample suspected of containing said antigen, comprising the steps of:
- a. forming a primary chromatographic support consisting essentially of particulate resins or porous plastics, the primary chromatographic support having a flocculating agent immobilized thereon;
- b. flocculating onto said primary support a secondary support of microparticles having first antibody to the target antigen affixed thereon;
- c. contacting the sample with the support formed in step b and excess labeled second antibody to the target antigen to form a first antibody-antigen-second antibody sandwich attached to such microparticles;
- d. separating bound label from unbound label; and
- e. measuring either bound or unbound label to determine the presence of the target antigen.
- 19. A method of performing a competitive immunoassay comprising the steps of:
- a. forming a primary chromatographic support consisting essentially of particulate resins or porous plastics, the primary chromatographic support having a flocculating agent immobilized thereon;
- b. flocculating onto said primary support a secondary support of microparticles having target antigen or its analog affixed thereon;
- c. contacting a sample suspected of containing a target antigen with the support formed in step b and a labeled antibody to the target antigen;
- d. separating bound label from unbound label; and
- e. measuring either bound or unbound label to determine the presence of the target antigen.
