Claims
- 1. A method for conducting column chromatography, comprising the steps of:
- (a) mounting a chromatographic column in a rack, the column having a top end and a seal member closing the top end;
- (b) lowering a pressure cylinder with a protruding head toward the column until the head ruptures the seal member, the pressure cylinder additionally having a bore which communicates with the head;
- (c) raising the pressure cylinder from the column;
- (d) introducing fluid into the column through the ruptured seal member;
- (e) lowering the pressure cylinder toward the column again;
- (f) sealing the pressure cylinder to the column using a resilient pad carried by the pressure cylinder; and
- (g) exposing the interior of the column to pressure through the bore in the pressure cylinder.
- 2. The method of claim 1, further comprising the steps of mounting a plurality of additional chromatographic columns with seal members in the rack during step (a), lowering a plurality of additional pressure cylinders with protruding heads to rupture the seal members of the additional columns during step (b), raising the additional pressure cylinders during step (c), introducing fluid into the additional columns through their ruptured seal members during step (d), lowering the additional pressure cylinders toward the additional columns again during step (e), sealing the additional pressure cylinders to the additional columns using resilient pads carried by the additional pressure cylinders during step (f), and exposing the interiors of the additional columns to pressure through bores in the additional pressure cylinders during step (g).
- 3. The method of claim 1, further comprising the steps of collecting eluate from the column in a cuvette while step (g) is conducted, moving the cuvette to a measurement station after the eluate is collected, and sensing at least one property of the eluate at the measurement station.
- 4. The method of claim 1, wherein the protruding head has a conical portion that extends to the bore of the pressure cylinder, a flat portion that extends to the bore, and a cutting edge adjacent the bore, and wherein step (b) comprises puncturing the seal member with the cutting edge of the protruding head.
- 5. The method of claim 1 wherein the chromatographic column has a tip portion, the tip portion having a removable cap thereon, the method further comprising;
- moving a decapper member having a forked end into engagement with the tip portion; and
- pressing the forked end against the cap to remove the cap from the tip portion of the column.
- 6. The method of claim 5 and further comprising the steps of:
- receiving the cap in a first cell of a cuvette; and
- moving the relative positions of the column and the cuvette for collecting fluid from the column in a second cell of the cuvette.
- 7. The method of claim 6, further comprising the steps of moving the cuvette to a measurement station after collecting fluid from the column.
- 8. The method of claim 5 wherein the decapper member is flexed to move the forked end into engagement with the tip portion.
- 9. The method of claim 5 wherein the decapper member is biased by a spring to move the forked end into engagement with the tip portion.
- 10. A decapper for use in the method of claim 1.
- 11. A pressure cylinder for use in the method of claim 1.
- 12. A chromatographic column for use in the method of claim 1.
- 13. A chromatographic column for use in the method of claim 1 wherein the seal member includes a foil member.
- 14. A chromatographic column for use in the method of claim 5.
