Claims
- 1. A pharmaceutical composition comprising a glucagon like peptide-1 (GLP-1) derivative and a dual Peroxisome Proliferator-Activated Receptor-α/Peroxisome Proliferator-Activated Receptor-γ (PPAR-α/PPAR-γ) activator.
- 2. The pharmaceutical composition according to claim 1 wherein the dual PPAR-α/PPAR-γ activator is a compound of formula (I)
- 3. The pharmaceutical composition according to claim 2 wherein X is aryl, heteroaryl or heterocyclyl optionally substituted with one or more substituents selected from halogen, perhalomethyl, C1-6-alkoxy, C1-6alkylthio, aryl, aryloxy, arylthio, aralkyl, aralkoxy, heteroaryl, heteroaralkyl, heteroaryloxy or heteroaralkoxy.
- 4. The pharmaceutical composition according to claim 2 wherein X is aryl, heteroaryl or heterocyclyl each of which is optionally substituted with one or more substituents selected from halogen, perhalomethyl, C1-6-alkoxy, C1-6-alkylthio, aryl, aryloxy, arylthio, aralkyl, aralkoxy, heteroaryl, heteroaralkyl, heteroaryloxy or heteroaralkoxy.
- 5. The pharmaceutical composition according to claim 2 wherein X is aryl optionally substituted with one or more substituents selected from halogen, perhalomethyl, C1-6-alkoxy, C1-6alkylthio, aryl, aryloxy, arylthio, aralkyl, aralkoxy, heteroaryl, heteroaralkyl, heteroaryloxy or heteroaralkoxy.
- 6. The pharmaceutical composition according to claim 2 wherein X is phenyl or naphthyl each of which is optionally substituted with one or more substituents selected from halogen or perhalomethyl.
- 7. The pharmaceutical composition according to claim 2 wherein X is phenyl optionally substituted with one or more substituents selected from halogen.
- 8. The pharmaceutical composition according to claim 2 wherein X is phenyl.
- 9. The pharmaceutical composition according to claim 2 wherein X is heteroaryl optionally substituted with one or more substituents selected from halogen, perhalomethyl, C1-6-alkoxy, C1-6-alkylthio, aryl, aryloxy, arylthio, aralkyl, aralkoxy, heteroaryl, heteroaralkyl, heteroaryloxy or heteroaralkoxy.
- 10. The pharmaceutical composition according to claim 2 wherein X is heterocyclyl optionally substituted with one or more substituents selected from halogen, perhalomethyl, C1-6-alkoxy, C1-6-alkylthio, aryl, aryloxy, arylthio, aralkyl, aralkoxy, heteroaryl, heteroaralkyl, heteroaryloxy or heteroaralkoxy.
- 11. The pharmaceutical composition according to claim 2 wherein Y is hydrogen, C1-12-alkyl or aryl.
- 12. The pharmaceutical composition according to claim 2 wherein Y is hydrogen or methyl.
- 13. The pharmaceutical composition according to claim 2 wherein Y is hydrogen.
- 14. The pharmaceutical composition according to claim 2 wherein Z is hydrogen or C1-6-alkoxy.
- 15. The pharmaceutical composition according to claim 2 wherein Z is hydrogen.
- 16. The pharmaceutical composition according to claim 2 wherein Q is O.
- 17. The pharmaceutical composition according to claim 2 wherein Ar is arylene optionally substituted with one or more substituents selected from C1-6-alkyl or C1-6-alkoxy each of which can be optionally substituted with carboxy.
- 18. The pharmaceutical composition according to claim 2 wherein Ar is phenylene.
- 19. The pharmaceutical composition according to claim 2 wherein R1 is hydrogen or R1 forms a bond together with R2.
- 20. The pharmaceutical composition according to claim 2 wherein R1 is hydrogen.
- 21. The pharmaceutical composition according to claim 2 wherein R2 is hydrogen or R2 forms a bond together with R1.
- 22. The pharmaceutical composition according to claim 2 wherein R2 is hydrogen.
- 23. The pharmaceutical composition according to claim 2 wherein R3 is C1-6-alkyl.
- 24. The pharmaceutical composition according to claim 2 wherein R3 is C1-2-alkyl.
- 25. The pharmaceutical composition according to claim 2 wherein R4 is hydrogen.
- 26. The pharmaceutical composition according to claim 2 wherein n is 1.
- 27. The pharmaceutical composition according to claim 2 wherein m is 1.
- 28. The pharmaceutical composition according to claim 2 wherein alkyl is methyl or ethyl.
- 29. The pharmaceutical composition according to claim 2 wherein alkenyl is vinyl or 1-propenyl.
- 30. The pharmaceutical composition according to claim 2 wherein alkynyl is 1-propynyl.

- 49. The pharmaceutical composition according to claim 2 wherein the dual PPAR-α/PPAR-γ activator of formula (I) is selected from the following:

- 51. The pharmaceutical composition according to claim 1 wherein the GLP-1 derivative comprises a sequence wherein at least one amino acid residue of the sequence has a lipophilic substituent attached, with the proviso that if only one lipophilic substituent is present and this substituent is attached to the N-terminal or to the C-terminal amino acid residue of the parent peptide then this substituent is an alkyl group or a group which has an ω-carboxylic acid group.
- 52. The pharmaceutical composition according to claim 2 wherein the GLP-1 derivative comprises a sequence wherein at least one amino acid residue of the sequence has a lipophilic substituent attached, with the proviso that if only one lipophilic substituent is present and this substituent is attached to the N-terminal or to the C-terminal amino acid residue of the parent peptide then this substituent is an alkyl group or a group which has an ω-carboxylic acid group.
- 53. The pharmaceutical composition according to claim 52 wherein the GLP-1 derivative comprises a peptide selected from the group GLP-1(1-35); GLP-1(1-36); GLP-1(1-36)amide; GLP-1(1-37); GLP-1(1-38); GLP-1(1-39); GLP-1(1-40); GLP-1(1-41).
- 54. The pharmaceutical composition according to claim 52 wherein the GLP-1 derivative comprises a peptide selected from the group GLP-1(7-35); GLP-1(7-36); GLP-1(7-36)amide; GLP-1(7-37); GLP-1(7-38); GLP-1(7-39); GLP-1(740) and GLP-1(7-41).
- 55. - 56. A GLP-1 derivative according to claim 52 wherein the lipophilic substituent is the acyl group of a straight-chain or branched fatty acid.
- 57. A GLP-1 derivative according to claim 56 wherein the acyl group is selected from the group comprising CH3(CH2)nCO—, wherein n is 4 to 38.
- 58. A GLP-1 derivative according to claim 56 wherein the acyl group is selected from CH3(CH2)6CO—, CH3(CH2)8CO—, CH3(CH2)10CO—, CH3(CH2)12CO—, CH3(CH2)14CO—, CH3(CH2)16CO—, CH3(CH2)18CO—, CH3(CH2)20CO— and CH3(CH2)22CO—.
- 59. The pharmaceutical composition according to claim 55 wherein the GLP-1 derivative is Arg34-GLP-1(7-37),
- 60. The pharmaceutical composition of claim 59, wherein the lipophilic substituent is linked to the ε-amino group of Lys via a spacer.
- 61. The pharmaceutical composition of claim 60, wherein the spacer is selected from the group: γ-glutamyl, β-asparagyl, glycyl, and β-alanyl.
- 62. The pharmaceutical composition according to claim 59 wherein the GLP-1 derivative is selected from the following:
Lys 26 (Nε-tetradecanoyl)-GLP-1(7-37), Arg34Lys26(Nε-lithocholyl)-GLP-1(7-37)-OH, Arg34Lys26(Nε-(γ-glutamyl(Nα-hexadecanoyl)))-GLP-1(7-37)-OH, and Arg34Lys26(Nε-(γ-glutamyl(Nα-tetradecanoyl)))-GLP-1(7-37)-OH.
- 63. A pharmaceutical composition comprising the composition of claim 2 together with a pharmaceutically acceptable carrier or diluent.
- 64. A pharmaceutical composition comprising the pharmaceutical composition of claim 52 together with a pharmaceutically acceptable carrier or diluent.
- 65. A pharmaceutical composition according to claim 64 in unit dosage form, comprising from about 0.05 to about 100 mg, preferably from about 0.1 to about 50 mg of the compound according to any one of the preceding compound claims or a pharmaceutically acceptable salt thereof.
- 66. A process for making a pharmaceutical composition comprising combining a GLP-1 derivative and a dual PPAR-α/PPAR-γ activator and a pharmaceutically acceptable carrier.
- 67. A method of treating or preventing disorders or diseases mediated by nuclear receptors, comprising administering to a patient in need of such treatment a therapeutically effective amount of the pharmaceutical composition of claim 2.
- 68. A method of treating or preventing metabolic disorders or diseases mediated by the PPARs, comprising administering to a patient in need of such treatment a therapeutically effective amount of the pharmaceutical composition of claim 2.
- 69. A method of treating or preventing metabolic syndrome, diabetes and/or obesity, comprising administering to a patient in need of such treatment a therapeutically effective amount of the pharmaceutical composition of claim 2 or a pharmaceutically acceptable salt thereof together with a pharmaceutically acceptable carrier or diluent.
- 70. A pharmaceutical composition according to claim 64 for oral, nasal, transdermal, pulmonal, or parenteral administration.
- 71. A kit comprising an amount of a GLP-1 derivative and a pharmaceutically acceptable carrier, vehicle, or diluent in a first unit dosage form; an amount of a dual PPAR-α/PPAR-γ activator and a pharmaceutically acceptable carrier, vehicle, or diluent in a second unit dosage form; and a container.
- 72. A method for treating or preventing diseases or disorders involving the metabolic syndrome, insulin resistance, dyslipidemia, obesity, diabetes, hypertriglyceridaemia, impaired glucose tolerance or hypertension, said method comprising administering to a patient in need of said treatment (i) a first amount of a GLP-1 derivative and (ii) a second amount of a dual PPAR-α/PPAR-γ activator, wherein said first and second amounts together are effective for said treatment.
