Claims
- 1. A composition comprising recombinant coagulation factor VIII, a non-ionic surfactant in an amount of at least 0.01 mg/ml, sodium or potassium chloride in an amount of more than 0.1 M, and L-histidine for buffering the composition and protecting the factor VIII in the amorphous state, whereby the factor VIII activity is stabilized during storage, said composition being either (a) initially and finally an aqueous solution ready for use, or (b) initially an aqueous solution which is subsequently dried and finally reconstituted as an aqueous solution before use.
- 2. The composition according to claim 1 in which said factor VIII has a specific activity of more than 2,000 IU/mg protein.
- 3. The composition according to claim 1 in which the specific activity of said factor VIII is more than 5,000 IU/mg protein and is stabilized without the addition of albumin.
- 4. The composition according to claim 1 in which factor VIII is full-length or a deletion derivative of recombinant factor VIII having full factor VIII activity.
- 5. The composition according to claim 1 in which the amount of factor VIII is 10 to 100,000 IU/ml.
- 6. The composition according to claim 5 in which the amount of factor VIII is 50 to 10,000 IU/ml.
- 7. The composition according to claim 1 in which the non-ionic surfactant is present in an amount above the critical micelle concentration.
- 8. The composition according to claim 1 in which the non-ionic surfactant is selected from the group consisting of block co-polymers and polyoxyethylene (20) fatty acid esters.
- 9. The composition according to claim 8 in which the block co-polymer is a polyoxamer.
- 10. The composition according to claim 8 in which the polyoxyethylene (20) fatty acid ester is selected from the group consisting of polysorbate 20 and polysorbate 80.
- 11. The composition according to claim 1 which comprises mono- or disaccharides or sugar alcohols.
- 12. The composition according to claim 11 in which the disaccharide is sucrose.
- 13. The composition according to claim 1 in which the ratio of sodium chloride to L-histidine is more than 1:1.
- 14. The composition according to claim 1 which further comprises a calcium salt.
- 15. The composition according to claim 14 in which the calcium salt is in an amount of more than 0.5 mM.
- 16. The composition according to claim 14 in which the calcium salt is selected from the group consisting of calcium chloride and calcium gluconate.
- 17. The composition according to claim 1, comprising:
- i) 10-100,000 IU/ml of recombinant factor VIII,
- ii) at least 0.01 mg/ml of polyoxyethylene (20) fatty acid ester,
- iii) sodium chloride in an amount of more than 0.1 M,
- iv) calcium salt,
- v) L-histidine in an amount of more than 1 mM.
- 18. The composition according to claim 1 wherein said calcium is calcium chloride or calcium gluconate in an amount of more than 0.5 mM.
- 19. The composition according to claim 1 which is dried.
- 20. The composition according to claim 19 which is lyophilized.
- 21. The composition according to claim 1 which is a stable aqueous solution ready for use.
- 22. The composition according to claim 1 in which said factor VIII activity is stabilized during storage for at least 6 months.
- 23. The composition according to claim 22 wherein storage is performed at 5.+-.3.degree. C.
- 24. A process for preparation of the composition according to claim 1 comprising mixing said factor VIII with the non-ionic surfactant in an aqueous solution, with L-histidine and sodium or potassium chloride, and optionally further with sucrose and a calcium salt.
- 25. A process for preparation of the composition according to claim 1 comprising eluting said factor VIII from a last purification step with a buffer containing a non-ionic surfactant in an aqueous solution, with L-histidine and sodium or potassium chloride, and optionally further with sucrose and a calcium salt.
