Claims
- 1. An x-ray contrast composition for oral or retrograde examination of the gastrointestinal tract comprising on a % weight per volume basis:
- (a) from about 5 to 45% of an x-ray contrast producing agent having the formula, or a pharmaceutically acceptable salt thereof ##STR39## or a pharmaceutically acceptable salt thereof wherein Z=H, halo, C.sub.1 -C.sub.20 alkyl, cycloalkyl, lower alkoxy, cyano, where the alkyl and cycloalkyl groups can be substituted with halogen or halo-lower-alkyl groups;
- R=methyl, ethyl, propyl, C.sub.9 -C.sub.25 alkyl, cycloalkyl, or halo-lower-alkyl, optionally substituted with halo, fluoro-lower-alkyl, aryl, lower-alkoxy, hydroxy, carboxy, lower-alkoxy carbonyl or lower-alkoxy-carbonyloxy, (CR.sub.1 R.sub.2).sub.p --(CR.sub.3 .dbd.CR.sub.4).sub.m Q, or (CR.sub.1 R.sub.2).sub.p --C.dbd.C--Q;
- R.sub.1, R.sub.2, R.sub.3 and R.sub.4 are independently lower-alkyl, optionally substituted with halo;
- x is 1-4;
- n is 1-5;
- m is 1-15;
- p is 1-10; and
- Q is H, lower-alkyl, lower-alkenyl, lower-alkynyl, lower-alkylene, aryl, or aryl-lower alkyl;
- (b) from about 0.1 to 10% of a pharmaceutically acceptable clay selected from the group consisting of: montmorillonite, beidelite, nontronite, hectorite and saponite;
- (c) from about 1.0 to 20% of a surfactant selected from the group consisting of nonionic, anionic, cationic and zwitterionic surfactants;
- (d) from about 0 to 15% of an excipient; and
- (e) water to make 100% by volume.
- 2. The x-ray contrast composition of claim 1 wherein said x-ray contrast producing agent is present in an amount of from about 10 to 35%.
- 3. The x-ray contrast composition of claim 1 wherein said pharmaceutically acceptable clay constitutes from 0.5 to 5% of the composition.
- 4. The x-ray contrast composition of claim 1 wherein said surfactant constitutes from 2 to 10% of the composition.
- 5. The x-ray contrast composition of claim 1 wherein said excipient constitutes from 0.5 to 5% of the composition.
- 6. The x-ray contrast composition of claim 1 wherein said nonionic surface active agent is selected from the group consisting of carboxylic esters, carboxylic amides, ethoxylated alklyphenols, ethoxylated aliphatic alcohols, ethylene oxide polymer, ethylene oxide/propylene oxide co-polymer, polyvinylpyrrolidone and polyvinylalcohol.
- 7. The x-ray contrast composition of claim 1 wherein said surfactant is sorbitan ester having the formula: ##STR40## wherein R.sub.1 =R.sub.2 =OH, R.sub.3 =R for sorbitan monoesters,
- R.sub.1 =OH, R.sub.2 =R.sub.3 =R for sorbitan diesters,
- R.sub.1 =R.sub.2 =R.sub.3 =R for sorbitan triesters,
- where
- R=(C.sub.11 H.sub.23) COO for laurate,
- (C.sub.17 H.sub.33) COO for oleate,
- (C.sub.15 H.sub.31) COO for palmitate or
- (C.sub.17 H.sub.35) COO for stearate.
- 8. The x-ray contrast composition of claim 1 wherein said surface active agent is polyoxyethylene stearate.
- 9. The x-ray contrast composition of claim 1 wherein said surfactant is polyoxyethylene sorbitan fatty acid ester of the formulas (1) and (2) ##STR41## wherein the sum of w+x+y+z is selected from the group consisting of 4, 5 and 20.
- 10. The x-ray contrast composition of claim 1 wherein said x-ray contrast producing agent is selected from the group consisting of: 2-(4-iodophenoxy)-decane, 2-(2,4,6-triiodophenoxy)-pentadecane, 2-(2,4,6-triiodophenoxy)decane, (2,4,6-triiodophenoxy)-1H, 1H, 2H, 2H-perfluorooctane and 1-(2,4,6-triiodo-3-trifluorophenoxy)octane.
- 11. The x-ray contrast composition of claim 1 wherein said x-ray contrast producing agent is selected from the group consisting of: 2-(2,4,6-triiodophenoxy)-nonane, 2-ethyl-1-(2,4,6-triiodophenoxy)-hexane and 3,3-diphenyl-1-(2,4,6-triiodophenoxy)propane.
- 12. The x-ray contrast composition of claim I wherein said x-ray contrast producing agent is selected from the group consisting of: 3-(2,4,6-triiodophenoxy)-nonane, 2-(4-iodophenoxy)-undecane, 2-iodophenoxycyclopentane, 3-iodophenoxycyclopentane and (3,5-dimethyl-2,4,6-triiodophenoxy)cyclopentane.
- 13. The x-ray contrast composition of claim 1 wherein said x-ray contrast producing agent is selected from the group consisting of: 2-(4-iodophenoxy)-pentadecane, 4-iodophenoxycyclopentane, 2,4,6-triiodophenoxycyclopentane, 2,4,6-triiodophenoxymethylcyclopentane and 2-(2,4,6-triiodophenoxy)ethylcyclopentane.
- 14. The x-ray contrast composition of claim 1 wherein said x-ray contrast producing agent is selected from the group consisting of: (E,E)-1-(2,4,6-triiodophenoxy)-3,7,11-trimethyl-2,6,10-dodecatriene, 1-(2,4,6-triiodophenoxy)-3,7-dimethyl-6-octene, (E)-1-(3,5-dimethyl-2,4,6-triiodophenoxy)-3,7-dimethyl-2,6-octadiene and (E)-1-(2,4,6-triiodophenoxy)-3,7-dimethyl-2,6-octadiene.
- 15. The x-ray contrast composition of claim 1 wherein said x-ray contrast producing agent is selected from the group consisting of: 1-(2,4,6-triiodophenoxy)-3-octyne, 2-(2,4,6-triiodophenoxy)-4-octyne, 1-(2,4,6-triiodophenoxy)-3-octyne, diethyl 2-(2,4,6-triiodophenoxy)-1,3-propanedioate and diisopropyl 2-(2,4,6-triiodophenoxy)-1,3propanedioate.
- 16. The x-ray contrast composition of claim 1 wherein said x-ray contrast producing agent is selected from the group consisting of: ethyl 2,2-bis-(3-iodophenoxy)acetate, ethyl-5-(2,4,6-triiodophenoxy)-hexanoate, 5-(2,4,6-triiodophenoxy)-hexan-1-ol, 10-(4-iodophenoxy)-undecan-1-ol, ethyl 5-(2,4,6-triiodophenoxy)-hexyl carbonate and ethyl 10-(3-iodophenoxy)-undecanoate.
- 17. A method of carrying out x-ray examination of the gastrointestinal tract of a patient, said method comprises the oral or rectal administration to the patient an x-ray contrast composition comprising:
- (a) from about 5 to 45% of an x-ray contrast producing agent having the formula, or a pharmaceutically acceptable salt thereof ##STR42## wherein Z is H, halo, C.sub.1 -C.sub.20 alkyl, cycloalkyl, lower alkoxy, cyano, where the alkyl and cycloalkyl groups can be substituted with halogen or halo-lower-alkyl groups;
- R is methyl, ethyl, propyl, C.sub.9 -C.sub.25 alkyl, cycloalkyl, or halo-lower-alkyl, each of which may be optionally substituted with halo, fluoro-lower-alkyl, aryl, lower-alkoxy, hydroxy, carboxy, lower-alkoxy carbonyl or lower-alkoxy-carbonyloxy; (CR.sub.1 R.sub.2).sub.p --(CR.sub.3 .dbd.CR.sub.4).sub.m Q, or (CR.sub.1 R.sub.2).sub.p --C.tbd.C--Q;
- R.sub.1, R.sub.2, R.sub.3 and R.sub.4 are independently lower-alkyl, optionally substituted with halo;
- x is 1-3
- y is 1-4;
- n is 1-5;
- m is 1-15;
- p is 1-10; and
- Q is H, lower-alkyl, lower-alkenyl, lower-alkynyl, lower-alkylene, aryl, or aryl-lower alkyl;
- (b) from about 0.1 to 10% of a pharmaceutically acceptable clay selected from the group consisting of: montmorillonite, beidelite, nontronite, hectorite and saponite;
- (c) from about 1.0 to 20% of a surfactant selected from the group consisting of nonionic, anionic, cationic and zwitterionic surfactants;
- (d) from about 0.0 to 15% of an excipient; and
- (e) water to make 100% by volume.
- 18. The method of claim 17 wherein said x-ray contrast producing agent is present in an amount of from about 10 to 35%.
- 19. The method of claim 17 wherein said pharmaceutically acceptable clay constitutes from 0.5 to 5% of the composition.
- 20. The method of claim 17 wherein said surfactant constitutes from 2 to 10% of the composition.
- 21. The method of claim 17 wherein said excipient constitutes from 0.5 to 5% of the composition.
- 22. The method of claim 17 wherein said nonionic surface active agent is selected from the group consisting of carboxylic esters, carboxylic amides, ethoxylated alklyphenols, ethoxylated aliphatic alcohols, ethylene oxide polymer, ethylene oxide/propylene oxide co-polymer, polyvinylpyrrolidone and polyvinylalcohol.
- 23. The method of claim 17 wherein said surfactant is sorbitan ester having the formula: ##STR43## wherein R.sub.1 =R.sub.2 =OH, R.sub.3 =R for sorbitan monoesters,
- R.sub.1 =OH, R.sub.2 =R.sub.3 =R for sorbitan diesters,
- R.sub.1 =R.sub.2 =R.sub.3 =R for sorbitan triesters,
- where R=
- (C.sub.11 H.sub.23) COO for laurate,
- (C.sub.17 H.sub.33) COO for oleate,
- (C.sub.15 H.sub.31) COO for palmitate or
- (C.sub.17 H.sub.35) COO for stearate.
- 24. The method of claim 17 wherein said surface active agent is polyoxyethylene stearate.
- 25. The method of claim 17 wherein said surfactant is polyoxyethylene sorbitan fatty acid ester of the formulas (1) and (2) ##STR44## wherein the sum of w+x+y+z is selected from the group consisting of 4, 5 and 20.
- 26. The method of claim 17 wherein said x-ray producing agent is selected from the group consisting of: 2-(4-Iodophenoxy)-decane, 2-(2,4,6-triiodophenoxy)-pentadecane, 2-(2,4,6-triiodophenoxy)decane, (2,4,6-triiodophenoxy)-1H, 1H, 2H, 2H-perfluorooctane and 1-(2,4,6-triiodo-3-trifluorophenoxy)octane.
- 27. The method of claim 17 wherein said x-ray producing agent is selected from the group consisting of: 2-(2,4,6-triiodophenoxy)nonane, 2-ethyl-1-(2,4,6-triiodophenoxy)-hexane and 3,3-diphenyl-1-(2,4,6-triiodophenoxy)propane.
- 28. The method of claim 17 wherein said x-ray producing agent is selected from the group consisting of: 3-(2,4,6-triiodophenoxy)nonane, 2-(4-iodophenoxy)-undecane, 2-iodophenoxycyclopentane, 3-iodophenoxycyclopentane and (3,5-dimethyl-2,4,6-triiodophenoxy)cyclopentane.
- 29. The method of claim 17 wherein said x-ray producing agent is selected from the group consisting of: 2-(4-iodophenoxy)-pentadecane, 4-iodophenoxycyclopentane, 2,4,6-triiodophenoxycyclopentane, 2,4,6-triiodophenoxymethylcyclopentane and 2-(2,4,6-triiodophenoxy)ethylcyclopentane.
- 30. The method of claim 17 wherein said x-ray producing agent is selected from the group consisting of: (E,E)-1-(2,4,6-triiodophenoxy)-3,7,11-trimethyl-2,6,10-dodecatriene, 1-(2,4,6-triiodophenoxy)-3,7-dimethyl-6-octene, (E)-1-(3,5-dimethyl-2,4,6-triiodophenoxy)-3,7-dimethyl-2,6-octadiene and (E)-1-(2,4,6-triiodophenoxy)-3,7-dimethyl-2,6-octadiene.
- 31. The method of claim 17 wherein said x-ray producing agent is selected from the group consisting of: 1-(2,4,6-triiodophenoxy)-3-octyne, 2-(2,4,6-triiodophenoxy)-4-octyne, 1-(2,4,6-triiodophenoxy)-3-octyne, diethyl 2-(2,4,6-triiodophenoxy)-1,3-propanedioate and diisopropyl 2-(2,4,6-triiodophenoxy)-1,3-propanedioate.
- 32. The method of claim 17 wherein said x-ray producing agent is selected from the group consisting of: ethyl 2,2-bis-(3-iodophenoxy)acetate, ethyl-5-(2,4,6-triiodophenoxy)-hexanoate, 5-(2,4,6-triiodo-phenoxy)-hexan-1-ol, 10-(4-iodophenoxy)-undecan-1-ol, ethyl 5-(2,4,6-triiodophenoxy)-hexyl carbonate and ethyl 10-(3-iodophenoxy)undecanoate.
