Claims
- 1. A D-amidase capable of specifically hydrolyzing D-.alpha.-alanineamide into D-.alpha.-alanine, with no substantial hydrolysis of D-.alpha.-alanineamide, having an optimum pH of 7 to 8 at 30.degree. C.; an optimum temperature of 40.degree. to 45.degree. C. at pH of 7.5; a molecular weight of 50,000.+-.5,000 as determined by SDS-polyacrylamide electrophoresis; and an isoelectric point at pH 5.2.+-.0.3, wherein said D-amidase is stable within a range of pH 6.5 to 10 at 30.degree. C.; is inactivated when allowed to stand at a temperature above 60.degree. C. for 10 minutes and further requires no coenzyme for activation.
- 2. The process for producing at least one of D-.alpha.-alanine which comprises specifically hydrolyzing D-.alpha.-alanineamide by reacting said D-.alpha.-alanineamide with the D-amidase of claim 1 in an aqueous medium of and recovering at least one of D-.alpha.-alanine from the resulting reaction mixture.
- 3. The process according to claim 2, wherein the potency of D-amidase is 1 to 300 kU/l, wherein U is the activity for hydrolyzing D-alanineamide into 1 micromole of D-alanine per minute.
- 4. The process according to claim 2, wherein the aqueous medium contains 5 to 50% (v/v) glycerol.
- 5. The process according to claim 2, wherein the concentration of D-.alpha.-alanineamide is 1 to 300 g/l.
- 6. The process according to claim 2, wherein the concentration of DL-.alpha.-alanineamide is 1 to 500 g/l.
