Claims
- 1. A method for detecting a restriction endonuclease mediated reaction by change in a fluorescence parameter comprising:
- (a) conducting a nucleic acid primer extension reaction to synthesize a double-stranded, fluorescently labeled nucleic acid molecule containing a nucleic acid sequence recognizable by the restriction endonuclease;
- (b) contacting the restriction endonuclease with said nucleic acid molecule to cause a cleavage or nicking of said nucleic acid sequence thereby providing a change in a fluorescence parameter; and
- (c) detecting the change in a fluorescence parameter.
- 2. The method of claim 1 wherein the nucleic acid primer extension reaction is part of a nucleic acid amplification reaction.
- 3. The method of claim 2 wherein the fluorescence label comprises a donor fluorophore and an acceptor dye in a configuration such that fluorescence of the donor fluorophore is quenched.
- 4. The method of claim 3 wherein the change in a fluorescence parameter is an increase in donor fluorophore intensity or a decrease in acceptor dye fluorescence intensity.
- 5. The method of claim 1 wherein the change in fluorescence parameter is detected in real-time.
- 6. The method of claim 1 wherein the change in fluorescence parameter is detected at an endpoint.
- 7. The method of claim 1 wherein the change in fluorescence parameter is detected periodically over time.
- 8. A method for detecting a restriction endonuclease mediated reaction by change in a fluorescence parameter comprising:
- (a) using a nucleic acid primer extension reaction to prepare a reaction mixture comprising a restriction endonuclease and a double-stranded, fluorescently labeled nucleic acid molecule containing a nucleic acid sequence recognizable by the restriction endonuclease; and
- (b) detecting a change in a fluorescence parameter as a result of nicking or cleavage of said nucleic acid molecule by the restriction endonuclease.
- 9. The method of claim 8 wherein the fluorescence label comprises a donor fluorophore and an acceptor dye in a configuration such that fluorescence of the donor fluorophore is quenched.
- 10. The method of claim 9 wherein the change in a fluorescence parameter is an increase in donor fluorophore intensity or a decrease in acceptor dye fluorescence intensity.
- 11. The method of claim 8 wherein the change in fluorescence parameter is detected in real-time.
- 12. The method of claim 8 wherein the change in fluorescence parameter is detected at an endpoint.
- 13. The method of claim 8 wherein the change in fluorescence parameter is detected periodically over time.
- 14. A method for detecting a restriction endonuclease mediated reaction by change in a fluorescence parameter comprising:
- (a) conducting a nucleic acid primer extension reaction to synthesize a double-stranded, nucleic acid molecule wherein one strand comprises a donor fluorophore, an acceptor dye and a nucleic acid sequence recognizable by the restriction endonuclease between said donor fluorophore and said acceptor dye;
- (b) contacting a restriction endonuclease with said nucleic acid molecule to cause a cleavage or nicking of said nucleic acid sequence thereby providing a change in a fluorescence parameter; and
- (c) detecting the change in a fluorescence parameter.
- 15. The method of claim 14 wherein the nucleic acid primer extension reaction is part of a nucleic acid amplification reaction.
- 16. The method of claim 14 wherein the donor fluorophore and the acceptor dye are in a configuration such that fluorescence of the donor fluorophore is quenched.
- 17. The method of claim 16 wherein the change in a fluorescence parameter is an increase in donor fluorophore intensity or a decrease in acceptor dye fluorescence intensity.
- 18. The method of claim 14 wherein the change in fluorescence parameter is detected in real-time.
- 19. The method of claim 14 wherein the change in fluorescence parameter is detected at an endpoint.
- 20. The method of claim 14 wherein the change in fluorescence parameter is detected periodically over time.
- 21. A method for detecting a restriction endonuclease mediated reaction by change in a fluorescence parameter comprising:
- (a) using a nucleic acid primer to prepare a double-stranded nucleic acid molecule wherein one strand of said double-stranded nucleic acid molecule comprises a donor fluorophore, an acceptor dye and a nucleic acid sequence recognizable by the restriction endonuclease between said donor fluorophore and said acceptor dye, wherein said double-stranded nucleic acid molecule is part of a reaction mixture also comprising a restriction endonuclease; and
- (b) detecting a change in a fluorescence parameter as a result of nicking or cleavage of said nucleic acid molecule by the restriction endonuclease.
- 22. The method of claim 21 wherein the donor fluorophore and the acceptor dye are in a configuration such that fluorescence of the donor fluorophore is quenched.
- 23. The method of claim 22 wherein the change in a fluorescence parameter is an increase in donor fluorophore intensity or a decrease in acceptor dye fluorescence intensity.
- 24. The method of claim 21 wherein the change in fluorescence parameter is detected in real time.
- 25. The method of claim 21 wherein the change in fluorescence parameter is detected at an endpoint.
- 26. The method of claim 21 wherein the change in fluorescence parameter is detected periodically over time.
