Claims
- 1. A method for detecting receptivity of endometrium to embryo implantation comprising:
a. obtaining a sample of said endometrium, b. contacting said sample with a monoclonal antibody specific for β3, and c. detecting the presence or absence of β3 in said sample.
- 2. The method of claim 1 wherein said monoclonal antibody is SSA6.
- 3. The method of claim 1 wherein said β3 is combined with another integrin subunit.
- 4. The method of claim 3 wherein said other integrin subunit is αv.
- 5. The method of claim 4 wherein said monoclonal antibody is 23C6.
- 6. The method of claim 1 wherein said β3 is detected by immunohistochemically staining said sample of endometrium contacted with said monoclonal antibody specific for β3.
- 7. The method of claim 1 wherein said β3 is detected by immunoblotting β3 from said sample of endometrium contacted with said monoclonal antibody specific for β3.
- 8. The method of claim 1 wherein said endometrium is human.
- 9. A method for diagnosing infertility in a mammal comprising:
a. obtaining a sample of endometrium from said mammal, b. contacting said sample with a monoclonal antibody specific for β3, c. monitoring the appearance β3 in said sample, and d. repeating steps a.-c. with samples of said endometrium at a plurality of stages of the menstrual cycle.
- 10. The method of claim 9 wherein said monoclonal antibody is SSA6.
- 11. The method of claim 9 wherein said β3 is combined with another integrin subunit.
- 12. The method of claim 11 wherein said other integrin subunit is αv.
- 13. The method of claim 12 wherein said monoclonal antibody is 23C6.
- 14. The method of claim 9 wherein said mammal is human.
- 15. The method of claim 9 wherein said β3 is monitored by immunohistochemically staining said sample of endometrium contacted with said monoclonal antibody specific for β3.
- 16. The method of claim 9 wherein said β3 is monitored by immunoblotting β3 from said sample of endometrium contacted with said monoclonal antibody specific for β3.
- 17. The method of claim 9 wherein said stages of the menstrual cycle are selected from the group consisting of follicular phase and secretory phase.
- 18. A method of detecting the window of embryo implantation in endometrium comprising:
a. obtaining a sample of endometrium, b. contacting said sample of endometrium with a monoclonal antibody specific for β3, c. detecting the presence or absence of β3 in said sample, and d. repeating steps a.-c. with samples of endometrium at a plurality of stages of the menstrual cycle.
- 19. The method of claim 18 wherein said monoclonal antibody is SSA6.
- 20. The method of claim 18 wherein said β3 is combined with another integrin subunit.
- 21. The method of claim 20 wherein said other integrin subunit is αv.
- 22. The method of claim 21 wherein said monoclonal antibody is 23C6.
- 23. The method of claim 18 wherein said level of β3 is detected by a staining method selected from the group consisting of immunohistochemically staining said sample of endometrium contacted with said monoclonal antibody specific for β3 and immunoblotting β3 from said sample of endometrium contacted with said monoclonal antibody specific for β3.
- 24. A method of in vitro fertilization comprising:
a. obtaining a sample of endometrium from a uterus, b. contacting said sample with a monoclonal antibody specific for β3, c. detecting the presence or absence of β3 in said sample, d. selecting for a sample having β3 present, and e. introducing a zygote into the uterus.
- 25. The method of claim 24 further comprising monitoring said zygote for implantation.
- 26. The method of claim 24 wherein said zygote is formed by the combination of an egg and sperm in vitro.
- 27. The method of in vitro fertilization of claim 24 wherein said monoclonal antibody is SSA6.
- 28. The method of in vitro fertilization of claim 24 wherein said β3 is combined with another integrin subunit.
- 29. The method of in vitro fertilization of claim 28 wherein said other integrin subunit is αv.
- 30. The method of in vitro fertilization of claim 29 wherein said monoclonal is 23C6.
- 31. The method of in vitro fertilization of claim 26 wherein said egg and said sperm are combined according to the group consisting of a petri dish, a test tube and the fallopian tubes.
- 32. A method of preventing embryo implantation in a mammal comprising contacting the endometrium of said mammal with a neutralizing Fab fragment specific for β3.
- 33. The method of claim 32 wherein said Fab fragment is combined with a pharmaceutically acceptable carrier.
- 34. The method of claim 32 wherein said Fab fragment is from monoclonal antibody SSA6.
- 35. The method of claim 33 wherein said β3 is combined with another integrin subunit.
- 36. The method of claim 35 wherein said other integrin subunit is αv.
- 37. The method of claim 36 wherein said Fab fragment is from monoclonal antibody 23C6.
- 38. The method of preventing embryo implantation in a mammal comprising a contacting the endometrium of said mammal with a peptide containing the sequence arginine-glycine-aspartic acid.
- 39. A method of monitoring endometrial maturation in a mammal comprising:
a. obtaining a sample of the endometrium, b. contacting said sample with a monoclonal antibody specific for β3, c. monitoring the appearance of β3 in said sample, and d. repeating steps a.-c. with samples of endometrium at a plurality of stages of the endometrial cycle.
- 40. The method of claim 39 wherein said monoclonal antibody is SSA6.
- 41. The method of claim 39 wherein said β3 is combined with another integrin subunit.
- 42. The method of claim 41 wherein said other integrin subunit is αv.
- 43. The method of claim 42 wherein said monoclonal antibody is 23C6.
- 44. The method of claim 39 wherein said β3 is monitored by a staining method selected from the group consisting of immunohistochemically staining said sample of endometrium contacted with said monoclonal antibody specific for β3 and immunoblotting β3 from said sample of endometrium contacted with said monoclonal antibody specific for β3.
- 45. The method of claim 39 wherein said stages of the endometrial cycle are selected from the group consisting of follicular phase and secretory phase.
- 46. A contraceptive comprising a neutralizing Fab fragment specific for β3 in a therapeutically effective amount and a pharmaceutically acceptable carrier.
- 47. The contraceptive of claim 46 wherein said Fab fragment is from monoclonal antibody SSA6.
- 48. The contraceptive of claim 46 wherein said β3 is combined with another integrin subunit.
- 49. The contraceptive of claim 48 wherein said other integrin subunit is αv.
- 50. The contraceptive of claim 49 wherein said Fab fragment is from monoclonal antibody 23C6.
- 51. The contraceptive comprising a peptide containing the sequence arginine-glycine-aspartic acid.
- 52. A diagnostic kit comprising:
a. a monoclonal antibody specific for β3, b. Fab fragments specific for β3, and c. means for immunostaining said β3.
