Claims
- 1. A device for isolating and identifying a target cell from a cellular mixture, said device comprising:
a substantially planar surface having a bioactive coating applied thereon; and at least one bifunctional compound capable of binding to said target cell and to said bioactive coating; and a delivery conduit communicating said bifunctional compound with said substantially planar surface.
- 2. The device of claim 1 further comprising a binder molecule conjugated with a visual indicator probe.
- 3. The device of claim 1 wherein said bioactive coating is selected from the group consisting of avidin, streptavidin, avidin derivatives, streptavidin derivatives, or blends thereof.
- 4. The device of claim 1 wherein said bifunctional molecule comprises a binding molecule conjugated with a biotin molecule or a biotin derivative.
- 5. The device of claim 4 wherein said binding molecule is selected from the group consisting of antibodies, antigens, glycoconjugates, lectins, hormones, cell receptors, vitamins, amino acids, sugars, lipids, fatty acids, liposomes, DNA probes, and RNA probes.
- 6. The device of claim 2 wherein said probe is selected from the group consisting of flurochromes, radiolabels, fluorescent agents, and chromophores.
- 7. The device of claim 1 wherein said delivery conduit includes a capillary annulus in spaced relationship with said planar surface.
- 8. The device of claim 7 further comprising a fluid absorbing media surrounding said capillary annulus.
- 9. The device of claim 8 further comprising a specimen chamber in communication with said capillary annulus.
- 10. The device of claim 1 wherein said delivery conduit includes a housing in spaced relationship with said planar surface, said housing having an inlet located at a first end of said housing.
- 11. The device of claim 10 further comprising a fluid, absorbing media opposite said inlet and adjacent to said bioactive coating.
- 12. A device for isolating and identifying a target cell from a cellular mixture, said device comprising:
a substantially planar surface having a bioactive coating applied thereon; a conduit in spaced relationship with said surface, wherein said conduit includes at least one channel capable of delivering said cellular mixture to said bioactive coating; and a fluid absorbing media provided on said surface and positioned adjacent to said bioactive coating.
- 13. The device of claim 12 wherein said surface comprises a material selected from the group consisting of metal, plastic, glass, or ceramic.
- 14. The device of claim 12 wherein said bioactive coating is applied to a defined area of said surface.
- 15. The device of claim 13 wherein said bioactive coating is selected from the group consisting of avidin, streptavidin, avidin derivatives, streptavidin derivatives, and blends thereof.
- 16. The device of claim 12 wherein said conduit is donut-shaped and said conduit is positioned above said bioactive coating.
- 17. The device of claim 16 further including a specimen loading chamber axially aligned with said donut-shaped conduit.
- 18. The device of claim 12 wherein said conduit has an inverted housing having a recess and said conduit extends across said bioactive coating.
- 19. The device of claim 18 wherein said conduit includes an inlet in communication with said recess of said conduit.
- 20. A method of isolating a target cell from a cellular mixture, said method comprising:
providing a substantially planar surface having a bioactive coating applied thereon; providing a plurality of bifunctional compounds capable of binding to said target cell and to said bioactive coating; combining said cellular mixture with said bifunctional compound; exposing said cellular mixture to said bioactive coating through a delivery conduit; and analyzing said bioactive coating for said target cell.
- 21. The method of claim 20 further comprising combining said cellular mixture with a marker compound.
- 22. The method of claim 21 wherein said marker compound comprises a binder molecule and a probe.
- 23. The method of claim 22 wherein said binder molecule is selected from the group consisting of antibodies, antigens, glycoconjugates, lectins, hormones, cell receptors, vitamins, amino acids, sugars, lipids, fatty acids, liposomes, DNA probes, and RNA probes.
- 24. The method of claim 22 wherein said probe is selected from the group consisting of flurochromes, radiolabels, fluorescent agents, and chromophores.
- 25. The device of claim 20 wherein said bioactive coating comprises bioactive compounds selected from the group consisting of avidin, streptavidin, avidin derivatives, streptavidin derivatives, and blends thereof.
- 26. The device of claim 20 wherein said bifunctional molecule comprises a binding molecule conjugated with a biotin molecule or a biotin derivative.
- 27. A method of isolating target cells from a cellular mixture comprising:
exposing a cellular mixture to a targeting solution to form a target cell mixture; delivering said target cell mixture to a substantially planar reaction surface; controlling the delivery of said target mixture so as to optimize the flow over said reaction surface; retaining said target cells on said reaction surface; and analyzing said target cells remaining on said reaction surface.
- 28. A method as set forth in claim 27, wherein the controlling of the delivery of the target cell mixture is performed by establishing a predetermined delivery distance for said target cell solution to traverse to said reaction surface.
- 29. A method as set forth in claim 28, wherein the controlling further includes absorbing non-reacting amounts of said target cell solution from said reaction surface at a predetermined rate.
