Claims
- 1. A compound comprising the formula:
- 2. The compound according to claim 1 wherein X and Y are each independently selected from the group consisting of C(O), C(NH), S(O) and S(O)2.
- 3. The compound according to claim 1 wherein Y is C(O) and X is selected from the group consisting of C(O), C(NH), S(O) and S(O)2.
- 4. The compound according to claim 1 wherein n is 1.
- 5. The compound according to claim 1 wherein R7 is selected from the group consisting of a substituted or unsubstituted 5 or 6 membered ring, a substituted or unsubstituted amino, alkyl, and alkoxy moiety.
- 6. The compound according to claim 1 wherein R7 is a substituted or unsubstituted 5 or 6 membered ring.
- 7. The compound according to claim 1 wherein R7 is selected from the group consisting of —R23, —OR23, —(C1-8)alkyleneR23, —(C1-8)alkyleneOR23 and —(C1-8)alkyleneNR23R24; wherein R23 and R24 are each independently hydrogen or are each independently selected from the group consisting of a substituted or unsubstituted (C1-10)alkyl, (C3-12)cycloalkyl, hetero(C4-12)cycloalkyl, (C6-12)aryl, hetero(C5-12)aryl, (C9-12)bicycloalkyl, hetero(C9-12)bicycloalkyl, (C9-12)bicycloaryl and hetero(C8-12)bicycloaryl.
- 8. The compound according to claim 1 wherein R7 is substituted through available atoms by one to three radicals selected from a group consisting of substituted or unsubstituted (C1-6)alkyl, cyano, halo, nitro, halo-substituted(C1-6)alkyl, —NR20R20, —NR20C(O)OR20, —NR20C(O)NR20R20, —NR20C(NR20)NR20R20, —OR20, —SR20, —C(O)OR20, —C(O)NR20R20, —S(O)2NR20R20, —P(O)(OR20)OR20, —OP(O)(OR20)OR20, —NR20C(O)R20, —S(O)R20, —S(O)2R20, —(C1-8)alkyleneC(O)R20, —(C1-8)alkyleneNR20R20, —(C1-8)alkyleneNR20C(O)OR20, —(C1-8)alkyleneNR20C(O)NR20R20, —(C1-8)alkyleneNR20C(NR20)NR20R20, —(C1-8)alkyleneOR20, —(C1-8)alkyleneSR20, —(C1-8)alkyleneC(O)OR20, —(C1-8)alkyleneC(O)NR20R20, —(C1-8)alkyleneS(O)2NR20R20, —(C1-8)alkyleneP(O)(OR20)OR20, —(C1-8)alkyleneOP(O)(OR20)OR20, —(C1-8)alkyleneNR20C(O)R20, —(C1-8)alkyleneS(O)R20, —(C1-8)alkyleneS(O)2R20 and —(C1-8)alkyleneC(O)R20 where each R20 substituent is independently selected from the group consisting of hydrogen and a substituted or unsubstituted (C1-8)alkyl.
- 9. The compound according to claim 1 wherein R7 is a substituted or unsubstituted 5 membered ring.
- 10. The compound according to claim 1 wherein R7 a substituted or unsubstituted 6 membered ring.
- 11. The compound according to claim 1 wherein R7 comprises the formula
- 12. The compound according to claim 1 wherein R7 comprises the formula
- 13. The compound according to claim 12 wherein at least one of R10 and R11, or at least one of R16 and R17 is a cyano group.
- 14. The compound according to claim 12 wherein at least one of R10 and R11, or at least one of R16 and R17 is a cyano or boronic acid substituent.
- 15. The compound according to claim 1 wherein R7 comprises the formula —NR14R15 where R14 is a substituted or unsubstituted (C3-12)cycloalkyl and R15 is selected from the group consisting of hydrogen and a substituted or unsubstituted (C1-8)alkyl.
- 16. The compound according to claim 1 wherein R7 comprises the formula —NR14R15 where R14 is a substituted or unsubstituted hetero(C3-12)cycloalkyl and R15 is selected from the group consisting of hydrogen and a substituted or unsubstituted (C1-8)alkyl.
- 17. The compound according to claim 1 wherein R7 comprises the formula —NR14R15 where R14 is a substituted or unsubstituted (C6-12)aryl or hetero(C5-12)aryl and R15 is selected from the group consisting of hydrogen and a substituted or unsubstituted (C1-8)alkyl.
- 18. The compound according to claim 1 wherein R7 comprises the formula —NR14R15 where R14 is a substituted or unsubstituted (C9-12)bicycloaryl or hetero(C8-12)bicycloaryl and R15 is selected from the group consisting of hydrogen and a substituted or unsubstituted (C1-8)alkyl.
- 19. The compound according to claim 1 wherein R7 is selected from the group consisting of 2-cyano-pyrrolidin-1-yl, 4-fluoro-2-cyanopyrrolidin-1-yl, thiazolidin-3-yl and phenylamino.
- 20. The compound according to claim 1 wherein Z is a substituted or unsubstituted 5 membered ring.
- 21. The compound according to claim 1 wherein Z is a substituted or unsubstituted 6 membered ring.
- 22. The compound according to claim 1 wherein Z is a substituted or unsubstituted 5 membered heteroaryl ring.
- 23. The compound according to claim 1 wherein Z is a substituted or unsubstituted 5 membered heterocycloalkyl ring.
- 24. The compound according to claim 1 wherein the atom of Z that is bound to Y is a nitrogen atom.
- 25. The compound according to claim 1 wherein Y, taken together with the 5 or 6 membered ring of substituent Z forms an amino carbonyl.
- 26. The compound according to claim 1 wherein Y, taken together with the 5 or 6 membered ring of substituent Z forms an amino sulfonyl.
- 27. The compound according to claim 1 wherein Y, taken together with the 5 or 6 membered ring of substituent Z forms an amino sulfinyl.
- 28. The compound according to claim 1 wherein Y, taken together with the 5 or 6 membered ring of substituent Z forms an amino imino.
- 29. The compound according to claim 1 wherein Z is substituted with a non-hydrogen substituent at a 2 position on the ring, where the 1-position is attached to Y.
- 30. The compound according to claim 1 wherein Z is substituted with cyano at a 2-position on the ring, where the 1-position is attached to Y.
- 31. The compound according to claim 1 wherein the substituent Z comprises the formula
- 32. The compound according to claim 1 wherein at least one of R10 and R11, or at least one of R16 and R17 is a cyano group.
- 33. The compound according to claim 1 wherein at least one of R10 and R11, or at least one of R16 and R17 comprises a cyano or boronic acid substituent.
- 34. The compound according to claim 1 wherein Z comprises the formula —NR14R15 where R14 is a substituted or unsubstituted (C3-12)cycloalkyl or hetero(C3-12)cycloalkyl and R15 is selected from the group consisting of hydrogen and a substituted or unsubstituted (C1-8)alkyl.
- 35. The compound according to claim 1 wherein Z comprises the formula —NR14R15 where R14 is a substituted or unsubstituted (C6-12)aryl or hetero(C5-12)aryl and R15 is selected from the group consisting of hydrogen and a substituted or unsubstituted (C1-8)alkyl.
- 36. The compound according to claim 1 wherein Z comprises the formula —NR14R15 where R14 is a substituted or unsubstituted (C9-12)bicycloaryl or hetero(C8-12)bicycloaryl and R15 is selected from the group consisting of hydrogen and a substituted or unsubstituted (C1-8)alkyl.
- 37. The compound according to claim 1 wherein Z is selected from the group consisting of 2-cyano-pyrrolidin-1-yl, 4-fluoro-2-cyanopyrrolidin-1-yl, and thiazolidin-3-yl.
- 38. The compound according to claim 1 wherein R1 and R2 are taken together to form a ring.
- 39. The compound according to claim 1 wherein R5 and R6 are taken together to form a ring.
- 40. The compound according to claim 1 wherein one of R1 and R2 is taken together with one of R5 and R6 to form a ring.
- 41. The compound according to claim 1 wherein at least one of R1, R2, R5 and R6 is a substituted or unsubstituted —(C1-8)alkyleneR23, wherein R23 is selected from the group consisting of (C1-10)alkyl, (C3-12)cycloalkyl, hetero(C4-12)cycloalkyl, (C6-12)aryl, hetero(C5-12)aryl, (C9-12)bicycloalkyl, hetero(C9-12)bicycloalkyl, (C9-12)bicycloaryl and hetero(C8-12)bicycloaryl.

The compound according to claim 1 wherein at least one of R5 and R6 is selected from the group consisting of a substituted or unsubstituted —OR23 or a substituted or unsubstituted —(C1-8)alkyleneR23, wherein R23 is selected from the group consisting of (C1-10)alkyl, (C3-12)cycloalkyl, hetero(C4-12)cycloalkyl, (C6-12)aryl, hetero(C5-12)aryl, (C9-12)bicycloalkyl, hetero(C9-12)bicycloalkyl, (C9-12)bicycloaryl and hetero(C8-12)bicycloaryl.

The compound according to claim 1 wherein at least one of R1, R2, R5 and R6 is substituted through available atoms by one to three radicals selected from the group consisting of (C1-6)alkyl, cyano, halo, nitro, halo-substituted(C1-6)alkyl, —NR20R20, —NR20C(O)OR20, —NR20C(O)NR20R20, —NR20C(NR20)NR20R20, —OR20, —SR20, —C(O)OR20, —C(O)NR20R20, —S(O)2NR20R20, —P(O)(OR20)OR20, —OP(O)(OR20)OR20, —NR20C(O)R20, —S(O)R20, —S(O)2R20, —(C1-8)alkyleneC(O)R20, —(C1-8)alkyleneNR20R20, —(C1-8)alkyleneNR20C(O)OR20, —(C1-8)alkyleneNR20C(O)NR20R20, —(C1-8)alkyleneNR20C(NR20)NR20R20, —(C1-8)alkyleneOR20, —(C1-8)alkyleneSR20, —(C1-8)alkyleneC(O)OR20, —(C1-8)alkyleneC(O)NR20R20, —(C1-8)alkyleneS(O)2NR20R20, —(C1-8)alkyleneP(O)(OR20)OR20, —(C1-8)alkyleneOP(O)(OR20)OR20, —(C1-8)alkyleneNR20C(O)R20, —(C1-8)alkyleneS(O)R20, —(C1-8)alkyleneS(O)2R20 and —(C1-8)alkyleneC(O)R20 where each R20 substituent is independently selected from the group consisting of hydrogen and a substituted or unsubstituted (C1-8)alkyl.
- 44. The compound according to claim 1 wherein at least one of R1, R2, R5 and R6 is substituted through available atoms by one to three radicals selected from the group consisting of —NH2, —NHC(NH)NH2, —OH, —SH, —C(O)OH and —C(O)NH2.
- 45. The compound according to claim 1 wherein at least one of R1 and R2 is hydrogen.
- 46. The compound according to claim 1 wherein both R1 and R2 are hydrogen.
- 47. The compound according to claim 1 wherein at least one of R5 and R6 is hydrogen.
- 48. The compound according to claim 1 wherein both R5 and R6 are hydrogen.
- 49. The compound according to claim 1 wherein R3 is hydrogen.
- 50. The compound according to claim 1 wherein R3 is a substituted or unsubstituted (C1-10)alkyl.

- 52. The compound according to claim 1 wherein the compound is in the form of a pharmaceutically acceptable salt.

- 54. The compound according to claim 1 wherein the compound comprises a single stereoisomer.
