Claims
- 1. A pharmaceutical composition for drug release comprising a multipolymeric drug delivery vehicle selected from the group consisting of microparticles, microcapsules, nanoparticles, and microscopic gelled solid objects of slab geometry, said vehicle comprising a core having at least two charged polymers and a shell surrounding the core, said shell having at least two polymers of opposite charge to the charge of the core polymers, the vehicle exhibiting a charged surface, and wherein said core further comprises covalently crosslinked conjugates of a drug and one or more polymers.
- 2. The composition of claim 1 wherein said drug exhibits a charge and becomes an integral part of said vehicle.
- 3. The composition of claim 1 wherein said conjugate is of neutral charge and said core further comprises at least two charged polymers.
- 4. The composition of claim 1 wherein the drug delivery vehicle is a nanoparticle or microparticle having droplet forming core polymers in which the droplet forming core polymers and the drug are crosslinked.
- 5. The composition of claim 1 wherein the vehicle is enzymatically degradable.
- 6. The composition of claim 1 in which the drug is selected from the group consisting of genes, antigens, small peptides, and proteins.
- 7. The composition of claim 1 in which the crosslinking between the drug and polymer is a Schiff-base crosslink.
- 8. A pharmaceutical composition for drug release comprising a multipolymeric drug delivery vehicle selected from the group consisting of microparticles, microcapsules, nanoparticles and microscopic gelled solid objects of slab geometry, said vehicle comprising:(a) a core having at least two anionically charged polymers and further comprising covalently crosslinked conjugates of a drug and one or more polymers; and (b) a shell surrounding said core, said shell comprising at least two cationically charged polymers and further having both an inner region in which said cationically charged polymers complex with said anionically charged polymers and an outer peripheral region exhibiting a positive charge.
- 9. A pharmaceutical composition for drug release comprising a multipolymeric drug delivery vehicle selected from the group consisting of microparticles, microcapsules, nanoparticles and microscopic gelled solid objects of slab geometry, said vehicle comprising:(a) a core having at least two cationically charged polymers and further comprising covalently crosslinked conjugates of a drug and one or more polymers; and (b) a shell surrounding said core, said shell comprising at least two anionically charged polymers and further having both an inner region in which said anionically charged polymers complex with said cationically charged polymers and an outer peripheral region exhibiting a positive charge.
- 10. A pharmaceutical composition for drug release comprising a multipolymeric drug delivery vehicle selected from the group consisting of microparticles, microcapsules, nanoparticles, and microscopic gelled solid objects of slab geometry, said vehicle comprising a core having at least one charged polymer and a shell surrounding the core, said shell having at least one polymer of opposite charge to the charge of the core polymer, the vehicle exhibiting a charged surface, and wherein said core further comprises covalently crosslinked conjugates of a drug and of one or more polymers and one of said core or said shell further comprises at least two polyanionic polymers and the other of said core or said shell comprises at least one polycationic polymer and at least one small inorganic cation.
