Claims
- 1. A kit for measuring enzymes capable of forming or hydrolyzing amide bonds, or the amine, amide, or carboxylic acid substrates thereof, comprising premeasured amounts of a bidentate aromatic heterocyclic nitrogen-containing ligand of ruthenium or osmium reagent and a standard comprising an enzyme capable of forming or hydrolyzing amide bonds, or the amine, amide, or carboxylic acid substrate thereof, wherein the premeasured amounts are sufficient to perform a single sample measurement.
- 2. The kit according to claim 1 wherein the kit contains a series of containers having the premeasured amounts of reagents within them.
- 3. The kit according to claim 2 wherein the series of containers are prepared for use in an automated fashion.
- 4. The kit according to claim 1 wherein the standard is an amine, amide, or carboxylic acid substrate of an enzyme capable of forming or hydrolyzing an amide.
- 5. The kit according to claim 1 wherein the standard is an enzyme capable of forming or hydrolyzing amide bonds.
- 6. The kit according to claim 4 wherein the standard is a known amount of amine, amide, or carboxylic acid substrate of an enzyme capable of forming or hydrolyzing amide or ester bonds.
- 7. The kit according to claim 5 wherein the standard is a known amount of an enzyme capable of forming or hydrolyzing amide bonds.
- 8. The kit according to claim 1 wherein the bidentate aromatic heterocyclic nitrogen containing ligand of ruthenium is Ru(bpy).sub.3.sup.2+.
- 9. A method for measuring the presence of an enzyme capable of forming or hydrolyzing amide bonds, comprising:
- (a) contacting a sample suspected of containing said enzyme with an amine, amide, or carboxylic acid substrate thereof and a bidentate aromatic heterocyclic nitrogen-containing ligand of ruthenium or osmium reagent;
- (b) subjecting the contacted sample of step (a) to conditions which result in electrochemiluminescence; and
- (c) measuring resulting luminescence.
- 10. The method of claim 9, wherein the sample is a biological material selected from urine, pus, secretions, throat swab, blood or serum.
- 11. The method to claim 9 further comprising:
- (d) performing steps (a) through (c) with a second sample known to contain none of said enzyme, and correlating the difference in luminescence between the two samples, if any, to the amount of enzyme in said first sample.
- 12. A method for measuring the presence of an amine, amide, or carboxylic acid substrate of an enzyme capable of forming or hydrolyzing amide bonds, comprising:
- (a) contacting a sample suspected of containing said amine, amide, or carboxylic acid substrate with said enzyme and a bidentate aromatic heterocyclic nitrogen-containing ligand of ruthenium or osmium reagent;
- (b) subjecting the contacted sample of step (a) to conditions which result in electrochemiluminescence;
- (c) measuring resulting luminescence.
- 13. The method of claim 12 further comprising:
- (d) performing steps (a) through (c) with a second sample known to contain none of said substrate, and correlating the difference in luminescence between the two samples, if any, to the amount of substrate in said first sample.
- 14. The method of claim 12 wherein the sample is a biological material selected from urine, pus, secretions, food, blood, throat swab or serum.
- 15. The method of claim 12 wherein the steps are performed in a continual fashion.
- 16. The method of either claim 12 wherein the process in performed in the absence of an added radical-forming reductant reagent.
- 17. The method according to claim 16 wherein the reductant reagent is tripropylamine.
- 18. The method of claim 13 wherein the bidentate aromatic heterocyclic nitrogen-containing ligand of ruthenium or osmium is Ru(bpy).sub.3.sup.2+ or Os(bpy).sub.3.sup.2+, respectively.
