Claims
- 1. A dehydrogenase enzyme-aminoglycoside conjugate.
- 2. An enzyme-aminoglycoside conjugate of the formula; ##STR6## wherein: R.sup.2 is amino or hydroxyl
- R.sup.3 is hydroxyl;
- R.sup.4 and R.sup.5 are amino except for amikacin, where R.sup.4 is 2-hydroxy-4-aminobutyramido;
- R.sup.6 is hydrogen or methyl;
- R.sup.7 and R.sup.9 are hydroxyl;
- R.sup.8 is amino or methylamino;
- R.sup.10 is hydrogen or hydroxymethyl;
- R.sup.11 and R.sup.12 are hydrogen or hydroxy;
- R.sup.1 is hydroxymethyl or FCHNHF.sup.1 wherein F and F.sup.1 are the same or different and are hydrogen or methyl;
- with the proviso that any of the amino groups may be substituted with an inert acylating agent and one of the R groups when amino has a linking group involving a thioether replacing a hydrogen to form a covalent bond to ENZ;
- ENZ is an enzyme; and
- z is on the average from about 1 to 20.
- 3. An enzyme-aminoglycoside conjugate according to claim 2 wherein said linking group is of the formula: ##STR7## wherein: Q is C.dbd.W, wherein W is O, NH or S
- T is a linking group having at least one carbon atom and not more than nine atoms other than hydrogen
- a is zero or one;
- Y and Y.sup.1 are O, NH or S;
- A is NH or O;
- D is a chain of from one to nine atoms other than hydrogen;
- k and m are zero or one.
- 4. An enzyme-aminoglycoside conjugate of the formula according to claim 3, wherein said enzyme is an oxidoreductase and said aminoglycoside is a gentamicin.
- 5. An enzyme-aminoglycoside conjugate of the formula according to claim 4, wherein said oxidoreductase is a dehydrogenase.
- 6. An enzyme-aminoglycoside conjugate of the formula according to claim 5, wherein said dehydrogenase is glucose-6-phosphate dehydrogenase.
- 7. An enzyme-aminoglycoside conjugate of the formula according to claim 3, wherein said enzyme is an oxidoreductase and said aminoglycoside is a tobramycin.
- 8. An enzyme-aminoglycoside conjugate of the formula according to claim 7, wherein said oxidoreductase is a dehydrogenase.
- 9. An enzyme-aminoglycoside conjugate of the formula according to claim 8, wherein said dehydrogenase is glucose-6-phosphate dehydrogenase.
- 10. A gentamicin-dehydrogenase enzyme conjugate.
- 11. A conjugate according to claim 10, wherein said enzyme is an oxidoreductase and said gentamicin is linked through a thioether group.
- 12. A tobramycin-enzyme conjugate.
- 13. A conjugate according to claim 12, wherein said enzyme is an oxidoreductase and said tobramycin is linked through a thioether group.
- 14. A gentamicin-enzyme conjugate wherein said gentamicin is linked at the 3-amino position with a linking group of the formula
- --COCH.sub.2 SCH.sub.2 CO(NHCH.sub.2 CO).sub.m --
- wherein m is zero or one.
- 15. A conjugate according to claim 14 wherein said enzyme is an oxidoreductase.
- 16. A tobramycin-enzyme conjugate, wherein said tobramycin is linked at the 6'-amino position with a linking group of the formula
- --COCH.sub.2 SCH.sub.2 CO(NHCH.sub.2 CO).sub.m --
- wherein m is zero or one.
- 17. A conjugate according to claim 15, wherein said enzyme is an oxidoreductase.
