Claims
- 1. A DNA construct encoding a rhamnogalacturonase enzyme, wherein the DNA construct comprises
- (a) the DNA sequence of nucleotides 64-1587 of SEQ ID NO:1;
- (b) a DNA sequence which hybridizes to the same probe as nucleotides 64-1587 of SEQ ID NO: 1 under conditions of presoaking in 5.times.SSC and prehybridizing for 1 hour at .about.40.degree. C. in a solution of 5.times.SSC, 5.times.Denhardt's solution, 50 mM sodium phosphate, pH 6.8, and 50 mg of denatured sonicated calf thymus DNA, followed by hybridization in the same solution supplemented with 50 .mu.Ci 32-P-dCTP labelled probe for 18 h at .about.40.degree. C., followed by washing three times in 2.times.SSC, 0.2% SDS at 40.degree. C. for 30 minutes; or
- (c) a DNA sequence encoding an amino acid sequence having amino acids 20-527 of the sequence of SEQ ID NO:2.
- 2. The DNA construct of claim 1, wherein the probe comprises nucleotides 64-1587 of SEQ ID NO:1.
- 3. The DNA construct of claim 1, wherein the DNA sequence is obtained from a strain of Aspergillus aculeatus.
- 4. The DNA construct of claim 1, wherein the DNA sequence is isolated from a DNA library of Aspergillus aculeatus CBS 101.43.
- 5. A DNA construct comprising a DNA sequence encoding a rhamnogalacturonase enzyme, wherein the rhamnogalacturonase enzyme exhibits the characteristics of:
- (a) cleaving a rhamnogalacturonan backbone, wherein galacturonic acids are left as the non-reducing ends;
- (b) depolymerizing hairy regions from a soy bean material; and
- (c) depolymerizing saponified hairy regions from a sugar beet material.
- 6. The DNA construct of claim 5 wherein the DNA sequence is obtained from a strain of Aspergillus aculeatus.
- 7. The DNA construct of claim 5, wherein the rhamnogalacturonase enzyme further exhibits the characteristics of:
- (d) a pH optimum above pH 5; and
- (e) a relative activity of at least 30% at a pH in the range of 5.5-6.5.
- 8. A recombinant expression vector comprising the DNA construct of claim 1.
- 9. A recombinant expression vector comprising the DNA construct of claim 5.
- 10. A cell comprising the recombinant expression vector of claim 8.
- 11. A cell comprising the recombinant expression vector of claim 9.
- 12. The cell of claim 11, wherein the cell is a eukaryotic cell selected from the group consisting of a fungal cell, a yeast cell, or a filamentous fungal cell.
- 13. The cell of claim 12, wherein the cell is an Aspergillus strain selected from Aspergillus niger or Aspergillus oryzae.
- 14. A method of producing a rhamnogalacturonase enzyme, the method comprising culturing the cell of claim 10 under conditions permitting the production of the enzyme, and recovering the enzyme from the culture.
- 15. A method of producing a rhamnogalacturonase enzyme, the method comprising culturing the cell of claim 11 under conditions permitting the production of the enzyme, and recovering the enzyme from the culture.
