Claims
- 1. A continuous process for producing ethanol by fermentation in an aqueous medium which comprises cultivating a bacterium of the genus Zymomonas, which is capable of producing ethanol by fermentation, in two stages, said process consisting essentially of:
- (i) producing a bacterial biomass of Zymomonas together with ethanol in an ethanol concentration range which does not substantially inhibit production of the Zymomonas cells in a medium containing a source of nitrogen and a source of carbon; and
- (ii) producing ethanol in the absence of substantial production of Zymomonas cells by the addition of fermentable sugar to the bacterial cell biomass produced in (i), wherein the sugar concentration in stage (ii) does not exceed about 6% w/v;
- wherein the two stages of the process are conducted in at least two fermenters with the bacterial cells produced by continuous culture in the first fermenter being continuously transferred to the second fermenter, and
- wherein the ethanol concentration in the effluent from the last fermenter is increased over ethanol concentration in a fermentation medium obtained by cultivating the bacterium in a single stage and wherein the ethanol reaches a steady stage concentration of about 10-11% w/v in stage (ii).
- 2. The process of claim 1, wherein in stage (i) the source of nitrogen is a mineral salt medium containing NH.sub.4.sup.+ ions as the sole source of nitrogen or an organic source of nitrogen supplemented with NH.sub.4.sup.+ ions.
- 3. The process of claim 2, wherein stage (ii) of the process is conducted in a fermenter in which the contents have a substantially constant volume, said fermenter having a continuous feed of the medium required for the second stage of the process.
- 4. The process of claim 1, wherein a portion of the bacterial cells from the second fermenter are removed therefrom and separated from the fermentation broth by continuous filtration or sedimentation and are then returned to stage (ii) to thereby incrase ethanol productivity in stage (ii).
- 5. The process as claimed in claim 4, wherein the bacterial cell and ethanol production stages are conducted at a pH of about 5.5 and a temperature in the range of about 28.degree.-33.degree. C., wherein the fermenter in stage (i) and the fermenter in stage (ii) have a working volumetric ratio of about 2:1, and wherein ethanol is recovered from the fermentation broth.
- 6. The process as claimed in claim 1, 2 or 4, wherein bacterial cell and ethanol production are conducted with a pH range of 4 to 8 and a temperature range of 20.degree.-40.degree. C.
- 7. The process as claimed in claim 1, 2 or 4, wherein the source of carbon is glucose.
- 8. The process as claimed in claim 1, 2 or 4, wherein the microorganism strain is Zymomonas mobilis ATCC 29191.
- 9. The process as claimed in claim 1, 2 or 4, wherein the microorganism is Zymomonas mobilis ATCC 10988.
- 10. The process as claimed in claim 1, 2 or 4, wherein the bacterial cell and ethanol production stages are conducted at a pH of about 5.5 and a temperature in the range of 28.degree.-33.degree. C.
- 11. The process as claimed in claim 1, wherein the bacterial cell and ethanol production stages are conducted at a pH of about 5.5 and a temperature in the range of about 28.degree.-33.degree. C., wherein the fermenter in stage (i) and the fermenter in stage (ii) having a working volumetric ratio of about 1:2.
