Claims
- 1. An isolated DNA construct comprising a polynucleotide at least about 150 nucleotides in length and having a sequence at least about 80% identical to SEQ. ID. No. 1 or the complement thereof, wherein sequence identity is determined by the Needleman and Wunsch algorithm using standard parameters.
- 2. The DNA construct of claim 1, wherein the polynucleotide has the sequence of SEQ. ID. No. 1.
- 3. The DNA construct of claim 1, further comprising a promoter operably linked to the polynucleotide.
- 4. The DNA construct of claim 3, wherein the polynucleotide is linked to the promoter in an antisense orientation.
- 5. The DNA construct of claim 3, wherein the promoter is a plant promoter.
- 6. The DNA construct of claim 5, wherein the promoter is a seed-specific promoter.
- 7. A transgenic plant comprising a recombinant expression cassette comprising a plant promoter operably linked to a polynucleotide at least about 150 nucleotides in length and having a sequence at least about 80% identical to SEQ. ID. No. 1, wherein sequence identity is determined by the Needleman and Wunsch algorithm using standard parameters.
- 8. The transgenic plant of claim 7, wherein the plant promoter is a heterologous promoter.
- 9. The transgenic plant of claim 7, wherein the polynucleotide is linked to the promoter in an antisense orientation.
- 10. The transgenic plant of claim 7, wherein the plant is Brassica napus.
- 11. The transgenic plant of claim 7, wherein the polynucleotide has the sequence of SEQ. ID. No. 1.
- 12. A method of increasing very long chain fatty acid content in a plant, the method comprising:
- introducing into plant tissue a recombinant expression cassette comprising a plant promoter operably linked to a full-length polynucleotide having a sequence at least about 80% identical to SEQ ID No:1, wherein sequence identity is determined by the Needleman and Wunsch algorithm using standard parameters;
- regenerating the plant tissue into a whole plant, whereby the regenerated plant transcribes the polynucleotide; and
- selecting plants having an increased content of very long chain fatty acids.
- 13. The method of claim 12, wherein the plant tissue is from Brassica.
- 14. The method of claim 12, wherein the recombinant expression cassette is introduced into the plant tissue using Agrobacterium.
- 15. The method of claim 12, wherein the polynucleotide is linked to the promoter in a sense orientation.
- 16. The method of claim 12, wherein the polynucleotide has the sequence of SEQ. ID. No. 1.
- 17. The method of claim 12, wherein the promoter is a seed-specific promoter.
- 18. An isolated polynucleotide which encodes a polypeptide having the sequence of SEQ. ID. No. 2.
- 19. The DNA construct of claim 1, wherein the polynucleotide has the sequence of SEQ. ID. No. 3 or the complement thereof.
- 20. A recombinant DNA molecule comprising a plant promoter operably linked to a polynucleotide sequence at least about 150 nucleotides in length and having a sequence at least about 80% identical to SEQ. ID. No. 1, wherein sequence identity is determined by the Needleman and Wunsch algorithm using standard parameters and wherein the plant promoter is heterologous to the polynucleotide sequence.
- 21. The recombinant DNA molecule of claim 20, wherein the polynucleotide has the sequence of SEQ. ID. No. 1.
- 22. A recombinant expression cassette comprising a plant promoter operably linked to a polynucleotide sequence at least about 150 nucleotides in length and having a sequence at least about 80% identical to SEQ. ID. No. 1, wherein sequence identity is determined by the Needleman and Wunsch algorithm using standard parameters and wherein the plant promoter is heterologous to the polynucleotide sequence.
- 23. The recombinant expression cassette of claim 22, wherein the polynucleotide has the sequence of SEQ. ID. No. 1.
- 24. The recombinant DNA molecule of claim 20, wherein the polynucleotide has the sequence of SEQ ID NO:3.
