Claims
- 1. A method of identifying transformed plant cells comprising the steps of:
a) introducing into a plant cell at least one copy of an expression cassette comprising a coding region that codes for a fumonisin esterase enzyme operably linked to a promoter; b) introducing a fumonisin or a structurally related analog of fumonisin into the culture medium; and c) identifying transformed cells as the surviving cells in the culture.
- 2. The method of claim 1 wherein the fumonisin esterase enzyme comprises a coding sequence capable of hybridizing under stringent conditions to SEQ ID NO: 11 or SEQ ID NO: 15.
- 3. The method of claim 1 wherein the fumonisin esterase enzyme comprises the coding sequence of SEQ ID NO: 11 or SEQ ID NO: 15.
- 4. The method of claim 1 wherein the plant cell is a monocot plant cell.
- 5. The method of claim 1 wherein the plant cell is a dicot plant cell.
- 6. The method of claim 1 wherein the plant cell is selected from the group consisting of soybean, maize, sorghum, alfalfa, rice, clover, cabbage, banana, coffee, celery, tobacco, cowpea, cotton, melon and pepper.
- 7. A method of identifying transformed plant cells comprising the steps of:
a) introducing into a plant cell at least one copy of an expression cassette comprising a coding region that codes for a fumonisin esterase enzyme operably linked to a promoter; b) culturing the cell on medium containing a phytohormone linked to TCA, such that the phytohormone is inactive unless cleaved by an esterase; and c) identifying transformed cells as the surviving cells in the culture.
- 8. The method of claim 7 wherein the flimonisin esterase enzyme comprises a coding sequence capable of hybridizing under stringent conditions to SEQ ID NO: 11 or SEQ ID NO: 15.
- 9. The method of claim 7 wherein the fumonisin esterase enzyme comprises the coding sequence of SEQ ID NO: 11 or SEQ ID NO: 15.
- 10. The method of claim 7 wherein the plant cell is a monocot plant cell.
- 11. The method of claim 7 wherein the plant cell is a dicot plant cell.
- 12. The method of claim 7 wherein the plant cell is selected from the group consisting of soybean, maize, sorghum, alfalfa, rice, clover, cabbage, banana, coffee, celery, tobacco, cowpea, cotton, melon and pepper.
