Claims
- 27. An isolated polynucleotide encoding a lignin biosynthetic enzyme selected from the group consisting of: 4-coumarate:CoA ligase, cinnamate-4-hydroxylase, and ferulate-5-hydroxylase, wherein said encoded enzyme is functional in Zea mays and wherein said polynucleotide comprises at least 85% sequence identity, as determined by the GAP program under default parameters, to the full length of a polynucleotide selected from the group consisting of SEQ ID NOS: 21, 26, and 76.
- 2. The isolated nucleic acid of claim 27, wherein said polynucleotide has a sequence selected from the group consisting of SEQ ID NOS: 21, 26, and 76.
- 3. A recombinant expression cassette, comprising a nucleic acid of claim 27 operably linked to a promoter.
- 4. The recombinant expression cassette of claim 3, wherein said nucleic acid is operably linked in antisense orientation to said promoter.
- 5. A host cell introduced with the recombinant expression cassette of claim 3.
- 6. The host cell of claim 5, wherein said host cell is a sorghum (Sorghum bicolor) or maize (Zea mays) cell.
- 7. The isolated nucleic acid of claim 27, wherein the polynucleotide is DNA.
- 20. A transgenic plant comprising a recombinant expression cassette comprising a plant promoter operably linked to an isolated nucleic acid of claim 27.
- 21. The transgenic plant of claim 20, wherein said plant is Zea mays.
- 22. A transgenic seed comprising a recombinant expression cassette comprising an isolated polynucleotide of claim 27.
- 23. The transgenic seed of claim 22, wherein the seed is from Zea mays.
- 28. An isolated polynucleotide encoding a lignin biosynthetic enzyme selected from the group consisting of: 4-coumarate:CoA ligase, cinnamate-4-hydroxylase, and ferulate-5-hydroxylase, wherein said encoded enzyme is functional in Zea mays and wherein said polynucleotide encodes a polypeptide selected from the group consisting of SEQ ID NOS: 3, 8, and 73.
- 29. An isolated polynucleotide encoding a lignin biosynthetic enzyme selected from the group consisting of: 4-coumarate:CoA ligase, cinnamate-4-hydroxylase, and ferulate-5-hydroxylase, wherein said encoded enzyme is functional in Zea mays and wherein said polynucleotide is amplified from a Zea mays nucleic acid library using primers which selectively hybridize, under stringent hybridization conditions comprising a wash in 0.1×SSC at about 60-65° C., to loci within a polynucleotide of SEQ ID NO: 21, 26, or 76, respectively.
- 33. An isolated polynucleotide encoding a lignin biosynthetic enzyme selected from the group consisting of: 4-coumarate:CoA ligase, cinnamate-4-hydroxylase, and ferulate-5-hydroxylase, wherein said encoded enzyme is functional in Zea mays and wherein said polynucleotide selectively hybridizes, under stringent hybridization conditions comprising a wash in 0.1×SSC at about 60-65° C., to the complement of a polynucleotide selected from the group consisting of SEQ ID NOS: 21, 26, and 76.
- 34. An isolated polynudleotide encoding a lignin biosynthetic enzyme selected from the group consisting of: 4-coumarate:CoA ligase, cinnamate-4-hydroxylase, and ferulate-5-hydroxylase, wherein said encoded enzyme is functional in Zea mays and wherein said polynucleotide is selected from the group consisting of SEQ ID NOS: 21, 26, and 76.
