Abstract
A glucocerebroside compound, a pharmaceutical composition thereof, and the use of the glucocerebroside compound and the pharmaceutical composition thereof in the preparation of drugs for preventing or treating immune-related diseases.
Description
FIELD OF THE INVENTION

The invention belongs to the field of medicine, and specifically relates to a glucocerebroside compound, a pharmaceutical composition thereof and use thereof. Further, the glucocerebroside compound and the pharmaceutical composition thereof can be used as a drug for preventing or treating immune-related diseases.


BACKGROUND ART

Cerebroside or glycoceramide belongs to glycosphingolipids, mainly found in animals. It is also distributed in plants and microorganisms. Cerebroside is composed of three parts: glycosyl, fatty acid chain and sphingosine chain. Although these compounds are all composed of three parts: glycosyl, fatty acid chain and sphingosine chain, they have structural diversity due to different types of sugars, different lengths and branching degrees of glycosidic bonds, fatty acid chains and sphingosine chains, different number and positions of hydroxyl groups and double bonds, etc. (J. Nat. Prod. 2017, 80, 6, 1734-1741) Compounds with different structures often have different types or degrees of pharmacological activity, so cerebroside compounds have been reported to have different degrees of antitumor, antiviral, antimicrobial, and neuroprotective activities (Xu Jie, Studies on the Purification, Analysis and Bioactivities of Cerebrosides from Sea Cucumbers, PhD thesis of Ocean University of China, 2011). Most of the glycosyls in cerebrosides are galactose and glucose, so they are divided into galactosylcerebrosides and glucocerebrosides according to the type of glycosyls. The earliest discovery was galactocerebroside, and its structure and activity were reported more, while the glucocerebroside discovered later was relatively less reported.



Cordyceps sinensis is a dry complex of the stromata and larval corpses of the ergot fungus Cordyceps sinensis (BerK.) Sacc. parasitizing on the larvae of bat moths. It has various pharmacological effects such as regulating immunity, anti-tumor, anti-oxidation, and anti-aging. Literature reports that the chemical constituents of Cordyceps sinensis mainly include polysaccharides, proteins, sugar alcohols, nucleosides, amino acids, sterols and organic acids. Extracting new active ingredients from Cordyceps sinensis can provide scientific basis for the comprehensive quality evaluation of Cordyceps sinensis and the development and application of related products.


SUMMARY

The applicant of the present application conducted an in-depth analysis of the ethyl acetate part of Cordyceps sinensis, found that it contained cerebroside components, and they were separated and purified to obtain 4 new cerebroside compound monomers. The immunomodulatory activity of these cerebroside monomers was studied, and it was found that they had excellent immunosuppressive activity.


The purpose of the present invention is to provide a new class of cerebroside compound with immunosuppressive activity and the pharmaceutical composition thereof. The compound and the pharmaceutical composition thereof arc used to prevent treat immune-related diseases.


In one aspect, the present invention provides a compound having the structure described in general formula (I), (II), (III) or (IV), or a solvate, a metabolite, a pharmaceutically acceptable salt or a prodrug thereof,

In another aspect, the present invention provides a pharmaceutical composition, which comprises the compound described in the present invention, and a pharmaceutically acceptable adjuvant.


In some embodiments, the pharmaceutical composition of the present invention further comprises other additional therapeutic agent selected from a chemotherapeutic agent or anti-proliferative agent, an anti-inflammatory drug, an immunomodulatory or immunosuppressive agent, a neurotrophic factor, an agent for treating cardiovascular disease, an agent for treating diabetes, and an agent for treating immunodeficiency disorders.


In another aspect, the present invention provides the use of the compound of the present invention and the pharmaceutical composition of the present invention in the manufacture of a medicament for preventing or treating immune-related diseases.


In some embodiments, the immune-related disease is organ transplantation anti-rejection or autoimmune disease, wherein the autoimmune disease is lupus, multiple sclerosis, amyotrophic lateral sclerosis, rheumatoid arthritis, psoriasis, complications from organ transplantation, xeno transplantation, diabetes, asthma, atopic dermatitis, autoimmune thyroid disorders, ulcerative colitis, Crohn's disease, Alzheimer's disease, leukemia or lymphoma.


In another aspect, the present invention provides the use of the compound of the present invention and the pharmaceutical composition of the present invention in the manufacture of a medicament as an immunosuppressant.


In another aspect, the present invention also provides the compound of the present invention or the pharmaceutical composition of the present invention for use in preventing or treating immune-related diseases.

In certain embodiments, the salt is a pharmaceutically acceptable salt. The term “pharmaceutically acceptable” means that the substance or composition must be chemically and/or toxicologically compatible with the other ingredients comprising the preparation and/or with the mammal being treated therewith.


The compounds of the invention, including their salts, may also be obtained in the form of their hydrates, or include other solvents used for their crystallization. The compounds of the present invention may inherently or by design form solvates with pharmaceutically acceptable solvents (including water); therefore, the invention also include solvated and unsolvated forms.


In other aspect, the present invention relates to a method of preparing, separating or purifying the compound of Formula (I), (II), (III) or (IV).

The present application discovers new cerebroside compounds in Cordyceps sinensis for the first time, and these compounds have excellent immunosuppressive activity.

The term “alkenyl” refers to a linear or branched chain monovalent hydrocarbon radical of 2 to 12 carbon atoms, or 2 to 8 carbon atoms, or 2 to 6 carbon atoms, or 2 to 4 carbon atoms, with at least one site of C—C is a sp2 double bond, wherein the alkenyl radical may be independently substituted with one or more substituents described herein, including radicals having “cis” and “trans” orientations, or “Z” and “E” orientations. In an embodiment of the present invention, the alkenyl group contains 2-6 carbon atoms. The term “cycloalkyl” refers to a saturated monovalent carbocyclic ring system. In an embodiment of the present invention, the cycloalkyl group contains 3-6 carbon ring atoms, specific examples of which include cyclopropyl, cyclopentyl and cyclohexyl.


The term “solvate” refers to an association or complex of one or more solvent molecules and a compound disclosed herein. Some non-limiting examples of the solvent that form solvates include water, isopropanol, ethanol, methanol, dimethylsulfoxide (DMSO), ethyl acetate, acetic acid and ethanolamine. The term “hydrate” refers to the complex where the solvent molecule is water,


A “metabolite” is a product produced through metabolism in the body of a specified compound or salt thereof. The metabolites of a compound may be identified using routine techniques known in the art and their activities determined using tests such as those described herein. Such products may result for example from oxidation, reduction, hydrolysis, amidation, deamidation, esterification, deesterification, enzyme cleavage, and the like, of the administered compound. Accordingly, the invention includes metabolites of compounds disclosed herein, including metabolites produced by contacting a compound disclosed herein with a mammal for a sufficient time period.


The “pharmaceutically acceptable salt” used in the present invention refers to organic and inorganic salts of the compounds of the present invention. Pharmaceutically acceptable salts are well known in the art. For example, S. M. Berge et al., describe pharmaceutically acceptable salts in detail in J. Pharmaceutical Sciences, 1977, 66: 1-19, which is incorporated herein by reference. Some non-limiting examples of pharmaceutically acceptable and nontoxic salts include salts of an amino group formed with inorganic acids such as hydrochloric acid, hydrobromic acid, phosphoric acid, sulfuric acid and perchloric acid or with organic acids such as acetic acid, oxalic acid, maleic acid, tartaric acid, citric acid, succinic acid and malanic acid or by using other methods used in the art such as ion exchange. Other pharmaceutically acceptable salts include adipate, alginate, ascorbate, aspartate, benzenesulfonate, benzoate, bisulfate, borate, butyrate, camphorate, camphorsulfonate, cyclopentanepropionate, digluconate, dodecylsulfate, ethanesulfonate, formate, fumarate, glucoheptonate, glycerophosphate, gluconate, hemisulfate, heptanoate, hexanoate, hydroiodide, 2-hydroxy-ethanesulfonate, lactobionate, lactate, laurate, laurylsulfate, malate, malonate, methanesulfonate, 2-naphthalenesulfonate, nicotinate, nitrate, oleate, palmitate, pamoate, pectate, persulfate, 3-phenylpropionate, picrate, pivalate, propionate, stearate, thiocyanate, p-toluenesulfortate, undecanoate, valerate, and the like. Salts derived from appropriate bases include alkali metal ; alkaline earth metal, ammonium and N+(C1-4 alkyl)4 salts. This invention also envisions the quatemization of any basic nitrogen-containing groups of the compounds disclosed herein. Water or oil soluble or dispersable products may be obtained by such quatemization. Representative alkali or alkaline earth metal salts include sodium, lithium, potassium, calcium, magnesium, and the like. Further pharmaceutically acceptable salts further include, when appropriate, nontoxic ammoniu quaternary ammonium, and amine cations formed using counterions such as halide, hydroxide, carboxylate, sulfate, phosphate, nitrate, C1-8 sultanate or aryl sulfonate.


The term “prodrug” used in the present invention refers to a compound that is transfotmed in vivo into a compound of formula (I), (II), (III) or (IV). Such a transforination can be affected, for example, by hydrolysis of the prodrug form in blood or enzymatic transformation to the parent form in blood or tissue. Prodrugs of the compounds disclosed herein may be, for example, esters. Some common esters which have been utilized as prodrugs are phenyl esters, aliphatic (C1-24) esters, acyloxymethyl esters, carbonates, carbamates and amino acid esters. For example, a compound disclosed herein that contains a hydroxy group may be acylated at this position in its prodrug form. Other prodrug forms include phosphates, such as, those phosphate compounds derived from the phosphonation of a hydroxy group on the parent compound. The term “treatment” as used herein refers to both therapeutic and prophylactic treatment. For example, therapeutic treatment includes alleviating or ameliorating the progression, severity, and/or or duration of an immune-mediated condition, or amelioration of one or more symptoms (particularly, one or more identifiable symptoms) of an immune-mediated condition as a result of administering one or more therapies (e.g., one or more therapeutic agents (e.g., compounds and compositions of the invention)). In certain embodiments, therapeutic treatment includes amelioration of at least one measurable physical parameter of an immune-mediated condition. In other embodiments, therapeutic treatment includes inhibiting the progression of an immune-mediated condition physically, for example, by stabilizing discernible symptoms, or physiologically, for example, by stabilizing physical parameters, or both. In other embodiments, therapeutic treatment includes reducing or stabilizing the severity of immune-mediated diseases. The medicament of the invention can be used in the community to treat people already suffering from immune diseases to reduce the severity of symptoms and reduce the number of days they are sick.

The invention provides a class of novel cerebroside compound with immunosuppressive activity, the pharmaceutical composition thereof, and the use thereof in the preparation of drugs for preventing or treating immune-related diseases.

The compounds or compositions of this invention may be administered by any suitable means. The present invention also provides a method of treating, preventing or alleviating immune-related diseases in a subject, the method comprises administering a therapeutically effective amount of the compound of the present invention or the pharmaceutical composition thereof to a subject. Moreover, the above-mentioned compounds provided by the present invention or their pharmaceutical compositions can be co-administered with other therapies or therapeutic agents. The mode of administration can be carried out simultaneously, sequentially or at certain time intervals. Wherein the other therapy or therapeutic agent is selected from a chemotherapeutic agent or anti-proliferative agent, an anti-inflammatory agent, an immunomodulatory or immunosuppressive agent, a neurotrophic factor, an agent for treating cardiovascular disease, an agent for treating diabetes, or an agent for treating immunodeficiency disorders.


The dosage of the compound or pharmaceutical composition required to implement the effects of treatment, prevention or delay usually depends on the specific compound to be administered, the patient, the specific disease or condition and its severity, the route and frequency of administration, etc., and needs to be determined by the attending physician according to the specific situation. EXAMPLE 1: SEPARATION AND PREPARATION OF CEREBROSIDE FRACTION (CSE-E7) FROM CORDYCEPS SINENSIS
1.1 Preparation Method of CSE-E at Ethyl Acetate Part in Cordyeeps sinensis

15 Kg of Cordyceps sinensis was ground, heated and refluxed with 120 L of water to extract for 3 times, each time for 2 h. The mixture was filtered, and the water extraction filtrate was combined to obtain the water extract. The residue was heated and refluxed with 120 L of 95% ethanol to extract for 3 times, each time for 2 h. The mixture was filtered, and the 95% ethanol extraction filtrate was combined to obtain the 95% ethanol extract. The water extract was adsorbed with 20 Kg HP20 macroporous adsorption resin, and then eluted with 250 L of 95% ethanol. The part not absorbed by HP-20 macroporous resin was concentrated under reduced pressure and the obtained extract was marked as CSW (yield was 27.6%), the 95% ethanol eluted part and the 95% ethanol extract were combined and concentrated under reduced pressure, and the resulting extract was marked as CSE (yield was 20.9%). The CSE part was added with water and stirred, and then extracted successively with cyclohexane and ethyl acetate to obtain CSE-C at the cyclohexane part (extraction yield was 55.0%) and CSE-E at ethyl acetate part (extraction yield was 1.2%).


1.2 Preparation Method of Cerebroside Fraction CSE-E7 Fraction From Cordyceps sinensis

CSE-E (38.10 g) was adsorbed and mixed with 50 g of silica gel (100-200 mesh, Qingdao Ocean Chemical Co., Ltd., Qingdao, China), eluted with 280 mL of petroleum ether, and the remaining sample that was still adsorbed by silica gel was evaporated and connected to the ODS column (500 g) (10.0*250 mm) (50 μm, YMC Co. Ltd, Tokyo, Japan), which was separated at medium and low pressure, and eluted with 5%, 10%, 30%, 50%, 70%, 90%, and 100% methanol-water in sequence, with 5 L for each gradient elution. The 100% methanol eluate was combined and concentrated under reduced pressure, and the resulting extract was CSE-E7 (1.4 g).


EXAMPLE 2: SEPARATION AND IDENTIFICATION OF MONOMERIC COMPOUNDS 1, 2, 3, and 4 IN THE CEREBROSIDE FRACTION CSE-E7 FROM CORDYCEPS SINENSIS

CSE-E7 (1.4 g) was dissolved in methanol, analyzed by preparative high-performance liquid chromatography, and separated by medium and low pressure ODS column chromatography (3.1×20.0 cm, Japan YMC Company). The mobile phase was eluted with a gradient of 85%-100% (v/v) methanol-water for 3 hours, and at the same time, the eluent was connected with a graduated centrifuge tube, about 30 mL into one tube, and every 15 tubes were combined into one fraction. A total of 7 sub-fractions were obtained, namely F7.1, F7.2, F7.3, F7.4, F7.5, F7.6, F7.7. Fraction F7.4 (10.8 mg) was separated by a semi-preparative chromatographic column Phenomenex Biphenyl (10.0×250 mm, 5 μm) and eluted isocratically with a mobile phase of 85% methanol-ater (v/v) at a flow rate of 3 L/min to obtain compound 2 (tR: 43.8 min, 3.5 mg). Fraction F7.5 (93.8 mg) was separated by a semi-preparative chromatographic column Phenomenex Gemini C18 (10.0×250 mm, 5 μm) and eluted isocratically with a mobile phame of 92% methanol-water (b/v) at a flow rate of 3 mL/min to obtain compound 3 (tR: 51.6 min, 4.1 mg). Fraction F7.6 (523.4 mg) was separated by a preparative chromatographic column Phenomenex Gemini C18 (20.0×250 mm, 5 μm) and eluted isocratically with a mobile phase of 93% methanol-water (v/v) at a flow rate of 8 mL/min to obtain fraction F7.6.1 (tR: 50.0˜55.0 min), F7.6.2 (tR: 60.0˜65.0 min), F7.6.3 (tR: 66.0˜70.0 min) and F7.6.4 (tR: 74.0˜78.0 min). Fraction F7.6.2 (20.7 mg) was separated by a semi-preparative chromatographic column Phenomenex Biphenyl (10.0×250 mm, 5 μm) and eluted isocratically with a mobile phase of 87% methanol-water (b/v) at a flow rate of 3 mL/min to obtain compound 1 (tR: 31.6 min, 9.3 mg). Fraction F7.6.3 (8.8 mg) was separated by a semi-preparative chromatographic column Phenomenex Biphenyl (10.0×250 mm, 5 μm) and eluted isocratically with a mobile phase of 87% methanol-water (v/v) at a flow rate of 3 mL/min to obtain compound 4 (tR: 29.2 min, 2.5 mg).

EXAMPLE 3: THE IMMUNOSUPPRESSIVE ACTIVITY TEST OF COMPOUNDS 1, 2, 3, 4
3.1 Solution Preparation

Medium: 89% medium+10% fetal bovine serum+1% double antibody, stored at 2-8° C.


PBS: 1 pack of PBS was dissolved completely with IL of pure water, subpackaged and sterilized at high temperature.


10% CCK-8: 1 mL of CCK8 was added to 9 mL of culture medium to prepare 10% CCK-8 detection solution, which was prepared and used immediately.


ConA: 5 mg of ConA was accurately weighed and added with 1 mL of sterile PBS to fully dissolve to prepare a 5 mg/mL mother solution, which was subpackaged into 50 μL per tube and stored at −20° C. When in use, after thawing, complete medium was used to prepare a working solution ith a concentration of 150 μg/mL.


3.2 Experimental Method

3.2.1 Cell acquisition: BABL/C male mice ere sacrificed after eyeball extraction and bloodletting, soaked in 75% ethanol for 10 mint, and the spleen was aseptically removed in an ultra-clean workbench, and placed in a petri dish filled with PBS. A syringe was used to absorb PBS to rinse the surface and interior of the spleen until the spleen turned white, then the spleen was gently torn up with tweezers. The spleen was gently crushed with a 5 mL syringe on the steel mesh to make a single-cell suspension, which was filtered through a 200-mesh sieve. The filtrate was centrifuged for 10 min (1000 rpm/min); after centrifugation, the supertnatant was discarded. After the cell pellet was resuspended with PBS, the mixture was centrifuged again for 10 min (1000 rpm/min), and the operation was repeated once; 2 mL of erythrocyte lysate was added to the cell pellet. After the mixture was mixed evenly and allowed to stand for 5 min, 8 mL of PBS was added to nx evenly, and then the mixture was centrifuged at 1000 rpm min for 5 min. After centrifugation, the supernatant was discarded, the cell pellet was resuspended with PBS, then the mixture was centrifuged again for 10 min (1000 rpm/min), and the operation was repeated once; after centrifugation, the supematant ias discarded, the cell pellet was suspended in 1 mL of RPMI-1640 complete medium (containing 10% fetal bovine serum and 1% double antibody). 2-3 mL medium was added to dilute. A cell counter was used for cell counting (new cell type), and complete medium was used to adjust the cell concentration to 2*106 cells/mL.


3.2.2 Stunple preparation: appropriate amount of compounds 1, 2, 3, and 4 were dissolved in DMSO respectively to prepare 64 mM mother solution, and then diluted with medium to obtain solutions with a concentration of 128 μM respectively, and then gradiently diluted with medium to obtain sample dilutions with concentrations of 64 μM, 32 μM, 16 μM, 8 μM, and 4 μM, respectively.


3.2.3 Detection of cell viability: the prepared cell suspension was added to a 96-well plate. The seeding cell density was 2*105 cells/well, and the added cell volume was 100 μL. Compounds 1, 2, 3, and 4 were added at 100 μL/well, with 5 replicate wells for each concentration, and then incubated in a 37° C. 5% CO2 incubator for 60 h, and then incubated in a 37° C. incubator for 4-5 hours with serum-free medium and 10% CCK-8. The absorbance (A) value of each well was detected at 450 nm by a inicroplate reader. A control group was set up: cells+1640 medium. The results showed that compared with the control group, compounds 1, 2, 3, and 4 showed no cytotoxic effect on spleen lymphocytes at various concentrations (64, 32, 16, 8, 4, and 2 μM).


3.2.4 Detection of cell proliferation inhibition: the prepared cell suspension was added to the 96-well plate. The seeding cell density was 2*105 cells/well, and the added cell volume was 100 μL. Samples were added at 100 μL, with 5 replicate wells for each concentration, and then incubated in a 37° C., 5% CO2 incubator for 12 h. 10 μL of the supernatant were carefully suck out. 10 μL ConA with a concentration of 150 μg/mL (final concentration: 7.5 μg/mL) was added to the corresponding group, and then the mixture was incubated in a 37° C., 5% CO2 incubator for 48 h, and finally incubated in a 37° C. incubator for 4-5 hours with serum-free medium and 10% CCK-8. The absorbance (A) value of each well was detected at 450 nm by a microplate reader. A negative control group and a model control group were set up, which were cells+1640 medium, cells+ConA+1640 medium, respectively.


3.2.5 Data processing: the proliferation rate (viability) of the sample to spleen hymphocytes was calculated according to the following formula:





Proliferation rate %=[As/Ac]×100%

    • As: Absorbance of sample group (spleen lymphocytes+samples to be tested+CCK-8)
    • Ac: Absorbance of control group (spleen lymphocytes+CCK-8)


The proliferation inhibition rate of the sample to spleen lymphocytes was calculated according to the following formula:





Proliferation inhibition rate %=[(Am−As)/(Am−Ac)]×100%

    • Am: Absorbance of model control group (spleen lymphocytes+ConA+CCK-8)
    • As: Absorbance of sample group (spleen lymphocytes+samples to be tested+ConA+CCK-8)
    • Ac: Absorbance of negative control group (spleen lymphocytes+CCK-8)


3.3 Experimental Results

The experimental results of compounds 1. 2, 3. 4 on the cell proliferation inhibition rate of Con A-induced spleen lymphocyte proliferation are shown in Table 3 below; according to the log value of the concentration, linear fitting was performed with the log value x and the inhibition rate y, the fitted curve could calculate the IC 50 , and the IC 50 values corresponding to compounds 1, 2, 3, and 4 were 4.0, 10.4, 19.5, and 1.5 μM, respectively.

It can be seen from Table 3 that the compounds of the present invention all show good immunosuppressive activity in inhibiting Con A-induced proliferation of mouse splenocytes.

  • 15. The compound of claim 11, having any one of the following structures, or a solvate, a metabolite, a pharmaceutically acceptable salt or a prodrug thereof,
  • 16. A pharmaceutical composition, comprising the compound of claim 11, and a pharmaceutically acceptable adjuvant.

The pharmaceutical composition of claim 16, further comprising other additional therapeutic agent selected from a chemotherapeutic agent or anti-proliferative agent, an anti-inflammatory drug, an immunomodulatory or immunosuppressive agent, a neurotrophic factor, an agent for treating cardiovascular disease, an agent for treating diabetes, and an agent for treating immunodeficiency disorders.
  • 18. A method of preventing or treating immune-related diseases, comprising administering to a subject in need thereof a therapeutically effective amount of the compound of claim 11.
  • 19. A method of preventing or treating immune-related diseases, comprising administering to a subject in need thereof a therapeutically effective amount of the pharmaceutical composition of claim 16.
  • 20. The method of claim 18, wherein the immune-related disease is organ transplantation anti-rejection or autoimmune disease, wherein the autoimmune disease is lupus, multiple sclerosis, amyotrophic lateral sclerosis, rheumatoid arthritis, psoriasis, complications from organ transplantation, xeno transplantation, diabetes, asthma, atopic dermatitis, autoimmune thyroid disorders, ulcerative colitis, Crohn's disease, Alzheimer's disease, leukemia or lymphoma.
  • 21. The method of claim 19, wherein the immune-related disease is organ transplantation anti-rejection or autoimmune disease, wherein the autoimmune disease is lupus, multiple sclerosis, amyotrophic lateral sclerosis, rheumatoid arthritis, psoriasis, complications from organ transplantation, xeno transplantation, diabetes, asthma, atopic dermatitis, autoimmune thyroid disorders, ulcerative colitis, Crohn's disease, Alzheimer's disease, leukemia or lymphoma.
  • 22. A method of immunosuppression,comprising administering to a subject in need thereof a therapeutically effective amount of the compound of claim 11.
  • 23. A method of immunosuppression,comprising administering to a subject in need thereof a therapeutically effective amount of the pharmaceutical composition of claim 16.
