Claims
- 1. A composition which comprises a nutrient medium that has been conditioned by exposure to an irradiated or non-irradiated immortalized human stromal cell line and secretes a cytokine profile selected from the group consisting of:
a) IL-7, IL-8, SCF, LIF, M-CSF, TGF-beta1, MIP-1alpha, G-CSF, and MCP-1; b) IL-7, IL-8, FLT3L, SCF, LIF, M-CSF, TGF-beta1, GM-CSF, MIP-1alpha, G-CSF, and MCP-1; c) IL-7, IL-8, IL-11, FLT3L, SCF, LIF, M-CSF, TGF-beta1, GM-CSF, MIP-1alpha, G-CSF, and MCP-1; d) IL-7, IL-8, IL-11, FLT3L, SCF, LIF, M-CSF, MIP-1alpha, G-CSF, and MCP-1; e) IL-7, IL-8, FLT3L, SCF, LIF, M-CSF, MIP-1alpha, G-CSF, and MCP-1; f) IL-7, IL-8, IL-11, FLT3L, SCF, LIF, M-CSF, GM-CSF, MIP-1alpha, G-CSF, and MCP-1; and g) L-7, IL-8, IL-11L, SCF, LIF, M-CSF, MIP-1alpha, G-CSF, and MCP-1.
- 2. The composition of claim 1, wherein the immortalized stromal cell line is selected from the group consisting of: HGS2.11, HGS2.52, HGS3.18, HGS3.30, HGS3.65, HGS3.66, HGS3.103, and HGS3.114.
- 3. The composition of claim 1, wherein the immortalized stromal cell line is HGS2.52.
- 4. The composition of claim 1, wherein the immortalized stromal cell line is HGS3.18.
- 5. The composition of claim 1, wherein the immortalized stromal cell line is HGS3.65.
- 6. The composition of claim 1, wherein the nutrient medium is supplemented with at least one exogenously supplied growth factor.
- 7. The composition of claim 1, wherein the nutrient medium is supplemented with serum or plasma.
- 8. The composition of claim 7, wherein the nutrient medium is supplemented with human serum or plasma.
- 9. The composition of claim 1, wherein the nutrient medium has been conditioned by exposure to the immortalized human stromal cell line for one to five days.
- 10. The composition of claim 1, wherein the conditioned nutrient medium has been concentrated.
- 11. The composition of claim 10, wherein the conditioned nutrient medium has been concentrated by ultrafiltration.
- 12. The composition of claim 10, wherein the conditioned nutrient medium has been concentrated from two- to ten-fold.
- 13. The composition of claim 1, wherein the nutrient medium has been conditioned by exposure to two or more different immortalized human stromal cell lines.
- 14. A human stromal cell line selected from the group consisting of: HGS2.11, HGS2.52, HGS3.18, HGS3.30, HGS3.65, HGS3.66, HGS3.103, and HGS3.114.
- 15. The human stromal cell line of claim 14, wherein the human stromal cell line is HGS2.11.
- 16. The human stromal cell line of claim 14, wherein the human stromal cell line is HGS2.52.
- 17. The human stromal cell line of claim 14, wherein the human stromal cell line is HGS3.18.
- 18. The human stromal cell line of claim 14, wherein the human stromal cell line is HGS3.30.
- 19. The human stromal cell line of claim 14, wherein the human stromal cell line is HGS3.65.
- 20. The human stromal cell line of claim 14, wherein the human stromal cell line is HGS3.66.
- 21. The human stromal cell line of claim 14, wherein the human stromal cell line is HGS3.103.
- 22. The human stromal cell line of claim 14, wherein the human stromal cell line is HGS3.114.
- 23. A method of culturing hematopoietic cells comprising, incubating said cells in the composition of claim 1.
- 24. A method for increasing the number of human hematopoietic cells in vitro, comprising: inoculating the hematopoietic cells from a blood product into a culture vessel containing the composition of claim 1 and culturing the cells under conditions for a time sufficient to increase the number of hematopoietic cells relative to the number of cells present initially in the blood product.
- 25. The method of claim 24, wherein the human hematopoietic cells are separated from mature hematopoietic cells present in a blood product prior to inoculating the culture vessel containing the conditioned culture medium.
- 26. The method of claim 24, further comprising the step of freezing the separated hematopoietic cells and thawing the cells prior to inoculating the culture vessel containing the conditioned culture medium.
- 27. The method of claim 23 including, subsequent to the step of culturing, harvesting the human hematopoietic cells from the culture medium.
- 28. The method of claim 24, further comprising subsequent to the step of harvesting, freezing the hematopoietic cells which have been increased in number in the conditioned medium culture.
- 29. The method of claim 24, wherein the blood product is bone marrow, umbilical cord blood, or peripheral blood.
- 30. The method of claim 24, wherein the culture medium includes at least one exogenously added growth factor.
- 31. The method of claim 24, wherein the cells are cultured under conditions for a time sufficient to achieve at least an approximately two-fold expansion in the number of hematopoietic cells relative to the number of cells present initially in the blood product.
- 32. A method for conditioning culture media, comprising: innoculating culture media with a human stromal cell line selected from the group consisting of HGS2.11, HGS2.52, HGS3.18, HGS3.30, HGS3.65, HGS3.66, HGS3.103, and HGS3.114, and culturing the innoculated cell line for a time sufficient for the conditioned media to increase the number of hematopoietic cells cultured in said media.
- 33. The method of claim 32, wherein said stromal cell line is HGS2.52.
- 34. The method of claim 32, wherein said stromal cell line is HGS3.18.
- 35. The method of claim 32, wherein said stromal cell line is HGS3.65.
- 36. The method of claim 32, wherein said stromal cell line has been irradiated.
- 37. The method of claim 32, wherein said stromal cell line has not been irradiated.
- 38. The conditioned culture media generated according to the method of claim 32.
- 39. The conditioned culture media of claim 38, wherein said media is supplemented with exogenous growth factors, serum, or plasma.
