Claims
- 1. A compound of Formula I:

A compound according to claim 1, wherein A is pyrrolyl, thienyl, benzofuranyl, benzothiophenyl, pyrazolyl, indazolyl, phenyl or indolyl which which in each case is unsubstituted or is substituted by alkyl having 1 to 8 C atoms, alkoxy having 1 to 8 C atoms, halogen, carboxy, oxo, nitro, oxide, alkoxycarbonyl having 2 to 8 C atoms, alkyl-sulfonyl having 1 to 8 C atoms, Ar, Ar—CO—, Ar-sulfonyl, Ar—O—, Ar-alkyl-O— wherein the alkylene group has 1 to 8 C atoms, Ar-thio, Hetero, cyano, trifluoromethyl, halogenated-alkyl having 1 to 8 C atoms, halogenated-alkoxy having 1 to 8 C atoms, amino, monoalkylamino having 1 to 8 C atoms, dialkylamino wherein each alkyl group has 1 to 8 C atoms, hydroxyl, hydroxyalkyl having 1 to 8 C atoms, hydroxyalkoxy having 2 to 8 C atoms, acyl, alkylthio having 1 to 8 C atoms, alkylsulphinyl having 1 to 8 C atoms, acylamido, and acyloxy.
- 3. A compound according to claim 1, wherein A is dihydrobenzofuranyl which is unsubstituted or substituted by alkyl, alkoxy, halogen, carboxy, oxo, nitro, alkoxycarbonyl, cyano, trifluoromethyl, amino, monoalkylamino, dialkylamino, hydroxyl, hydroxyalkyl, or combinations thereof.
- 4. A compound according to claim 1, wherein A is dihydrobenzodioxinyl which is unsubstituted or substituted by alkyl, alkoxy, halogen, carboxy, oxo, nitro, alkoxycarbonyl, cyano, trifluoromethyl, amino, monoalkylamino, dialkylamino, hydroxyl, hydroxyalkyl or combinations thereof.
- 5. A compound according to claim 1, wherein A is chromanyl which is substituted or substituted by alkyl, alkoxy, halogen, carboxy, oxo, nitro, alkoxycarbonyl, cyano, trifluoromethyl, amino, monoalkylamino, dialkylamino, hydroxyl, hydroxyalkyl or combinations thereof.
- 6. A compound according to claim 1, wherein A is dihydrobenzodioxepinyl, which is unsubstituted or substituted by alkyl, alkoxy, halogen, carboxy, oxo, nitro, alkoxycarbonyl, cyano, trifluoromethyl, amino, monoalkylamino, dialkylamino, hydroxyl, hydroxyalkyl or combinations thereof.
- 7 A compound according to claim 1, wherein A is indolyl which is unsubstituted or substituted by alkyl, alkoxy, halogen, carboxy, oxo, nitro, alkoxycarbonyl, cyano, trifluoromethyl, amino, monoalkylamino, dialkylamino, hydroxyl, hydroxyalkyl, or combinations thereof.

A compound according to claim 1, wherein A is phenyl substituted by a heterocylic group, wherein A is unsubstituted or substituted by alkyl, alkoxy, cycloalkyl, cycloalkylalkyl, halogen, carboxy, oxo, nitro, alkoxycarbonyl, cyano, trifluoromethyl, amino, monoalkylamino, dialkylamino, hydroxyl, hydroxyalkyl, —COR5, or combinations thereof.

A compound according to claim 1, wherein A is benzoxazinyl which is unsubstituted or substituted by alkyl, alkoxy, cycloalkyl, cycloalkylalkyl, halogen, carboxy, oxo, nitro, alkoxycarbonyl, cyano, trifluoromethyl, amino, monoalkylamino, dialkylamino, hydroxyl, hydroxyalkyl, or combinations thereof.
- 10. A compound according to claim 1, wherein A is indazolyl which is unsubstituted or substituted by alkyl, alkoxy, halogen, carboxy, oxo, nitro, alkoxycarbonyl, cyano, trifluoromethyl, amino, monoalkylamino, dialkylamino, hydroxyl, hydroxyalkyl, or combinations thereof.
- 11. A compound according to claim 1, wherein A is benzoimidazolyl which is unsubstituted or substituted by alkyl, alkoxy, halogen, carboxy, oxo, nitro, alkoxycarbonyl, cyano, trifluoromethyl, amino, monoalkylamino, dialkylamino, hydroxyl, hydroxyalkyl, or combinations thereof.

A compound according to claim 1, wherein A is phenylpiperazinyl which is unsubstituted or substituted by alkyl, alkoxy, cycloalkyl, cycloalkylalkyl, halogen, carboxy, oxo, nitro, alkoxycarbonyl, cyano, trifluoromethyl, amino, monoalkylamino, dialkylamino, hydroxyl, hydroxyalkyl, Ar or combinations thereof.

A compound according to claim 1, wherein A is benzothiazolyl which is unsubstituted or substituted by alkyl, alkoxy, cycloalkyl, cycloalkylalkyl, halogen, carboxy, oxo, nitro, alkoxycarbonyl, cyano, trifluoromethyl, amino, monoalkylamino, dialkylamino, hydroxyl, hydroxyalkyl, or combinations thereof.

A compound according to claim 1, wherein A is pyrrolyl which is unsubstituted or substituted by alkyl, alkoxy, cycloalkyl, cycloalkylalkyl, halogen, carboxy, oxo, nitro, alkoxycarbonyl, cyano, trifluoromethyl, amino, monoalkylamino, dialkylamino, hydroxyl, hydroxyalkyl, aryl which is unsubstituted or substituted by halogen, halogenated alkyl, or halogenated alkoxy, arylalkyl which is unsubstituted or substituted by halogen, halogenated alkyl, or halogenated alkoxy, or combinations thereof.

A compound according to claim 1, wherein A is pyrazolyl which is unsubstituted or substituted by alkyl, alkoxy, cycloalkyl, cycloalkylalkyl, halogen, carboxy, oxo, nitro, alkoxycarbonyl, cyano, trifluoromethyl, amino, monoalkylamino, dialkylamino, hydroxyl, hydroxyalkyl, Ar which is unsubstituted or substituted by halogen, halogenated alkyl, or halogenated alkoxy, arylalkyl which is unsubstituted or substituted by halogen, halogenated alkyl, or halogenated alkoxy, or combinations thereof.

A compound according to claim 1, wherein A is diazabicycloheptyl which is unsubstituted or substituted by alkyl, alkoxy, cycloalkyl, cycloalkylalkyl, halogen, carboxy, oxo, nitro, alkoxycarbonyl, cyano, trifluoromethyl, amino, monoalkylamino, dialkylamino, hydroxyl, hydroxyalkyl, or combinations thereof.

A compound according to claim 1, wherein A is benzodioxolyl which is unsubstituted or substituted by alkyl, alkoxy, cycloalkyl, cycloalkylalkyl, halogen, carboxy, oxo, nitro, alkoxycarbonyl, cyano, trifluoromethyl, amino, monoalkylamino, dialkylamino, hydroxyl, hydroxyalkyl, or combinations thereof.

A compound according to claim 1, wherein A is dihydrobenzodioinyl which is unsubstituted or substituted by alkyl, alkoxy, cycloalkyl, cycloalkylalkyl, halogen, carboxy, oxo, nitro, alkoxycarbonyl, cyano, trifluoromethyl, amino, monoalkylamino, dialkylamino, hydroxyl, hydroxyalkyl, or combinations thereof.
- 19. A compound according to claim 1, wherein R3 and R4 are each H.

- 37. A method of treating a patient suffering from dementia and other conditions with memory loss comprising administering to the patient an effective amount of a compound according claim 1.

A method of treating a patient suffering from memory impairment due to mild cognitive impairment due to aging, Alzheimer's disease, schizophrenia, Parkinson's disease, Huntington's disease, Pick's disease, Creutzfeld-Jakob disease, depression, aging, head trauma, stroke, CNS hypoxia, cerebral senility, or multiinfarct dementia comprising administering an effective amount of a compound according to claim 1.
- 39. A method of treating and/or preventing dementia in an Alzheimer's patient comprising administering to the patient a therapeutically effective amount of a compound according to claim 1 to inhibit the binding of an amyloid beta peptide with nAChRs.
- 40. A method of treating a patient for alcohol withdrawal or treating a patient with anti-intoxication therapy comprising administering to the patient an effective amount of a compound according to claim 1.
- 41. A method of treating a patient to provide for neuroprotection against damage associated with strokes and ischemia and glutamate-induced excitotoxicity comprising administering to the patient an effective amount of a compound according to claim 1.
- 42. A method of treating a patient suffering from nicotine addiction, pain, jetlag, obesity and/or diabetes, or a method of inducing smoking cessation in a patient comprising administering to the patient an effective amount of a compound according to claim 1.

A method of treating a patient suffering from mild cognitive impairment (MCI), vascular dementia (VaD), age-associated cognitive decline (AACD), amnesia associated w/open-heart-surgery, cardiac arrest, general anesthesia, memory deficits from exposure to anesthetic agents, sleep deprivation induced cognitive impairment, chronic fatigue syndrome, narcolepsy, AIDS-related dementia, epilepsy-related cognitive impairment, Down's syndrome, Alcoholism related dementia, drug/substance induced memory impairments, Dementia Puglistica (Boxer Syndrome), or animal dementia comprising administering to the patient an effective amount of a compound according to claim 1.
- 44. A method of selectively activating/stimulating α-7 nicotinic receptors in a mammal wherein such activation/stimulation has a therapeutic effect, comprising administering to an animal in need thereof an effective amount of a compound of according to claim 21.
- 45. A method of treating a patient suffering from psychotic diseases, neurodegenerative diseases involving a dysfunction of the cholinergic system, and conditions of memory and/or cognition impairment comprising administering to the patient an effective amount of a compound according to claim 1.
- 46. A method of treating a patient suffering from dementia and other conditions with memory loss comprising administering to the patient an effective amount of a compound according claim 21.
