Claims
- 1. A promoter element derived from a bacterial alcohol dehydrogenase gene, the promoter element being operatively linked to a heterologous gene encoding a heterologous protein, wherein said promoter element provides elevated expression of the heterologous protein as measured relative to the rate of protein expression of the heterologous gene under control of its endogenous native promoter.
- 2. The promoter element of claim 1 wherein the bacterial alcohol dehydrogenase gene derives from a mesophilic strain of bacteria.
- 3. The promoter element of claim 2 wherein said mesophilic strain of bacterium is Clostridium beijerinckii.
- 4. The promoter element of claim 1 wherein the bacterial alcohol dehydrogenase gene derives from a thermophilic strain of bacteria.
- 5. The promoter element of claim 4 wherein said thermophilic strain of bacterium is Thermoanaerobacter brockii.
- 6. The promoter element of claim 5, wherein said promoter element comprises a nucleic acid sequence having SEQ ID NO: 1, or an active fragment thereof.
- 7. The nucleic acid sequence of claim 6 wherein said active fragment thereof comprises SEQ ID NO: 2, or SEQ ID NO: 3.
- 8. The promoter element of claim 3, wherein said promoter element comprises a nucleic acid sequence having SEQ ID NO: 4, or an active fragment thereof.
- 9. A construct comprising the a promoter element of claim 1, further comprising a nucleic acid sequence encoding a heterologous protein operatively linked to the promoter element.
- 10. A host cell transfected with the construct of claim 9.
- 11. A vector useful for transfecting host cells comprising the promoter element of claim 1.
- 12. A host cell transfected with a vector according to claim 11.
- 13. The promoter element of claim 1, wherein the promoter is a recombinant nucleic acid sequence comprising the sequence described in SEQ ID NO: 1, SEQ ID NO: 4 or active fragment thereof.
- 14. The promoter element of claim 13, wherein the active fragment thereof comprises SEQ ID NO: 2 or SEQ ID NO: 3.
- 15. A method of constitutively expressing a heterologous protein in a host cell, the method comprising the steps of:
transfecting a host cell with a construct or expression vector comprising the promoter element of claim 1;identifying a transfected host cell that actively expresses the heterologous protein; and culturing the transfected host cell.
- 16. The method of claim 15, wherein said heterologous protein is obtained in active folded conformation.
- 17. The method of claim 15, wherein the heterologous protein is expressed in a host cell cultured on enriched medium.
- 18. The method of claim 15, wherein the heterologous protein is selected from the group consisting of a growth factor, a growth factor receptor, a peptide hormone, a hormone receptor, a cytokine, a cytokine receptor, an agonist, an antagonist of a growth factor, a hormone receptor, a clotting factor, an enzyme, an enzyme inhibitor, an extracellular matrix protein, a viral antigen, and an antibody.
- 19. The method of claim 18, wherein the heterologous protein is selected from the group consisting of a FGF growth factor, FGF agonists, FGF antagonists, FGF truncated molecules, FGF mutants, FGF receptor molecules, and a collagen.
- 20. The method of claim 19, wherein the heterologous protein is selected from the group consisting of FGF9, FGF-9-2, FGF2, mouse Collagen type X NCI domain, human Collagen type X NCI domain, human collagen VIII, and FGF receptor 3 extracellular domain.
- 21. A promoter element comprising a nucleic acid sequence of SEQ ID NO: 1, SEQ ID NO: 4 or an active fragment thereof, the promoter element providing elevated expression of a heterologous protein when operatively linked to a heterologous gene encoding of the heterologous protein as measured relative to the rate of protein expression of the heterologous gene under control of its endogenous native promoter.
- 22. The promoter element of claim 21, wherein said active fragment thereof is SEQ ID NO: 2 or SEQ ID NO: 3.
- 23. A construct comprising the a promoter element of claim 22, further comprising a nucleic acid sequence encoding a heterologous protein operatively linked to the promoter element.
- 24. A host cell comprising the construct of claim 23, expressing the heterologous protein.
- 25. The host cell of claim 24, wherein the heterologous protein expressed is selected from the group consisting of a FGF growth factor, FGF agonists, FGF antagonists, FGF truncated molecules, FGF mutants, FGF receptor molecules, and a collagen.
- 26. The host cell of claim 25, wherein the heterologous protein expressed is selected from the group consisting of FGF9, FGF-9-2, FGF2, mouse Collagen type X NCI domain, human Collagen type X NCI domain, human collagen VIII, and FGF receptor 3 extracellular domain.
