Claims
- 1. A composition comprising a skin graft, wherein said skin graft comprises human keratinocyte cells obtained from a cell line that originated with ATCC CRL 12191, wherein the cell line:(a) is immortalized; (b) is nontumorigenic; (c) forms cornified envelopes when induced to differentiate; (d) undergoes normal squamous differentiation in organotypic culture; and (e) maintains cell type-specific growth requirements.
- 2. The composition of claim 1, wherein said cell line type-specific growth requirements comprise (1) exhibition of morphological characteristics of normal human keratinocytes when cultured in standard keratinocyte growth medium in the presence of mitomycin C-treated 3T3 feeder cells; (2) dependence on epidermal growth factor for growth; and (3) inhibition of growth by transforming growth factor β1.
- 3. The composition of claim 1, wherein said cell line is in an organotypic culture.
- 4. A method for closing a wound comprising:(a) providing (i) a subject having a wound; and (ii) a skin graft of claim 1; (b) applying said skin graft to said subject under conditions such that said wound is closed.
- 5. The method of claim 4 wherein said wound is selected from the group consisting of an ulcer, burn, surgery wound and donor site wound.
