Claims
- 1. An isolated polynucleotide comprising an infectious cDNA encoding a North American PRRSV viral RNA transcript.
- 2. The isolated polynucleotide of claim 1, wherein the cDNA comprises a sequence having at least 90% sequence identity to SEQ ID NO: 1.
- 3. The isolated polynucleotide of claim 1, wherein the cDNA comprises SEQ ID NO: 1, or a degenerate variant of SEQ ID NO: 1.
- 4. A vector comprising the polynucleotide of claim 1.
- 5. A vector comprising the polynucleotide of claim 2.
- 6. A vector comprising the polynucleotide of claim 3.
- 7. The isolated polynucleotide of claim 2, further comprising one or more mutations, wherein at least one mutation comprises nucleotide substitution, insertion or deletion, or combinations thereof.
- 8. The isolated polynucleotide of claim 7, wherein at least one mutation is a non-silent mutation.
- 9. The isolated polynucleotide of claim 8, wherein the non-silent mutation is in at least one of ORF 5 and ORF 7.
- 10. The isolated polynucleotide of claim 2, further comprising at least one foreign polynucleotide.
- 11. The isolated polynucleotide of claim 10, wherein at least one foreign polynucleotide is inserted in at least one of:
(a) immediately upstream of ORF 2; and (b) the 3′ untranslated region.
- 12. The isolated polynucleotide of claim 10, wherein at least one foreign polynucleotide is inserted:
(a) between nucleotide positions 12181-12182 of SEQ ID NO: 1; or (b) between nucleotide positions 15371-15375 of SEQ ID NO: 1.
- 13. The isolated polynucleotide of claim 10, wherein at least one foreign polynucleotide encodes a porcine viral polypeptide.
- 14. The isolated polynucleotide of claim 13, wherein the viral polypeptide is a capsid protein of Porcine Circovirus Type II, or a portion thereof.
- 15. The isolated polynucleotide of claim 9, wherein at least one foreign polynucleotide comprises at least one polylinker sequence.
- 16. The isolated polynucleotide of claim 7, comprising at least one nucleotide deletion.
- 17. The isolated polynucleotide of claim 16 wherein the deletion is in at least one of the 3′ end of ORF 7 and the 5′ end of the 3′ untranslated region.
- 18. The isolated polynucleotide of claim 16 wherein at least one nucleotide is deleted:
(a) between nucleotide positions 15364-15370 of SEQ ID NO: 1; or (b) between nucleotide positions 15371-115375 of SEQ ID NO: 1.
- 19. A method of producing a full-length infectious North American PRRSV cDNA, comprising the steps of:
(a) reverse transcribing viral RNAs to obtain cDNA fragments; (b) cloning the cDNA fragments to form primary clones; (c) combining the primary clones to form intermediate clones; and (d) combining the intermediate clones to obtain a full-length infectious cDNA clone.
- 20. The method of claim 19 further comprising the step of introducing a nucleotide or polynucleotide substitution, insertion or deletion, or a combination thereof, in at least one of the primary or intermediate clones.
- 21. A method of producing infectious virus particles from an isolated full-length North American PRRSV cDNA clone comprising the steps of:
(a) producing a full-length infectious North American PRRSV cDNA clone; (b) inserting a eukaryotic promoter sequence upstream of the 5′ end of the PRRSV cDNA to form a replication-competent clone; (c) transforming host cells with the replication-competent clone; and (d) isolating infectious virus particles from the host cells of step (c).
- 22. A virus produced according to the method of claim 21.
- 23. A composition comprising the isolated polynucleotide of claim 1.
- 24. A composition comprising the isolated polynucleotide of claim 7.
- 25. A composition comprising a full-length infectious PRRSV cDNA produced according to the method of claim 19.
- 26. A composition comprising a full-length infectious PRRSV cDNA produced according to the method of claim 20.
