Claims
- 1. A compound of the formula: ##STR54## wherein: each of Q.sub.1 and Q.sub.2 are independently selected from phenyl, 5-6 membered aromatic heterocyclic ring systems having one nitrogen heteroatom;
- Q.sub.1 is substituted with 1 to 4 substituents, independently selected from halo; C.sub.1 -C.sub.3 alkyl; C.sub.1 -C.sub.3 alkyl substituted with NR'.sub.2, OR', CO.sub.2 R' or CONR'.sub.2 ; O--(C.sub.1 -C.sub.3)-alkyl; O--(C.sub.1 -C.sub.3)-alkyl substituted with NR'.sub.2, OR', CO.sub.2 R' or CONR'.sub.2 ; NR'.sub.2 ; OCF.sub.3 ; CF.sub.3 ; NO.sub.2 ; CO.sub.2 R'; CONR'; SR'; S(O.sub.2)N(R').sub.2 ; SCF.sub.3 ; or CN; and
- Q.sub.2 is optionally substituted with up to 4 substituents, independently selected from halo; C.sub.1 -C.sub.3 straight or branched alkyl; C.sub.1 -C.sub.3 straight or branched alkyl substituted with NR', NR'.sub.2, OR', CO.sub.2 R', or CONR'.sub.2 ; O--(C.sub.1 -C.sub.3)-alkyl; O--(C.sub.1 -C.sub.3)-alkyl substituted with NR', NR'.sub.2, OR', CO.sub.2 R' or CONR'.sub.2 ; NR'.sub.2 ; OCF.sub.3 ; CF.sub.3 ; NO.sub.2 ; CO.sub.2 R'; CONR'; SR'; S(O.sub.2)N(R').sub.2 ; SCF.sub.3 ; or CN;
- wherein R' is selected from hydrogen, (C.sub.1 -C.sub.3)-alkyl or (C.sub.2 -C.sub.3)-alkenyl or alkynyl; and
- X is selected from --S--, --O--, --S(O.sub.2)--, --S(O)--, --C(O)--, --N(R)--, or --C(R).sub.2 --;
- each R is independently selected from hydrogen or (C.sub.1 -C.sub.3) alkyl;
- Y is C;
- A is CR';
- n is 1; and
- R.sub.1 is selected from hydrogen, (C.sub.1 -C.sub.3)-alkyl, OH, or O--(C.sub.1 -C.sub.3)-alkyl.
- 2. A compound of the formula: ##STR55## wherein: each of Q.sub.1 and Q.sub.2 are independently selected from phenyl, 5-6 membered aromatic heterocyclic ring systems, wherein each of said heterocyclic rings has a single nitrogen heteroatom;
- Q.sub.1 is substituted with 1 to 4 substituents, independently selected from halo; C.sub.1 -C.sub.3 alkyl; C.sub.1 -C.sub.3 alkyl substituted with NR'.sub.2, OR', CO.sub.2 R' or CONR'.sub.2 ; O--(C.sub.1 -C.sub.3)-alkyl; O--(C.sub.1 -C.sub.3)-alkyl substituted with NR'.sub.2, OR', CO.sub.2 R' or CONR'.sub.2 ; NR'.sub.2 ; OCF.sub.3 ; CF.sub.3 ; NO.sub.2 ; CO.sub.2 R'; CONR'; SR'; S(O.sub.2)N(R').sub.2 ; SCF.sub.3 ; or CN; and
- Q.sub.2 is optionally substituted with up to 4 substituents, independently selected from halo; C.sub.1 -C.sub.3 straight or branched alkyl; C.sub.1 -C.sub.3 straight or branched alkyl substituted with NR', NR'.sub.2, OR', CO.sub.2 R', or CONR'.sub.2 ; O--(C.sub.1 -C.sub.3)-alkyl; O--(C.sub.1 -C.sub.3)-alkyl substituted with NR', NR'.sub.2, OR', CO.sub.2 R' or CONR'.sub.2 ; NR'.sub.2 ; OCF.sub.3 ; CF.sub.3 ; NO.sub.2 ; CO.sub.2 R'; CONR'; R.sup.3 ; OR.sup.3 ; NR.sup.3 ; SR.sup.3 ; C(O)R.sup.3 ; C(O)N(R')R.sup.3 ; C(O)OR.sup.3 ; SR'; S(O.sub.2)N(R').sub.2 ; SCF.sub.3 ; or CN;
- wherein R' is selected from hydrogen, (C.sub.1 -C.sub.3)-alkyl or (C.sub.2 -C.sub.3)-alkenyl or alkynyl; and
- R.sup.3 is selected from phenyl, --(CH.sub.2).sub.m --W or W, wherein m is 1, 2, or 3;
- X is selected from --S--, --O--, --S(O.sub.2)--, --S(O)--, [--S(O.sub.2)--N(R)--, --N(R)--S(O.sub.2)--, --N(R)--C(O)O--, --O--C(O)--N(R),] --C(O)--, [--C(O)O--, --O--C(O)--, --C(O)--N(R)--, --N(R)--C(O)--,] --N(R)--, or --C(R).sub.2 --;
- each R is independently selected from hydrogen or (C1-C3)-alkyl;
- W is selected from --N(R').sub.2, --OR', --SR', --S(O)R', --S(O.sub.2)R', C(O)OR', ##STR56## or any 5 to 6-membered saturated or unsaturated heterocyclic ring having up to 2 nitrogen heteroatoms, wherein said heterocyclic ring is optionally substituted with up to 3 substituents independently selected from halo, C.sub.1 -C.sub.3 alkyl, O--(C.sub.1 -C.sub.3)-alkyl, NR'.sub.2, OCF.sub.3, CF.sub.3, NO.sub.2, CO.sub.2 R', CONR' or CN;
- Y is C;
- A is CR';
- n is 1; and
- R.sub.1 is selected from hydrogen, (C.sub.1 -C.sub.3)-alkyl, OH, or O--(C.sub.1 -C.sub.3)-alkyl.
- 3. The compound according to claim 1 or 2, wherein Q.sub.1 is selected from phenyl or pyridyl having 1 to 3 substituents independently selected from chloro, fluoro, bromo, --CH.sub.3, --OCH.sub.3, --OH, --CF.sub.3, --OCF.sub.3, --O(CH.sub.2).sub.2 CH.sub.3 or NH.sub.2, and wherein at least one of said substituents is in the ortho position and said substituents are independently selected from chloro, fluoro, bromo, --CH.sub.3, --OCH.sub.3, --OH, --CF.sub.3, --OCF.sub.3, --O(CH.sub.2).sub.2 CH.sub.3 or NH.sub.2.
- 4. The compound according to claim 3, wherein Q.sub.1 contains at least two substituents, both of which are in the ortho position.
- 5. The compound according to claim 3, wherein Q.sub.1 is selected from: ##STR57##
- 6. The compound according to claim 5, wherein Q.sub.1 is selected from 2-fluoro-6-trifluoromethylphenyl, 2,6-difluorophenyl or 2,6-dichlorophenyl.
- 7. The compound according to any one of claims 1 to 2, wherein Q.sub.2 is selected from phenyl or pyridyl and wherein Q.sub.2 optionally having up to 3 substituents, each of which is independently selected from chloro, fluoro, bromo, methyl, ethyl, isopropyl, --OCH.sub.3, --OH, --NH.sub.2, --CF.sub.3, --OCF.sub.3, --SCH.sub.3, --OCH.sub.3, --C(O)OH, C(O)OCH.sub.3, C(O)NH.sub.2 and --CH.sub.2 CH.sub.2 OH.
- 8. The compound according to claim 1, wherein Q2 is selected from:

The compound according to claim 8, wherein Q.sub.2 is selected from phenyl, [2-isopropylphenyl,] 3,4-dimethylphenyl, [2-ethylphenyl, 3-fluorophenyl, 2-methylphenyl, 3-chloro-4-fluorophenyl,] 3-chlorophenyl, [2-carbomethoxylphenyl,] 2-carboxyphenyl, [2-methyl-4-chlorophenyl, 2-bromophenyl, 2-pyridyl, 2-methylenehydroxyphenyl] or 4-fluorophenyl.
- 10. The compound according to claim 1 or 2, wherein X is selected from --S--, --O--, --S(O.sub.2)--, --S(O)--, --NR--, --C(R.sub.2)-- or --C(O)--.
- 11. The compound according to claim 9, wherein X is S.
- 12. The compound according to claim 1, wherein each R attached to Y is independently selected from hydrogen or methyl.
- 13. The compound according to claim 2, wherein Q.sub.1 is 2,6-dichlorophenyl.
- 14. The compound according to claim 2, wherein Q.sub.2 is selected from 2-carbomethoxyphenyl, 2-methylphenyl, 4-fluorophenyl, 2-carboxyphenyl, 2-carboxamidophenyl, 2-methyl-4-chlorophenyl, 2-bromophenyl, 2-methylenehydroxyphenyl or 2-pyridyl.
- 15. The compound of formula (Ic) according to claim 2, wherein:
- R and R.sub.1 are hydrogen;
- X is S;
- n is 1
- Q.sub.1 is 2,6-dichlorophenyl; and
- Q.sub.2 is phenyl.
- 16. The compound according to claim 2, wherein Q.sub.1 is selected from: ##STR58##
- 17. The compound according to claim 2, wherein, Q.sub.2 is selected from: unsubstituted 2-pyridyl or unsubstituted phenyl.
- 18. A pharmaceutical composition comprising an amount of a compound according to any one of claim 1 or 2 effective to inhibit p38, and a pharmaceutcially acceptable carrier.
