Claims
- 1. A method for determining the presence or amount of an analyte in a test sample, comprising the steps of:
- a) providing a test sample suspected of containing an analyte;
- b) forming a reaction mixture by contacting the test sample with a soluble capture reagent comprising a first binding member conjugated to a carboxymethylamylose polyanion and an indicator reagent comprising a labeled second binding member, wherein said first and second binding members specifically bind the analyte;
- c) incubating the reaction mixture under conditions sufficient for the analyte and reagents to react and form soluble capture reagent/analyte/indicator reagent complexes;
- d) after incubating, determining the presence or amount of analyte by
- (i) contacting the reaction mixture with a solid phase comprising a polymeric cation substance and having a pre-determined charge opposite to said carboxymethylamylose polyanion so that capture reagent/analyte/indicator reagent complexes and unreacted capture reagent are separated from the reaction mixture and coupled to the solid phase by the ionic attraction of the oppositely charged carboxymethylamylose polyanion and solid phase; and
- (ii) detecting label associated with the solid phase or with the unreacted indicator reagent in the reaction mixture.
- 2. The method according to claim 1, further comprising the addition of a non-specific binding blocker to said reaction mixture.
- 3. The method according to claim 2, wherein said non-specific binding blocker is a separate reagent or is contained within a member selected from the group consisting of said indicator reagent or said capture reagent.
- 4. The method according to claim 2, wherein said non-specific binding blocker is an unbound polymeric anion material.
- 5. The method according to claim 4, wherein said unbound polymeric anion material is selected from the group consisting of dextran sulfate, heparin, carboxymethyl dextran, carboxymethyl cellulose, pentosan polysulfate, inositol hexasulfate and .beta.-cyclodextrin sulfate.
- 6. The method according to claim 1, wherein said polymeric cation substance has a nitrogen content of at least about two percent.
- 7. The method according to claim 1, wherein the indicator reagent comprises an unlabeled second binding member which specifically binds the analyte and a labeled third binding member which specifically binds the second binding member.
- 8. A method for determining the presence or amount of an analyte in a test sample, comprising the steps of:
- a) providing a test sample suspected of containing an analyte;
- b) forming a reaction mixture by contacting the test sample with a soluble capture reagent comprising a carboxymethylamylose polyanion conjugated to a first specific binding member which specifically binds the analyte;
- c) incubating the reaction mixture under conditions sufficient for the analyte and capture reagent to react and form soluble capture reagent/analyte complexes;
- d) after incubating, determining the presence or amount of analyte by
- (i) contacting the reaction mixture with a solid phase comprising a polymeric cation substance and having a pre-determined charge opposite to said carboxymethylamylose polyanion so that capture reagent/analyte complexes and unreacted capture reagent are separated from the reaction mixture and coupled to the solid phase by the ionic attraction of the oppositely charged carboxymethylamylose polyanion and solid phase;
- (ii) contacting the solid phase with an indicator reagent comprising a detectable label conjugated to a second specific binding member which specifically binds the analyte;
- (iii) incubating the solid phase and the indicator reagent under conditions sufficient for the analyte and reagents to react and form capture reagent/analyte/indicator reagent complexes; and
- (iv) detecting label associated with the solid phase or with the unreacted indicator reagent.
- 9. The method according to claim 8, further comprising the step of washing said solid phase prior to contacting said solid phase with said indicator reagent.
- 10. The method according to claim 8, further comprising the addition of a non-specific binding blocker to said reaction mixture.
- 11. A method for determining the presence or amount of an analyte in a test sample, comprising the steps of:
- a) providing a test sample suspected of containing an analyte;
- b) forming a reaction mixture by contacting the test sample with a soluble capture reagent comprising a carboxymethylamylose polyanion conjugated to a binding member which specifically binds the analyte and an indicator reagent comprising an analyte analog conjugated to a detectable label;
- c) incubating the reaction mixture under conditions sufficient for the analyte and reagents to react and form soluble capture reagent/analyte or capture reagent/indicator reagent complexes;
- d) after incubating, determining the presence or amount of analyte by
- (i) contacting the reaction mixture with a solid phase comprising a polymeric cation substance and having a pre-determined charge opposite to said carboxymethylamylose polyanion so that capture reagent/analyte or capture reagent/indicator reagent complexes and unreacted capture reagent are separated from the reaction mixture and coupled to the solid phase by the ionic attraction of the oppositely charged carboxymethylamylose polyanion and solid phase; and
- (ii) detecting label associated with the solid phase or with the unreacted indicator reagent in the reaction mixture.
