Claims
- 1. A process of synthesizing a copolymer, said process comprising the steps of:
- reacting chitosan with a weak acid to produce a lower molecular weight polysaccharide;
- reacting between 1 and 50 percent of the free amines of said lower molecular weight polysaccharide with a first acylating agent, said first acylating agent selected from the group consisting of C.sub.4 -C.sub.34 polycarboxyalkane, C.sub.4 -C.sub.34 polycarboxyalkene, C.sub.8 -C.sub.40 polycarboxyarylalkane, C.sub.10 -C.sub.40 polycarboxyarylalkene, or an acylating derivative thereof; and
- reacting between 50 and 100 percent of the free amine of said lower molecular weight polysaccharide with a second acylating agent, said second acylating agent selected from the group consisting of C.sub.2-31 monocarboxyalkane, C.sub.3-31 monocarboxyalkene, C.sub.7-38 monocarboxyarylalkane, C.sub.9-35 monocarboxyarylalkene, or an acylating derivative thereof.
- 2. A process of claim 1, wherein the percentage of said free amines which are acylated is determined by the use of a free amine detecting agent.
- 3. A process of claim 1, wherein said first acylating agent is succinic anhydride, said second acylating agent is acetic anhydride, and said weak acid is nitrous acid.
- 4. A process of synthesizing a composition, said process comprising the steps of:
- reacting chitosan with a weak acid to produce a lower molecular weight polysaccharide;
- reacting between 1 and 50 percent of the free amines of said lower molecular weight polysaccharide with a first acylating agent, said first acylating agent selected from the group consisting of C.sub.4 -C.sub.34 polycarboxyalkane, C.sub.4 -C.sub.34 polycarboxyalkene, C.sub.8 -C.sub.40 polycarboxyarylalkane, C.sub.10 -C.sub.40 polycarboxyarylalkene, or an acylating derivative thereof;
- reacting between 50 and 100 percent of the free amine of said lower molecular weight polysaccharide with a second acylating agent, said second acylating agent selected from the group consisting of COOH, C.sub.2-31 monocarboxyalkane, C.sub.3-31 monocarboxyalkene, C.sub.7-38 monocarboxyarylalkane, C.sub.9-35 monocarboxyarylalkene, or an acylating derivative thereof;
- neutralizing said acylated lower molecular weight polysaccharide with a base; and
- mixing said neutralized lower acylated molecular weight polysaccharide with a polypeptide salt, wherein said polypeptide salt comprises at least one ionogenic amine, to form a polypeptide-copolymer ionic conjugate.
- 5. A process of claim 4, wherein the percentage of said free amines which are acylated is determined by the use of a free amine detecting agent.
- 6. A process of claim 4, wherein said first acylating agent is succinic anhydride, said second acylating agent is acetic anhydride, and said weak acid is nitrous acid.
- 7. A process according to claim 6, wherein the polypeptide salt is selected from the group consisting of growth hormone releasing peptide, luteinizing hormone releasing hormone, somatostatin, bombesin, gastrin releasing peptide, calcitonin, bradyknin, galanin, melanocyte stimulating hormone, growth hormone releasing factor, growth hormone, amylin, tachykinins, secretin, parathyroid hormone, enkaphelin, endothelin, calcitonin gene releasing peptide, neuromedins, parathyroid hormone related protein, glucagon, neurotensin, adrenocorticothropic hormone, peptide YY, glucagon releasing peptide, vasoactive intestinal peptide, pituitary adenylate cyclase activating peptide, motilin, substance P, neuropeptide Y and TSH, or an analog thereof.
- 8. A process according claim 7, wherein the polypeptide salt is selected from the group consisting of somatostatin, bombesin, gastrin releasing peptide, luteinizing hormone releasing hormone, growth hormone, growth hormone releasing factor, growth hormone releasing peptide, calcitonin, amylin, parathyroid hormone and parathyroid hormone related protein, or an analog thereof.
- 9. A process according to claim 8, wherein the polypeptide salt is somatostatin or an analog thereof.
- 10. A process according to claim 9, wherein the somatostatin analog is (D-Nal-c�Cys-Tyr-D-Trp-Lys-Val-Cys!-Thr-NH.sub.2.
