Claims
- 1. A compound having the formula: ##STR8## or a pharmaceutically acceptable salt, ester, amide or prodrug thereof wherein the ester is selected from C.sub.1 -to-C.sub.6 alkyl esters, C.sub.5 -to-C.sub.7 cycloalkyl esters and aryl-C.sub.1 -to-C.sub.12 -alkyl esters and wherein the amide results from reaction of a carboxylic acid moiety in the compound of formula I with NH.sub.3, NH.sub.2 (C.sub.1 -to-C.sub.6 alkyl), NH(C.sub.1 -to-C.sub.6 alkyl).sub.2 or a 5- or 6-membered heterocycle containing one nitrogen atom and wherein the prodrug is selected from the group consisting of (a) acyloxymethyl esters of carboxylic acids, (b) (5-methyl-2-oxo-1,3-dioxolen-4-yl)methyl esters of carboxylic acids, (c) esters derived from alcohol groups in the parent drug by reaction with succinic acid, phosphoric acid, dialkylaminoacetic acid or an amino acid (d) N-Mannich bases of amides or amines, (e) N-hydroxymethyl derivatives of amides, (f) N-acyloxyalkyl derivatives of amides or heterocyclic amines, (g) oxazolidinones derived from ketone groups in the parent drug by reaction with 2-aminoethanol, N-methyl-2-aminoethanol, N-(2-hydroxyethyl)-2-aminoethanol, 2-aminopropanol, 2-amino-2-methylpropanol, 2-amino-2-hydroxymethylpropanol, 3-amino-2-hydroxypropane, 2-amino-1-phenylethanol, 3-aminopropanol or N-methyl-2-amino-1-phenylpropanol and (h) enol esters derived from ketone groups in the parent drug,
- wherein n is zero or one;
- R and R' are chosen such that one of R and R' is hydrogen, and the other is selected from the group consisting of methyl, ethyl, 2-hydroxyethyl, propyl, cyclopropylmethyl, 2-oxopropyl, 2-ethanal and allyl;
- R.sup.1 is hydrogen, and R.sup.2 and R.sup.3 are independently selected from the group consisting of:
- (a) --OC(S)OR.sup.10, with the proviso that one of R.sup.20 and R.sup.21 is simultaneously --OC(S)OR.sup.12 wherein R.sup.10, R.sup.20, R.sup.21 and R.sup.12 are as defined herein;
- (b) --OC(O)R.sup.14 ; with the proviso that one of R.sup.20 and R.sup.21 is simultaneously --OC(O)R.sup.15 wherein R.sup.14, R.sup.20, R.sup.21 and R.sup.15 are as defined herein,
- (c) --C(=NH)CCl.sub.3 ;
- (d) --OC(O)CH.sub.2 l;
- (e) --OP(O)(OR.sup.10).sub.2 wherein R.sup.10 is as defined herein; and
- (f) C.sub.1 -to-C.sub.8 -loweralkoxy, with the proviso that R.sup.2 and R.sup.3 may not both be C.sub.1 -to-C.sub.8 -loweralkoxy;
- or, taken together, R.sup.1 and R.sup.2 are =NNHC(O)NH.sub.2 ;
- R.sup.4 and R.sup.5 are selected from the group consisting of:
- (a) hydrogen;
- (b) --OR.sup.11 wherein R.sup.11 is as defined herein;
- (c) halogen; and
- (d) hydroxy, with the proviso that when one of R.sup.4 and R.sup.5 is hydroxy, the other of R.sup.4 and R.sup.5 is hydrogen;
- or, taken together, R.sup.4 and R.sup.5 form a divalent radical selected from the group consisting of oxo and thiooxo;
- Y is a group selected from the subformulae: ##STR9## wherein R.sup.18 and R.sup.19 are chosen such that: (i) both of R.sup.18 and R.sup.19 are hydrogen, or
- (ii) one of R.sup.18 and R.sup.19, taken together with one of R.sup.20 and R.sup.21, forms a C-23/C-24 bond, and the other of R.sup.18 and R.sup.19 is hydrogen;
- R.sup.20 and R.sup.21 are chosen such that:
- (i) one of R.sup.20 and R.sup.21 is hydrogen, and the other of R.sup.20 and R.sup.21 is selected from the group consisting of:
- (a) hydrogen,
- (b) hydroxy,
- (c) amino,
- (d) --N(CH.sub.2 CH.sub.2).sub.2 N--R.sup.12 wherein R.sup.12 is as defined herein,
- (e) --N(CH.sub.2 CH.sub.2).sub.2 NH,
- (f) --OR.sup.12 wherein R.sup.12 is as defined herein,
- (g) --OC(O)NHR.sup.24, wherein R.sup.24 is selected from the group consisting of:
- (1) hydrogen,
- (2) C.sub.1 -to-C.sub.8 -loweralkyl, and
- (3) aryl,
- (h) --C(O)R.sup.12 wherein R.sup.12 is as defined herein,
- (i) --OC(S)OR.sup.12 wherein R.sup.12 is as defined herein,
- (j) --OC(O)R.sup.15 wherein R.sup.15 is as defined herein,
- (k) --OC(O)CH.sub.2 l, and
- (l) --O--[(hydroxy protecting group)]protected by a hydroxy-protecting group selected from methylthiomethyl tert-butyldimethylsilyl, tert-butyldiphenylsilyl, aryl-C(O)-- and C.sub.1 -to-C.sub.12 -alkyl-C(O)-- and; or
- (ii) one of R.sup.20 and R.sup.21, taken together with one of R.sup.18 and R.sup.19, forms a C-23/C-24 bond, and the other of R.sup.20 and R.sup.21 is selected from the group consisting of:
- (a) hydrogen,
- (b) hydroxy, and
- (c) C.sub.1 -to-C.sub.8 -loweralkoxy;
- or, taken together, R.sup.20 and R.sup.21 form an oxo group;
- R.sup.22 and R.sup.23 are chosen such that:
- (i) both are C.sub.1 -to-C.sub.8 -loweralkoxy; or
- (ii) one of R.sup.22 and R.sup.23 is hydrogen, and the other is selected from the group consisting of:
- (a) hydroxy,
- (b) --OC(O)R.sup.13 wherein r.sup.13 is as defined herein, and
- (c) --OC(O)O-(aryl-C.sub.1 -to-C.sub.12 -alkyl--);
- or, taken together, R.sup.22 and R.sup.23 form an oxo group;
- R.sup.6 and R.sup.7 are chosen such that:
- (i) both of R.sup.6 and R.sup.7 are hydrogen;
- (ii) one of R.sup.6 and R.sup.7 is hydrogen, and the other of R.sup.6 and R.sup.7, taken together with R.sup.8, forms a C-9/C-10 bond;
- (iii) one of R.sup.6 and R.sup.7 is hydroxy, and the other of R.sup.6 and R.sup.7 is selected from the group consisting of:
- (a) hydrogen,
- (b) C.sub.1 -to-C.sub.8 -loweralkyl,
- (c) aryl-C.sub.1 -to-C.sub.12 -alkyl, and
- (d) aryl;
- or, taken together, R.sup.6 and R.sup.7 form a divalent radical selected from the group consisting of:
- (a) oxo,
- (b) =NOH,
- (c) =NNHC(O)NH.sub.2,
- (d) =NNR.sup.25 R.sup.26, where R.sup.25 and R.sup.26 are independently selected from the group consisting of:
- (1) hydrogen,
- (2) C.sub.1 -to-C.sub.8 -loweralkyl, and
- (3) aryl,
- (e) =CH.sub.2,
- (f) --O--CH.sub.2 --, and
- (g) thiooxo;
- or, taken together with the carbon to which they are attached, R.sup.6 and R.sup.7 are absent such that C-8 is attached directly to C-10;
- R.sup.8 is selected from the group consisting of:
- (a) hydroxy,
- (b) halogen,
- (c) amino,
- (d) C.sub.1 -to-C.sub.8 -loweralkylamino,
- (e) aryl-C.sub.1 -to-C.sub.12 -alkylamino,
- (f) C.sub.1 -to-C.sub.8 -loweralkoxy, and
- (g) arylalkoxy;
- or, taken together with one of R.sup.6 and R.sup.7 as defined above, forms a C-9/C-10 double bond;
- or, taken together with R.sup.9 as defined herein forms a C-10/C-11 double bond;
- R.sup.9 is hydrogen or, taken together with R.sup.8 as defined above, forms a C-10/C-11 bond;
- R.sup.10, R.sup.11, R.sup.12 and R.sup.13 are each independently selected from the group consisting of C.sub.1 -to-C.sub.8 -loweralkyl, aryl-C.sub.1 -to-C.sub.12 -alkyl and aryl; and
- R.sup.14 and R.sup.15 are each independently selected from the group consisting of:
- (i) 2-nitro-aryl,
- (ii) 3-nitro-aryl,
- (iii) 4-nitro-aryl,
- (iv) 2-pyridyl,
- (v) 3-pyridyl, and
- (vi) 4-pyridyl
- wherein at each occurrence the aryl group is selected from phenyl, 1-naphthyl, 2-naphthyl, fluorenyl, (1,2,)-dihydronaphthyl, (1,2,3,4)-tetrahydronaphthyl, indenyl and indanyl substituted by R.sup.111, R.sup.112 and R.sup.113 wherein R.sup.111, R.sup.112 and R.sup.113 are independently selected from the group consisting of:
- (a) hydrogen,
- (b) --C.sub.1 -to-C.sub.7 -alkyl,
- (c) --C.sub.2 -to-C.sub.6 -alkenyl,
- (d) halogen,
- (e) --(CH.sub.2).sub.m N(C.sub.1 -to-C.sub.3 -alkyl).sub.2, where m is zero to six,

- at each occurrence the heterocycle or heterocyclic group is independently selected from the group consisting of pyrrolyl, pyrrolidinyl, pyrazolyl, pyrazolinyl, pyrazolidinyl, cytosinyl, thiocytosinyl, imidazolyl, imidazolinyl, imidazolidinyl, pyridyl, piperidinyl, pyrazinyl, piperazinyl, pyrimidinyl, pyridazinyl, xanthenyl, xanthonyl, xanthopterinyl, oxazoyl, oxazolidinyl, thiouracilyl, isoxazolyl, isoxazolidinyl, morpholinyl, indolyl, quinolinyl, uracilyl, urazolyl, uricyl, thiazolyl, thiazolidinyl, isothiazolyl, isothiazolidinyl, isoquinolinyl, thymidinyl, benzimidazolyl, benzothiazolyl, benzoxazolyl, furyl, thienyl and benzothienyl wherein any carbon or heteroatom with suitable valence may bear a substituent selected at each occurrence from R.sup.111, R.sup.112 and R.sup.113 independently as defined above.
- 2. A compound according to claim 1 wherein R and R' are selected from the group consisting of methyl, ethyl, propyl and allyl.
- 3. A compound according to claim 1 wherein R.sup.4 and R.sup.5 are both hydrogen.
- 4. A compound according to claim 1 wherein one of R.sup.4 and R.sup.5 is hydroxy, and the other of R.sup.4 and R.sup.5 is hydrogen.
- 5. A compound according to claim 1 wherein R.sup.20 is hydroxy.
- 6. A compound according to claim 1 wherein R.sup.20 and R.sup.21 are both hydrogen.
- 7. A compound according to claim 1 wherein R.sup.22 and R.sup.23, taken together, form an oxo group.
- 8. A compound according to claim 1 wherein R.sup.6 and R.sup.7, taken together, form an oxo group.
- 9. A compound according to claim 1 wherein R.sup.8 is hydroxy.
- 10. A compound according to claim 1 wherein R.sup.9 is hydrogen.
- 11. A compound according to claim 1, selected from the group consisting of:
