Claims
- 1. [Twice Amended] An isolated polynucleotide which encodes a polypeptide with cellulose synthase activity comprising a member selected from the group consisting of:
a. a polynucleotide having at least 85% sequence identity over the entire reference sequence, as determined by the BLAST 2.0 algorithm under default parameters, to a polynucleotide encoding a polypeptide of SEQ ID NO: 10; b. a polynucleotide encoding a polypeptide of SEQ ID NO: 10; c. a polynucleotide amplified from a Zea mays nucleic acid library using primers which selectively hybridize, under stringent hybridization conditions, to loci within a polynucleotide of SEQ ID NO: 9; d. a polynucleotide which selectively hybridizes, under stringent hybridization conditions and a wash in 0.5X SSC at 60° C., to a polynucleotide of SEQ ID NO: 9; e. a polynucleotide of SEQ ID NO: 9; f. a polynucleotide which is fully complementary to a polynucleotide of (a), (b), or (e); and g. a polynucleotide comprising at least 30 contiguous nucleotides from a polynucleotide of (a), (b), (c), (d), (e) or (f).
- 2. A recombinant expression cassette, comprising a member of claim 1 operably linked, in sense or anti-sense orientation, to a promoter.
- 3. A host cell comprising the recombinant expression cassette of claim 2.
- 4. A transgenic plant comprising a recombinant expression cassette of claim 2.
- 5. The transgenic plant of claim 4, wherein the plant is a monocot.
- 6. The transgenic plant of claim 4, wherein the plant is selected from the group consisting of: maize, soybean, sunflower, sorghum, canola, wheat, alfalfa, cotton, rice, barley, and millet.
- 7. A transgenic seed from the transgenic plant of claim 4.
- 8. [Once amended] A method of modulating the level of cellulose synthase in a plant cell capable of plant regeneration, comprising:
a. transforming the plant cell with a recombinant expression cassette comprising a cellulose synthase polynucleotide of claim 1 operably linked to a promoter; b. culturing the transformed plant cell; and c. inducing expression of said polynucleotide , which results in production of an encoded protein, for a time sufficient to modulate the level of cellulose synthase in said transformed plant cell.
- 9. The method of claim 8, wherein a plant is regenerated from the transformed plant cell.
- 10. The method of claim 9, wherein the plant is selected from the group consisting of: maize, soybean, sunflower, sorghum, canola, wheat, alfalfa, cotton, rice, barley, and millet.
- 11. The method of claim 8, wherein the promoter is a tissue-preferred promoter.
- 12. The method of claim 8, wherein the level of cellulose synthase is increased.
- 13. The method of claim 8 wherein the cellulose synthase polynucleotide is amplified from a Zea mays nucleic acid library using primers which selectively hybridize, under stringent hybridization conditions, to loci within a polynucleotide comprising SEQ ID NO: 9.
- 14. [Twice Amended] The method of claim 8 wherein the cellulose synthase polynucleotide is SEQ ID NO: 9.

- 16. [Twice Amended] The method of claim 8, wherein the encoded protein comprises a member selected from the group consisting of:
a. a polypeptide of at least 30 contiguous amino acids from a polypeptide comprising SEQ ID NO: 10; b. a polypeptide selected from the group consisting of SEQ ID NO: 10; c. a polypeptide having at least 85% sequence identity to a polypeptide of SEQ ID NO: 10, wherein said sequence identity is determined using GAP under default parameters; and, d. at least one polypeptide encoded by a member of claim 1.
- 17. [Twice Amended] The transgenic plant of claim 4, wherein the recombinant expression cassette confers in the transgenic plant expression of a protein comprising a member selected from the group consisting of:
a. a polypeptide of at least 30 contiguous amino acids from a polypeptide of SEQ ID NO: 10; b. a polypeptide of SEQ ID NO: 10; c. a polypeptide having at least 85% sequence identity to a polypeptide of SEQ ID NO: 10, wherein said sequence identity is determined using GAP under default parameters; and, d. at least one polypeptide encoded by a member of claim 1.
