Claims
- 1. An isolated polynucleotide encoding a polypeptide having Lysophosphatidic Acid Acyltransferase (LPAAT) activity, comprising a nucleotide sequence selected from the group consisting of:
(a) the DNA sequence of FIG. 9, FIG. 10, or FIG. 11 and biologically active fragments thereof; and (b) a DNA sequence which encodes the polypeptide of FIG. 9, FIG. 10, or FIG. 11 and biologically active fragments thereof.
- 2. An isolated polypeptide having LPAAT activity, comprising the amino acid sequence of FIG. 9, FIG. 10, or FIG. 11 and biologically active fragments thereof.
- 3. A method for screening one or more compounds to determine whether said one or more compounds increases or decreases LPAAT activity, comprising:
(a) contacting the polypeptide of claim 2 with one or more substrates for said polypeptide and with said one or more compounds; and (b) measuring whether the LPAAT activity of said polypeptide is increased or decreased by said one or more compounds.
- 4. The method of claim 3, wherein said one or more compounds is selected from a combinatorial chemical library.
- 5. A method of expressing the polypeptide of claim 2, comprising:
(a) introducing into a cell a polynucleotide comprising a nucleotide sequence selected from the group consisting of:
(i) the DNA sequence of FIG. 9, FIG. 10, or FIG. 11 and biologically active fragments thereof; and (ii) a DNA sequence which encodes the polypeptide of FIG. 9, FIG. 10, or FIG. 11 and biologically active fragments thereof,
wherein said polynucleotide is operably linked to a promoter; and (b) maintaining or growing said cell under conditions that result in the expression of said polypeptide.
- 6. The isolated polynucleotide of claim 1, wherein said polynucleotide comprises the DNA sequence of FIG. 9 or a DNA sequence encoding the polypeptide of FIG. 9.
- 7. The isolated polynucleotide of claim 1, wherein said polynucleotide comprises the DNA sequence of FIG. 10 or a DNA sequence encoding the polypeptide of FIG. 10.
- 8. The isolated polynucleotide of claim 1, wherein said polynucleotide comprises the DNA sequence of FIG. 11 or a DNA sequence encoding the polypeptide of FIG. 11.
- 9. The isolated polypeptide of claim 2, wherein said polypeptide comprises the amino acid sequence of FIG. 9.
- 10. The isolated polypeptide of claim 2, wherein said polypeptide comprises the amino acid sequence of FIG. 10.
- 11. The isolated polypeptide of claim 2, wherein said polypeptide comprises the amino acid sequence of FIG. 11.
- 12. An isolated polynucleotide encoding a polypeptide having Lysophosphatidic Acid Acyltransferase (LPAAT) activity, comprising a DNA sequence capable of hybridizing under high stringency conditions to the complement of (a) or (b) of claim 1 and which encodes a polypeptide having LPAAT activity.
- 13. A method of expressing a polypeptide having LPAAT activity, comprising:
(a) introducing into a cell a polynucleotide comprising a DNA sequence capable of hybridizing under high stringency conditions to the complement of (a) or (b) of claim 1 and which encodes a polypeptide having LPAAT activity,
wherein said polynucleotide is operably linked to a promoter, and (b) maintaining or growing said cell under conditions that result in the expression of said polypeptide.
