Claims
- 1. An isolated polynucleotide comprising a nucleotide sequence selected from the group consisting of: SEQ ID NO: 1-266, 350-375, 404 and 406.
- 2. An isolated polynucleotide comprising a nucleotide sequence selected from the group consisting of:
(a) complements of the sequences recited in SEQ ID NOS: 1-266, 350-375, 404 and 406; (b) reverse complements of the sequences recited in SEQ ID NOS: 1-266, 350-375, 404 and 406; and (c) reverse sequences of the sequences recited in SEQ ID NOS: 1-266, 350-375, 404 and 406.
- 3. An isolated polynucleotide comprising a sequence selected from the group consisting of:
(a) sequences having at least 75% identity to a sequence of SEQ ID NO: 1-266, 350-375, 404 and 406; (b) sequences having at least 90% identity to a sequence of SEQ ID NO: 1-266, 350-375, 404 and 406; and (c) sequences having at least 95% identity to a sequence of SEQ ID NO: 1-266, 350-375, 404 and 406.
- 4. The isolated polynucleotide of claim 3, wherein the polynucleotide encodes a polypeptide having activity in a lignin biosynthetic pathway.
- 5. An isolated polynucleotide comprising a nucleotide sequence that hybridizes to a sequence of SEQ ID NO: 1-266, 350-375, 404 and 406 under stringent hybridization conditions.
- 6. An isolated polynucleotide comprising a sequence selected from the group consisting of:
(a) nucleotide sequences that are 200-mers of a sequence recited in SEQ ID NO: 1-266, 350-375, 404 and 406; (b) nucleotide sequences that are 100-mers of a sequence recited in SEQ ID NO: 1-266, 350-375, 404 and 406; (c) nucleotide sequences that are 40-mers of a sequence recited in SEQ ID NO: 1-266, 350-375, 404 and 406; and (d) nucleotide sequences that are 20-mers of a sequence recited in SEQ ID NO: 1-266, 350-375, 404 and 406.
- 7. An isolated oligonucleotide probe or primer comprising at least 10 contiguous residues complementary to 10 contiguous residues of a nucleotide sequence recited in SEQ ID NO: 1-266, 350-375, 404 and 406.
- 8. A kit comprising a plurality of oligonucleotide probes or primers of claim 7.
- 9. A genetic construct comprising a polynucleotide of any one of claims 1-3.
- 10. A transgenic cell comprising a construct according to claim 9.
- 11. A construct comprising, in the 5′-3′ direction:
(a) a gene promoter sequence; (b) a polynucleotide sequence comprising at least one of the following: (1) a polynucleotide sequence comprising a coding region of a polynucleotide of any one of claims 1-3; and (2) a polynucleotide sequence comprising a non-coding region of a polynucleotide of any one of claims 1-3; and (c) a gene termination sequence.
- 12. The construct of claim 11 wherein the polynucleotide is in a sense orientation.
- 13. The construct of claim 11 wherein the polynucleotide is in an antisense orientation.
- 14. The construct of claim 11, wherein the gene promoter sequence is functional in a plant host to provide for transcription in xylem.
- 15. A transgenic plant cell comprising a construct of claim 11.
- 16. A plant comprising a transgenic plant cell according to claim 15, or fruit or seeds or progeny thereof.
- 17. A method for modulating one or more of the lignin content, the lignin composition and the lignin structure of a plant, comprising stably incorporating into the genome of the plant a polynucleotide of any one of claims 1-3.
- 18. The method of claim 17, wherein the plant is selected from the group consisting of eucalyptus and pine species.
- 19. The method of claim 17, comprising stably incorporating into the genome of the plant a construct of claim 11.
- 20. A method for producing a plant having one or more of altered lignin content, altered lignin composition and altered lignin structure, comprising:
(a) transforming a plant cell with a construct of claim 11 to provide a transgenic cell; and (b) cultivating the transgenic cell under conditions conducive to regeneration and mature plant growth.
- 21. A method for modifying the activity of a polypeptide involved in a lignin biosynthetic pathway in a plant comprising stably incorporating into the genome of the plant a construct of claim 11.
- 22. An isolated polypeptide comprising an amino acid sequence selected from the group consisting of: sequences recited in SEQ ID NOS: 267-349, 376-401, 405 and 407.
- 23. An isolated polypeptide comprising an amino acid sequence selected from the group consisting of:
(a) sequences having at least 75% identity to a sequence of SEQ ID NOS: 267-349, 376-401, 405 and 407; (b) sequences having at least 90% identity to a sequence of SEQ ID NOS: 267-349, 376-401, 405 and 407; and (c) sequences having at least 95% identity to a sequence of SEQ ID NOS: 267-349, 376-401, 405 and 407, wherein the polypeptide is active in a lignin biosynthetic pathway.
- 24. An isolated polynucleotide that encodes a polypeptide of claim 22.
- 25. An isolated polypeptide encoded by a polynucleotide of any one of claims 1-3.
- 26. A method for modifying the activity of a polypeptide involved in a lignin biosynthetic pathway in a plant, comprising introducing into cells of the plant RNA corresponding to a polynucleotide of any one of claims 1-3, thereby inhibiting expression of a polypeptide encoded by the polynucleotide.
- 27. A method for modifying the activity of a polypeptide involved in a lignin biosynthetic pathway in a plant, comprising introducing into cells of the plant double stranded RNA corresponding to a polynucleotide of any one of claims 1-3, thereby inhibiting expression of a polypeptide encoded by the polynucleotide.
