Claims
- 1. A container assembly for storing a cell or biological sample, said assembly comprising:
a container having a side, an open top end, and an internal dimension to contain a volume of a reagent sufficient to treat a biological sample; a cap for coupling to and closing said open top end of said container; and a sample holder having a closure member and an internal cavity with a dimension for receiving a biological sample, said sample holder having a plurality of fluid openings into said cavity to enable free flow of said reagent into said cavity, said sample holder having a dimension to fit in said container below said closure and to immerse said cavity in said reagent.
- 2. The container assembly of claim 1, wherein said sample holder is removably coupled to said container.
- 3. The container assembly of claim 2, wherein said container has a bottom wall with a recess, said recess having a dimension to receive said sample holder by a friction fit or interference fit.
- 4. The container assembly of claim 3, wherein said sample holder has a base dimensioned to fit in said recess in said bottom wall of said container to removably couple said sample holder to said base.
- 5. The container assembly of claim 1, wherein said sample holder is tethered to said cap.
- 6. The container assembly of claim 1, wherein said sample holder is tethered to said container.
- 7. The container assembly of claim 1, wherein said closure member cooperates with said sample holder to retain said sample holder in a predetermined location in said container.
- 8. The container assembly of claim 1, wherein said sample holder includes a permeable material defining said fluid openings.
- 9. The container assembly of claim 1, wherein said sample holder has a density greater that a density of said reagent to sink said sample holder in said container.
- 10. The container assembly of claim 1, wherein said cap includes a member to displace a portion of said reagent in said container.
- 11. The container assembly of claim 1, wherein said sample holder comprises a bottom and a side, an open top end, and at least one stabilizing member extending from said side of said sample holder a distance to cooperate with said side of said container to position said sample holder in a selected location in said container.
- 12. The container assembly of claim 11, comprising a plurality of said stabilizing members extending in a substantially radial outward direction from said side of said sample holder.
- 13. The container assembly of claim 1, wherein said closure member has an inner surface and a post extending from said inner surface and being oriented to extend into said internal cavity of said sample holder.
- 14. The container assembly of claim 13, wherein said post is oriented to extend in an axial direction into said internal cavity toward a bottom end of said sample holder.
- 15. The container assembly of claim 13, wherein said post extends into said internal cavity of said sample holder to capture a biological sample between said post and a bottom end of said sample holder.
- 16. The container assembly of claim 13, wherein said post has an axial end and at least one projection from said axial end to capture a biological sample.
- 17. The container assembly of claim 1, wherein said closure member is pivotally coupled to said sample holder to pivot between an open and closed position.
- 18. The container assembly of claim 1, wherein said closure member includes a permeable material to enable the flow of said reagent into said sample holder.
- 19. The container assembly of claim 18, wherein said side and said bottom of said sample holder include a permeable material to enable said reagent to flow into said sample holder.
- 20. The container assembly of claim 1, wherein said sample holder includes a base for contacting said bottom of said container and spacing said internal cavity of said sample holder from a bottom of said container.
- 21. The container assembly of claim 1, wherein said container has an internal volume and said sample holder has an internal volume, and wherein the ratio of said internal volume of said container and said internal volume of said sample holder is at least 10:1.
- 22. The container assembly of claim 1, wherein said closure member of said sample holder includes a top face and a handle member extending from said top face.
- 23. The container assembly of claim 22, wherein said handle member has a height to cooperate with said cap of said container.
- 24. The container assembly of claim 1, wherein said sample holder includes a plurality of stabilizing members extending outward from said side of said sample holder, said stabilizing members having a dimension to cooperate with said side of said container to position said sample holder in said container.
- 25. The container assembly of claim 11, wherein said sample holder includes a plurality of legs extending downwardly for supporting said sample holder.
- 26. The container assembly of claim 25, wherein said stabilizing member comprises a ring-shaped member extending radially outward from said side of said sample holder, said stabilizing member having a shape and dimension complementing said container.
- 27. The container assembly of claim 11, wherein said sample holder includes at least one tab extending upwardly and having a dimension for manipulating said sample holder.
- 28. The container assembly of claim 11, wherein said cap includes a plunger member having a dimension complementing an inner dimension of said internal cavity of said sample holder, said plunger member defining said closure member for said sample holder.
- 29. The container assembly of claim 28, wherein said plunger member has an axial dimension to contact and stabilize said sample holder in said container and to close said open top end of said sample holder.
- 30. The container assembly of claim 29, wherein said plunger member has a dimension to extend into said cavity of said sample holder to displace a portion of said reagent.
- 31. The container assembly of claim 1, wherein said sample holder is fixed to said container.
- 32. The container assembly of claim 1, wherein said sample holder is integrally molded and fixed to said container.
- 33. The container assembly of claim 32, wherein said sample holder includes a bottom, a side, an open top end, and wherein said closure member is positioned for closing said open to end of said sample holder.
- 34. The container assembly of claim 33, wherein said closure member is removably engageable with said sample holder.
- 35. The container assembly of claim 33, wherein said closure member is fixedly engageable with said open top end of said sample holder, said closure member providing selective accessibility to said internal cavity.
- 36. A container assembly, comprising:
a container having a bottom, a side and an open top end, and being dimensioned to contain a reagent; a closure cap removably coupled to said container and closing said open top end, said cap having an outer face and an inner face; a sample holder having an internal cavity for receiving a biological sample, said sample holder having at least one fluid opening into said internal cavity and having a dimension to fit within said container to completely immerse said internal cavity in said reagent and to limit linear movement of said sample holder in said container and to maintain said internal cavity immersed in said reagent; and a body coupled to said inner face of said closure to displace a portion of said reagent in said container.
- 37. The container assembly of claim 36, wherein said body is integrally formed with said cap and extends from said inner face in a substantially axial direction.
- 38. The container assembly of claim 37, wherein said cavity of said sample holder has an open top end, and wherein said body has an axial length and a width to close said open top end of said sample holder.
- 39. The container assembly of claim 37, wherein said sample holder includes a plurality of supporting legs extending from said bottom for engaging said bottom of said container.
- 40. The container assembly of claim 39, wherein said sample holder includes a stabilizing member extending from a side of said sample holder, said stabilizing member having a dimension to cooperate with said side of said container and to position said cavity in a predetermined location in said container.
- 41. A method of treating a biological sample with a liquid reagent comprising the steps of:
immersing a biological sample in a liquid reagent contained in a container, said liquid reagent being provided in an amount effective to treat said biological sample; and retaining said biological sample within a predetermined area of said container to maintain said biological sample completely immersed in said liquid reagent without regard to an orientation of said container.
- 42. The method of claim 41, comprising providing a ratio of at least 5:1 by volume of said liquid reagent to said biological sample.
- 43. The method of claim 41, comprising providing a ratio of at least 10:1 by volume of said liquid reagent to said biological sample.
- 44. The method of claim 41, comprising displacing a portion of said liquid reagent in said container to raise a level of said liquid reagent to maintain said biological sample immersed in said liquid reagent.
- 45. The method of claim 41, comprising providing a sample holder having a predetermined size to limit the size of said biological sample, and positioning said holder in said container.
- 46. The method of claim 41, wherein said holder has an internal cavity to receive said biological sample and to orient said biological sample in said predetermined area of said container.
- 47. The method of claim 41, comprising maintaining a level of said liquid reagent in said container to provide a headspace above said level sufficiently small to inhibit contact of said biological sample with air in said container.
- 48. The method of claim 41, wherein said container has an air head space above a level of said liquid reagent, said method comprising displacing said air to reduce said head space in said container.
- 49. The method of claim 41, wherein said container includes a cap having a body portion, and wherein said method comprises coupling said cap to said container to displace a portion of air above said liquid reagent.
- 50. The method of claim 49, wherein said cap retains said biological sample completely immersed in said liquid reagent.
- 51. The method of claim 41, wherein said biological sample is selected form the group consisting of organ specimen, tumor specimen, bone specimen, and connective tissue specimen.
- 52. The method of claim 41, wherein said biological sample is a tissue specimen.
- 53. The method of claim 41, wherein said liquid reagent is selected from the group consisting of nucleic acid stabilizing reagents, lysing reagents, tissue preserving reagents and drying reagents.
- 54. The method of claim 41, wherein said liquid reagent includes a cationic detergent.
- 55. The method of claim 41, wherein liquid reagent is selected from the group consisting of aqueous solutions and alcohol solutions.
- 56. A method of treating a biological sample comprising the steps of:
immersing a biological sample in treating reagent contained in container; and displacing a portion of said treating reagent in said container and raising a level of said treating reagent in said container to maintain said biological sample completely immersed in said treating reagent.
- 57. The method of claim 56, comprising the step of providing a ratio of at least 5:1 by volume of said treating reagent to said biological sample.
- 58. The method of claim 56, comprising providing a ratio of at least 10:1 by volume of said treating reagent to said biological sample.
- 59. The method of claim 56, comprising:
placing said biological sample in a sample holder, said sample holder having an internal cavity for receiving said biological sample and having a fluid opening into said cavity; positioning said sample holder in said container and completely immersing said internal cavity in said treating reagent; placing a closure member on said container and closing said container, said closure member cooperating with said sample holder to limit movement of said sample holder within said container and to retain said internal cavity immersed in said treating reagent.
- 60. The method of claim 56, wherein said closure member includes an inner face having a convex body portion extending in a substantially axial direction and having a dimension to displace a volume of said treating liquid in said container, said method comprising coupling said closure member to said container to displace a portion of said treating liquid and to raise the fluid level of said treating liquid above said sample holder.
