Claims
- 1. A method of controlling the rate of vapor diffusion between a crystal growth solution and a reservoir solution comprising
placing a device between the crystal growth solution and the reservoir solution, the device having defined therein discrete diffusion pathways, wherein said pathways control the vapor diffusion rate between the crystal growth solution and the reservoir solution.
- 2. A method for crystallizing a biological molecule comprising the steps of:
(a) placing a reservoir solution in the bottom of a container; (b) placing a first end of a device comprising a discrete diffusion pathway in contact with the top of the container; (c) placing a crystal growth solution comprising the biological molecule at an opposite end of the device from the reservoir solution; and (d) sealing the container, the device, and the solutions such that the diffusion pathway controls vapor diffusion kinetics between the reservoir solution and the crystal growth solution.
- 3. The method of claim 2 wherein the step of placing the crystal growth solution comprises placing the crystal growth solution in a well on the opposite end of the device.
- 4. The method of claim 2 wherein the step of placing the crystal growth solution comprises placing the crystal growth solution on a coverslip, wherein the crystal growth solution is hanging over the second end of the device.
- 5. The method of claim 2 further comprising the step of monitoring crystal growth.
- 6. The method of claim 5 further comprising the step of actively controlling the pathway in response to the monitoring step.
- 7. The method of claim 2 wherein the pathway comprises a material porous to vapor from the solutions.
- 8. The method of claim 2 wherein the pathway comprises a first channel between the crystal growth solution and the reservoir solution.
- 9. The method of claim 8 wherein the device comprises a second channel in vapor contact with the crystal growth solution.
- 10. The method of claim 9 wherein the container further comprises a second reservoir solution and wherein each of the channels are between the crystal growth solution and its respective the reservoir solution.
- 11. The method of claim 10 further comprising the step of actively controlling the first and second channels.
- 12. The method of claim 11 further comprising the step of monitoring crystal growth, wherein the first and second channels are actively controlled in response to information obtained from the monitoring step.
- 13. A method of controlling the rate of vapor diffusion between a crystal growth solution and a reservoir solution comprising the steps of selecting a device having defined therein a discrete diffusion pathway, wherein said pathway is configured to control the vapor diffusion rate between the crystal growth solution and the reservoir solution, and placing the device between the crystal growth solution and the reservoir solution.
- 14. The method of claim 13 further comprises the step of sealing the device such that vapor contact between the crystal growth solution and the reservoir solution is only through the pathway.
- 15. The method of claim 13 wherein the pathway has a predetermined geometry and controls the vapor diffusion rate by the geometry of the pathway.
- 16. The method of claim 15 further comprising the step selecting a second device having a second predefined geometry and placing the second device between a second sample of the crystal growth solution and a second sample of the reservoir solution, to achieve a second rate of vapor diffusion.
