Claims
- 1. A method of measuring cyclic-adenosine monophosphate (CAMP) levels in cells without disrupting the cells, said method comprising:
a) providing cells growing in culture, b) removing the growth media from the cells, c) adding cAMP collection medium to said cells, d) incubating said cells with said collection medium, e) optionally concentrating the collection medium; and f) determining the cAMP concentration in the collection medium.
- 2. The method of claim 1 wherein the cells are selected from the group consisting of mammalian, avian and insect cells.
- 3. The method of claim 1 further comprising concentrating the collection medium prior to determining the cAMP concentration.
- 4. The method of claim 1, wherein the cells are mammalian cells; wherein the collection medium comprises a buffered saline solution and imidazolidin.
- 5. A method of measuring cyclic-adenosine monophosphate (cAMP) levels in cells, without disrupting the cells, wherein the cells are selected from the group consisting of Madin-Darby canine kidney cells, mouse C2C12 myoblasts, and human fetal mesangial cells, said method comprising:
a) providing cells growing in culture; b) removing the growth media from the cells; c) adding cAMP collection medium to said cells, wherein the collection medium comprises phosphate-buffered saline (pH 7.5) or Tris-buffered saline (pH 7.5); d) removing the collection medium; e) concentrating the collection medium by lyophilization; f) determining the cAMP concentration in the collection medium by liquid scintillation.
- 6. A method of producing cyclic-adenosine monophosphate (cAMP) from cultured cells, said method comprising;
a) providing a culture of cells, b) removing the growth media from the cells, c) adding a cAMP collection medium, d) incubating said cells with said collection medium, e) removing the collection medium, and f) purifying the cAMP from the collection medium.
- 7. The method of claim 6 wherein the cells are selected from the group consisting of mammalian, avian and insect cells.
- 8. The method of claim 6 further comprising concentrating the collection medium prior to purifying the cAMP.
- 9. The method of claim 6, wherein the cells are mammalian cells; wherein the collection medium comprises a buffered saline solution, isoproteronol and imidazolidin.
- 10. The method of claim 9, wherein the cells are selected from the group consisting of Madin-Darby canine kidney cells, mouse C2C12 myoblasts, and human fetal mesangial cells; wherein the collection medium comprises phosphate-buffered saline (pH 7.5); wherein the collection medium is concentrated by lyophilization prior to determining the cAMP concentration, and wherein the cAMP concentration is determined by liquid scintillation.
- 11. A diagnostic kit for measuring extracellular cyclic-Adenosine monophosphate (cAMP) according to any of the methods of claims 1, 2, 3, 4, or 5.
