Claims
- 1. A process for generating a double-stranded polynucleotide containing a sequence complementary to a sequence of interest and a single-stranded polynucleotide containing a sequence complementary to said sequence of interest, said process comprising:obtaining a single-stranded nucleic acid comprising said sequence of interest; and contacting said single-stranded nucleic acid with a first primer and a second primer, wherein said first primer hybridizes to a first sequence of said single-stranded nucleic acid and said second primer hybridizes to a second sequence of said single-stranded nucleic acid, said second sequence overlapping with said first sequence, and said first and second sequences being located 3′ to said sequence of interest, and wherein said contacting occurs in the presence of a polymerase activity allowing extension of a primer with displacement, under conditions allowing hybridization and extension of both primers, whereby an extension product of one of the primers displaces an extension product of the other primer, thus obtaining a double-stranded polynucleotide containing a sequence complementary to said sequence of interest and a displaced single-stranded polynucleotide containing a sequence complementary to said sequence of interest.
- 2. The process of claim 1, wherein one of said first and second primers is present in molar excess relative to the other said primer.
- 3. The process of claim 1, wherein said single-stranded nucleic acid is DNA.
- 4. The process of claim 1, wherein said single-stranded nucleic acid is RNA.
