Claims
- 1. A method for obtaining a biologically active recombinant protein by reconstituting the protein from a denatured state to its active form, wherein a feed solution containing the recombinant protein of interest in its denatured form and/or in biologically inactive intermediate forms is subject to a chromatographic separation process, in which said protein is reconstituted under conditions that promote refolding of the protein and the intermediate forms are separated from the refolded protein, characterized in that the denatured form and/or the inactive intermediate forms of the protein are separated from the refolded protein in a continuous or quasi-continuous chromatographic method.
- 2. The method of claim 1, wherein the intermediate forms that have been separated from the refolded protein are reintroduced into the feed solution and thus undergo at least one more reconstitution process.
- 3. The method of claim 2, wherein the separation process is selected from ion-exchange chromatography, affinity chromatography, hydrophobic interaction chromatography, reversed phase chromatography, covalent chromatography, size exclusion chromatography and adsorption chromatography.
- 4. The method of claim 3, wherein the separation process is operated continuously in the form of annular chromatography.
- 5. The method of claim 3, wherein the separation process is operated quasi-continuously in the form of a simulated moving bed chromatography.
- 6. The method of claim 3, wherein the separation process is operated quasi-continuously in the form of a carrousel chromatography.
