Claims
- 1. A negative selection method for enriching and recovering desired cells in a sample containing the desired cells, erythrocytes and undesired cells comprising:
(1) contacting the sample with an antibody composition comprising (a) at least one antibody that binds to an antigen on the undesired cells linked, either directly or indirectly, to (b) at least one antibody that binds to the erythrocytes, under conditions to allow immunorosettes of the undesired cells and the erythrocytes to form; and (2) separating the immunorosettes from the sample to obtain a sample enriched in the desired cells.
- 2. A method according to claim 1 further comprising adding an aggregating agent in step (1).
- 3. A method according to claim 2 wherein the aggregating agent is dextran.
- 4. A method according to claim 2 wherein the aggregating agent is hetastarch.
- 5. A method according to claim 1 wherein antibody (b) comprises anti-glycophorin A antibody.
- 6. A method according to claim 1 wherein the immunorosettes are separated in step (2) in a specialized container.
- 7. A method according to claim 6 wherein the specialized container contains an insert which forms a barrier between sedimented immunorosettes and the rest of the sample.
- 8. A method according to claim 7 wherein the insert is a gel, plug, screen or funnel.
- 9. A method according to claim 6 wherein the specialized container is an evacuated blood collection tube containing anti-coagulant, buoyant density medium and a thixotropic gel.
- 10. A method according to claim 2 wherein the antibody (b) that binds to the erythrocyte is anti-glycophorin A.
- 11. A method according to claim 2 wherein the antibody composition comprises tetrameric antibody complexes comprising (a) an antibody that binds to the antigen on the undesired cells; and (b) an antibody that binds to the erythrocytes; and (c) two antibodies that bind to the Fc fragment of the antibodies defined in (a) and (b), wherein the antibodies in (a) and (b) are of the same animal species and the antibodies in (c) are of a different animal species from the antibodies in (a) and (b).
- 12. A method according to claim 6 wherein the antibody composition comprises tetrameric antibody complexes comprising (a) an antibody that binds to the antigen on the undesired cells; and (b) an antibody that binds to the erythrocytes; and (c) two antibodies that bind to the Fc fragment of the antibodies defined in (a) and (b), wherein the antibodies in (a) and (b) are of the same animal species and the antibodies in (c) are of a different animal species from the antibodies in (a) and (b).
- 13. An antibody composition for separating nucleated cells in a sample comprising (a) at least one antibody that binds to an antigen on the nucleated cells linked, either directly or indirectly to (b) at least one antibody that binds to erythrocytes.
- 14. An antibody composition according to claim 13, further comprising an aggregating agent.
- 15. An antibody composition according to claim 14 wherein the aggregating agent comprises dextran.
- 16. An antibody composition according to claim 14 wherein the aggregating agent comprises hetastarch.
- 17. An antibody composition according to claim 14 wherein antibody (b) comprises anti-glycophorin A antibody.
- 18. An antibody composition according to claim 14 wherein the antibody composition comprises tetrameric antibody complexes comprising (a) an antibody that binds to the antigen on the undesired cells; and (b) an antibody that binds to the erythrocytes; and (c) two antibodies that bind to the Fc fragment of the antibodies defined in (a) and (b), wherein the antibodies in (a) and (b) are of the same animal species and the antibodies in (c) are of a different animal species from the antibodies in (a) and (b).
- 19. A kit for separating nucleated cells from a sample comprising:
(1) an antibody composition comprising (a) at least one antibody that binds to an antigen on the nucleated cells to be separated, and (b) at least one antibody that binds to erythrocytes.
- 20. A kit according to claim 19 further comprising an aggregating agent.
- 21. A kit according to claim 20 wherein the aggregating agent is dextran or hetastarch.
- 22. A kit according to claim 19 further comprising a specialized tube.
- 23. A kit according to claim 22 wherein the specialized tube contains an insert which forms a barrier between sedimented immunorosettes and the rest of the sample.
- 24. A kit according to claim 23 wherein the insert is a gel, plug, screen or funnel.
- 25. A kit according to claim 24 wherein the specialized tube is an evacuated blood collection tube containing anti-coagulant, buoyant density medium and a thixotropic gel.
