Claims
- 1. A method of treating endotoxic shock in a mammal which comprises administering to said mammal an effective amount of a compound having the formula ##STR6## or a pharmaceutically acceptable base addition salt thereof, wherein R.sub.1 is hydrogen or R'OOC--;
- Z is --(CH.sub.2).sub.n -- or phenylene;
- n is 1-8; ##STR7## R.sub.2 is hydroxy, halo, or --O--(CH.sub.2).sub.m --Y; R.sub.3 is C.sub.1 -C.sub.6 alkyl, C.sub.1 -C.sub.6 alkanoyl, C.sub.2 -C.sub.4 alkenyl, C.sub.1 -C.sub.4 alkoxy, hydroxy-substituted C.sub.1 -C.sub.3 alkyl, or --CH.sub.2 --D;
- A is a bond or straight or branched chain C1-C10 alkylidene;
- R.sub.4 is C.sub.1 -C.sub.6 alkyl, C.sub.2 -C.sub.6 alkenyl, C.sub.2 -C.sub.6 alkynyl, 1,2,4-triazol-l-yl, hydroxy, --CN, halo, --N.sub.3, --NR.sub.5 R.sub.6, --COR.sub.7, --S(O).sub.p --(C.sub.1 -C.sub.4 alkyl), 5-tetrazolyl optionally substituted with a C.sub.1 -C.sub.4 alkyl group or --(CH.sub.2).sub.q --COOR', or phenyl optionally substituted with one or two groups selected from halo, cyano, C.sub.1 -C.sub.3 alkyl, trifluoromethyl, --CH.sub.2 CH, --CH.sub.2 Br, C.sub.1 -C.sub.4 alkoxy, --S(O).sub.p -- (C.sub.1 -C.sub.4 alkyl), acetenyl, --COOR', 5-tetrazolyl, or 5-tetrazolyl substituted with C.sub.1 -C.sub.4 alkyl or --(CH.sub.2).sub.q --COOR';
- where each R' is independently hydrogen or C.sub.1 -C.sub.4 alkyl;
- m is 1-4;
- q is 1-4
- Y is hydrogen or --CN;
- D is halo, C.sub.1 -C.sub.4 alkoxy, or --S--(C.sub.1 -C.sub.4 alkyl);
- R.sub.5 and R.sub.6 are independently hydrogen, C.sub.1 -C.sub.3 alkyl, or C.sub.2 -C.sub.4 alkanoyl, or when taken together with the nitrogen atom to which they are attached from a morpholino ring;
- R.sub.7 is hydroxy, C.sub.1 -C.sub.4 alkoxy, halo, --NR.sub.5 R.sub.6, --NHOH, --N(CH.sub.3)OH, ##STR8## or C.sub.1 -C.sub.3 alkyl; and each p is 0, 1, or 2,
- provided that when A is a bond, R.sub.4 must be C.sub.1 -C.sub.6 alkyl or an optionally substituted phenyl group, and further provided that when one of R.sub.5 and R.sub.6 is C.sub.2 -C.sub.4 alkanoyl, the other of R.sub.5 and R.sub.6 is hydrogen.
- 2. The method of claim 1 wherein said compound has the formula ##STR9## and pharmaceutically acceptable base addition salts thereof wherein R.sub.1 ' is methyl or ethyl;
- R.sub.2 ' is hydrogen or methyl;
- R.sub.3 ' is ethyl, propyl, --CH.sub.2 --S--CH.sub.3, or allyl; and
- R.sub.4 ' is hydroxy, cyano, --COOR', --CONR.sub.5 R.sub.6, --CO(C.sub.1 -C.sub.3 alkyl), --S(O).sub.p --(C.sub.1 -C.sub.4 alkyl), 5-tetrazolyl optionally substituted with C.sub.1 -C.sub.4 alkyl group, or phenyl optionally substituted in the meta position with one of the groups listed above, particularly cyano.
- 3. The method of claim 1 employing 5-ethyl-2-hydroxy-4-[6-methyl-6-(1H-tetrazol-5-yl)heptyloxy]-acetophenone or a parmaceutically acceptable base addition salt thereof.
- 4. The method of claim 1 employing 3-[(4-Acetyl-5-hydroxy-2-allylphenoxy)-methyl]-benzonitrile or a pharmaceutically acceptable base addition salt thereof.
- 5. The method of claim 1 employing 3-.differential.(4-acetyl-2-ethyl-5-hydroxyphenoxy)methyl]benzonitrile or a pharmaceutically acceptable base addition salt thereof.
