Claims
- 1. A method for the in vivo reduction of nitric oxide levels in a subject, said method comprising:
- administering to said subject an effective amount of at least one dithiocarbamate-containing nitric oxide scavenger, wherein said dithiocarbamate-containing nitric oxide scavenger comprises a dithiocarbamate moiety and, optionally a physiologically compatible di- or tri-valent transition metal ion, wherein said dithiocarbamate has the structure:
- R.sub.1 R.sub.2 N--C(S)--S
- wherein:
- each of R.sub.1 and R.sub.2 is independently selected from a C.sub.1 up to C.sub.18 alkyl substituted alkyl, cycloalkyl, substituted cycloalkyl, heterocyclic, substituted heterocyclic, alkenyl, substituted alkenyl, alkynyl, substituted alkynyl aryl, substituted aryl, heteroaryl, substituted heteroaryl, alkylaryl, substituted alkylaryl, arylalkyl, substituted arylalkyl or R.sub.1 and R.sub.2 can cooperate to form 5-, 6- or 7-membered ring including N, R.sub.1 and R.sub.2.
- 2. A method for treating nitric oxide overproduction in a subject, said method comprising:
- administering to said subject an effective amount of at least one dithiocarbamate-containing nitric oxide scavenger, wherein said dithiocarbamate-containing nitric oxide scavenger comprises a dithiocarbamate moiety and, optionally, a physiologically compatible di- or tri-valent transition metal ion, wherein said dithiocarbamate has the structure:
- R.sub.1 R.sub.2 N--C(S)--S
- wherein:
- each of R.sub.1 and R.sub.2 is independently selected from a C.sub.1 up to C.sub.18 alkyl, substituted alkyl, cycloalkyl, substituted cycloalkyl, heterocyclic, substituted heterocyclic, alkenyl, substituted alkenyl, alkynyl, substituted alkynyl, aryl, substituted aryl, heteroaryl, substituted heteroaryl, alkylaryl, substituted alkylaryl, arylalkyl, substituted arylalkyl, or R.sub.1 and R.sub.2 can cooperate to form a 5-, 6- or 7-membered ring including N, R.sub.1 and R.sub.2.
- 3. A method according to claim 2 wherein said nitric oxide overproduction is associated with septic shock, ischemia, administration of cytokines, overexpression of cytokines, ulcer, ulcerative colitis, diabetes, arthritis, asthma, Alzheimer's disease, Parkinson's disease, multiple sclerosis, cirrhosis, allograft rejection, encephalomyelitis, meningitis, pancreatitis, peritonitis, vasculitis, lymphocytic choriomeningitis, glomerulonephritis, uveitis, ileitis, liver inflammation, renal inflammation, hemorrhagic shock, anaphylactic shock, burn, Crohn's disease, infection, hemodialysis, chronic fatigue syndrome, stroke, cancer, cardiopulmonary bypass or ischemic/reperfusion injury.
- 4. A method according to claim 2 wherein said nitric oxide overproduction is associated with chronic fatigue syndrome.
- 5. A method according to claim 2 wherein said nitric oxide overproduction is associated with stroke.
- 6. A method according to claim 2 wherein said nitric oxide overproduction is associated with cancer.
- 7. A method according to claim 2 wherein said nitric oxide overproduction is associated with hemodialysis, cardiopulmonary bypass or ischemic/reperfusion injury.
- 8. A method according to claim 2 wherein the ratio of transition metal ion to dithiocarbamate moiety falls in the range of zero up to about 1:2.
- 9. A method according to claim 2 wherein said physiologically compatible di- or tri-valent transition metal is selected from iron, cobalt, copper or manganese.
- 10. A method according to claim 2 wherein said dithiocarbamate-containing nitric oxide scavenger is administered in combination with a cytokine, an antibiotic, a vasoactive agent, or mixtures thereof.
- 11. A method according to claim 10 wherein said cytokine is selected from interleukin-1 (IL-1), interleukin-2 (IL2), interleukin-6 (IL-6), interleukin-12 (IL-12) tumor necrosis factor (TNF) or interferon-.gamma..
- 12. A method according to claim 10 wherein said vasoactive agent is selected from a catecholamine, noradrenaline, dopamine or dobutamine.
- 13. A method according to claim 2 wherein said dithiocarbamate-containing nitric oxide scavenger is delivered orally, intravenously, subcutaneously, parenterally, rectally or by inhalation.
- 14. A method according to claim 2 wherein said dithiocarbamate-containing nitric oxide-scavenger is delivered in the form of a solid, solution, emulsion, dispersion, micelle or liposome.
- 15. A composition comprising a compound having the structure I and a pharmaceutically acceptable carrier therefor, wherein said pharmaceutically acceptable carrier is a sterile injectable solution or suspension, an organic or inorganic carrier suitable for enteral or parenteral administration, a non-toxic carrier for tablets, troches, lozenges, pellets, capsules, suppositories, emulsions, aqueous or oily suspension, dispersible powders or granules, liposomes, hard or soft capsules, syrups or elixirs, and wherein structure I is:
- R.sub.1 R.sub.2 N--C(S)--S.sup.- M.sup.+ (I)
- wherein:
- each of R.sub.1 and R.sub.2 is independently selected from a C.sub.1 up to C.sub.18 alkyl, substituted alkyl, cycloalkyl, substituted cycloalkyl, heterocyclic, substituted heterocyclic, alkenyl, substituted alkenyl, alkynyl, substituted alkynyl, aryl, substituted aryl, heteroaryl, substituted heteroaryl, alkylaryl, substituted alkylaryl arylalkyl, substituted arylalkyl or R.sub.1 and R.sub.2 can cooperate to form a 5, 6- or 7-membered ring including N, R.sub.1 and R.sub.2, and
- M is optionally present, and when present is a monovalent cation,
- provided, however, that the following compounds are excluded from the definition of Formula I, i.e., when R.sub.1 is ethyl, R.sub.2 is not ethyl; or when R.sub.1 is CH.sub.2 (CHOH).sub.4 CH.sub.2 OH, R.sub.2 is not methyl, isoamyl, benzyl, 4-methylbenzyl or p-isopropylbenzyl; or when R.sub.1 is CH.sub.2 CO.sub.2.sup.-, R.sub.2 is not CH.sub.2 CO.sub.2.sup.- ; or when R.sub.1 is CO.sub.2.sup.-, R.sub.2 is not CH.sub.3 ; or when R.sub.1 is CH.sub.2 CH.sub.2 --OH, R.sub.2 is not CH.sub.2 CH.sub.2 --OH; or when R.sub.1 and R.sub.2 combined, together with the carbamate nitrogen, form a pyrrolidinyl-2-carboxylate.
- 16. A composition according to claim 15 wherein M.sup.+ is selected from H.sup.+, Na.sup.+, NH.sub.4.sup.+ or tetraalkyl ammonium.
- 17. A composition according to claim 15 wherein said pharmaceutically acceptable carrier is selected from a solid, emulsion, dispersion, micelle or liposome.
- 18. A composition according to claim 15 wherein said composition further comprises an enteric coating.
- 19. A composition according to claim 15 wherein said pharmaceutically acceptable carrier is a sterile injectable suspension or solution.
- 20. A composition according to claim 15 wherein said pharmaceutically acceptable carrier is an organic or inorganic carrier suitable for enteral or parenteral administration.
- 21. A composition according to claim 15 wherein said pharmaceutically acceptable carrier is a non-toxic carrier for tablets, troches, lozenges, pellets, capsules, suppositories, emulsions, aqueous or oily suspensions, dispersible powders or granules, liposomes, hard or soft capsules, syrups or elixirs.
- 22. A composition comprising a compound having the structure II and a pharmaceutically acceptable carrier therefor, wherein said pharmaceutically acceptable carrier is a sterile injectable solution or suspension, an organic or inorganic carrier suitable for enteral or parenteral administration, a non-toxic carrier for tablets, troches, lozenges, pellets, capsules, suppositories, emulsions, aqueous or oily suspensions, dispersible powders or granules, liposomes, hard or soft capsules, syrups or elixirs, and wherein structure II is:
- �R.sub.1 R.sub.2 N--C(S)--S.sup.- !.sub.2 M.sup.+2,+3 (II)
- wherein:
- each of R.sub.1 and R.sub.2 is independently selected from a C.sub.1 up to C.sub.18 alkyl, substituted alkyl, cycloalkyl, substituted cycloalkyl, heterocyclic, substituted heterocyclic, alkenyl, substituted alkenyl, alkynyl, substituted alkynyl, aryl, substituted aryl, heteroaryl, substituted heteroaryl, alkylaryl, substituted alkylaryl, arylalkyl substituted arylalkyl, or R.sub.1 and R.sub.2 can cooperate to form a 5-, 6- or 7-membered ring including N, R.sub.1 and R.sub.2, and
- M is optionally present, and when present, is a physiologically compatible divalent or trivalent transition metal cation,
- provided, however, that the following compounds are excluded from the definition of Formula II, i.e,, when R.sub.1 is ethyl, R.sub.2 is not ethyl; or when R.sub.1 is CH.sub.2 (CHOH).sub.4 CH.sub.2 OH, R.sub.2 is not methyl, isoamyl, benzyl, 4-methylbenzyl or p-isopropylbenzyl; or when R.sub.1 is CH.sub.2 CO, R.sub.2 is not CH.sub.2 CO.sub.2 ; or when R.sub.1 is CO.sub.2.sup.-, R.sub.2 is not CH.sub.3 ; or when R.sub.1 is CH.sub.2 CH.sub.2 --OH, R.sub.2 is not CH.sub.2 CH.sub.2 --OH; or when R.sub.1 and R.sub.2 combined, together with the carbamate nitrogen, form a pyrrolidinyl-2-carboxylate.
- 23. A composition according to claim 22 wherein the ratio of transition metal ion to dithiocarbamate moiety falls in the range of zero up to about 1:2.
- 24. A composition according to claim 22 wherein M is selected from Fe.sup.+2, Fe.sup.+3, Co.sup.+2, Co.sup.+3, Cu.sup.+2, Mn.sup.+2 or Mn.sup.+3.
- 25. A composition according to claim 22 wherein:
- each of R.sub.1 and R.sub.2 =a C.sub.1 up to C.sub.12 alkyl, substituted alkyl, alkenyl, substituted alkenyl, alkynyl or substituted alkynyl, wherein said substituents are selected from carboxyl, --C(O)H, oxyacyl, phenol, phenoxy, pyridinyl, pyrrolidinyl, amino, amido, hydroxy, nitro or sulfuryl, and
- M=Fe.sup.+2 or Fe.sup.+3.
- 26. A composition according to claim 22 wherein:
- R.sub.1 =a C.sub.2 up to C.sub.8 alkyl or substituted alkyl, wherein said substituents are selected from carboxyl, acetyl, pyridnyl, pyrrolidinyl, amino, amido, hydroxy or nitro,
- R.sub.2 is selected from a C.sub.1 up to C.sub.6 alkyl or substituted alkyl, or R.sub.2 can cooperate with R.sub.1 to form a 5-, 6- or 7-membered ring including N, R.sub.2 and R.sub.1, and
- M=Fe.sup.+2.
- 27. A composition according to claim 22 wherein:
- R.sub.1 =a C.sub.2 up to C.sub.8 alkyl or substituted alkyl, wherein said substituents are selected from carboxyl, acetyl, amido or hydroxy,
- R.sub.2 =a C.sub.1 up to C.sub.4 alkyl or substituted alkyl, and
- M=Fe.sup.+2.
- 28. A composition according to claim 22 wherein said pharmaceutically acceptable carrier is selected from a solid, emulsion, dispersion, micelle or liposome.
- 29. A composition according to claim 22 wherein said composition further comprises an enteric coating.
- 30. A composition according to claim 22 wherein said pharmaceutically acceptable carrier is a sterile injectable solution or suspension.
- 31. A composition according to claim 22 wherein said pharmaceutically acceptable carrier is an organic or inorganic carrier suitable for enteral or parenteral administration.
- 32. A composition according to claim 22 wherein said pharmaceutically acceptable carrier is a non-toxic carrier for tablets, troches, lozenges, pellets, capsules, suppositories, emulsions, aqueous or oily suspensions, dispersible powders or granules, liposomes, hard or soft capsules, syrups or elixirs.
- 33. A composition comprising a compound having structure I in combination with a pharmacologically active agents wherein structure I is:
- R.sub.1 R.sub.2 N--C(S)--S.sup.- M.sup.+ (I)
- wherein:
- each of R.sub.1 and R.sub.2 is independently selected from a C.sub.1 up to C.sub.18 alkyl, substituted alkyl cycloalkyl substituted cycloalkyl, heterocyclic, substituted heterocyclic, alkenyl, substituted, alkenyl, alkynyl, substituted alkynyl, aryl, substituted aryl, heteroaryl, substituted heteroaryl, alkylaryl, substituted alkylaryl, arylalkyl, substituted arylalkyl or R.sub.1 and R.sub.2 can cooperate to form a 5-, 6- or 7-membered ring including N, R.sub.1 and R.sub.2, and
- M is optionally present and when present, is a monovalent cation,

- 35. A composition comprising a compound having structure II in combination with a pharmacologically active agent, wherein structure II is:

- wherein:
- each of R.sub.1 and R.sub.2 is independently selected from a C.sub.1 up to C.sub.18 alkyl, substituted alkyl, cycloalkyl, substituted cycloalkyl, heterocyclic, substituted heterocyclic, alkenyl, substituted alkenyl, alkynyl, substituted alkynyl, aryl, substituted aryl, heteroaryl, substituted heteroaryl, alkylaryl, substituted alkylaryl, arylalkyl substituted arylalkyl, or R.sub.1 and R.sub.2 can cooperate to form a 5-, 6- or 7-membered ring including N, R.sub.1 and R.sub.2, and

- 36. A composition according to claim 35 wherein said pharmacologically active agent is selected from a cytokine, an antibiotic, a vasoactive agent, or mixtures thereof.
- 37. A composition according to claim 35 wherein said pharmaceutically acceptable carrier is a sterile injectable solution or suspension.
- 38. A composition according to claim 35 wherein said pharmaceutically acceptable carrier is an organic or inorganic carrier suitable for enteral or parenteral administration.
- 39. A composition according to claim 35 wherein said pharmaceutically acceptable carrier is a non-toxic carrier for tablets, troches, lozenges, pellets, capsules, suppositories, emulsions, aqueous or oily suspensions, dispersible powders or granules, liposomes, hard or soft capsules, syrups or elixirs.
- 40. A composition according to claim 35 wherein said pharmaceutically acceptable carrier is selected from a solid, emulsion, dispersion, micelle or liposome.
- 41. A composition according to claim 35 wherein said composition further comprises an enteric coating.
