Claims
- 1. A method for producing a polypeptide, comprising:(a) cultivating a Bacillus cell in a medium conducive for the production of the polypeptide, wherein the Bacillus cell comprises a nucleic acid construct comprising a tandem promoter in which each promoter sequence of the tandem promoter is operably linked to a nucleic acid sequence encoding the polypeptide, wherein the tandem promoter comprises promoters selected from the group consisting of the amyL promoter, amyQ promoter, aprH promoter, cryIIIA promoter, subtilisin Carlsberg gene promoter, and consensus promoters of the amyL promoter, amyQ promoter, aprH promoter, cryIIIA promoter, and subtilisin Carlsberg gene promoter having the sequence TTGACA for the “−35” region and TATAAT for the “−10” region; and (b) isolating the polypeptide from the cultivation medium.
- 2. The method of claim 1, wherein the tandem promoter comprises at least two copies of a promoter selected from the group consisting of the amyL promoter, amyQ promoter, aprH promoter, cryIIIA promoter, subtilisin Carlsberg gene promoter, and consensus promoters of the amyL promoter, amyQ promoter, aprH promoter, cryIIIA promoter, and subtilisin Carlsberg gene promoter having the sequence TTGACA for the “−35” region and TATAAT for the “−10” region.
- 3. The method of claim 1, wherein the tandem promoter comprises the amyL promoter and the cryIIIA promoter.
- 4. The method of claim 1, wherein the Bacillus cell contains one or more copies of the nucleic acid construct.
- 5. The method of claim 1, wherein the nucleic acid construct further comprises a selectable marker gene.
- 6. The method of claim 1, wherein the Bacillus cell contains no selectable marker gene.
- 7. The method of claim 1, wherein the nucleic acid sequence encodes a polypeptide heterologous to the Bacillus cell.
- 8. The method of claim 1, wherein the polypeptide is a hormone or variant thereof, enzyme, receptor or portion thereof, antibody or portion thereof, or reporter.
- 9. The method of claim 8, wherein the enzyme is an oxidoreductase, transferase, hydrolase, lyase, isomerase, or ligase.
- 10. The method of claim 1, wherein the Bacillus host cell is a Bacillus alkalophilus, Bacillus amyloliquefaciens, Bacillus brevis, Bacillus circulans, Bacillus clausii, Bacillus coagulans, Bacillus firmus, Bacillus lautus, Bacillus lentus, Bacillus licheniformis, Bacillus megaterium, Bacillus pumilus, Bacillus stearothermophilus, Bacillus subtilis, or Bacillus thuringiensis cell.
- 11. A method for producing a polypeptide, comprising:(a) cultivating a Bacillus cell in a medium conducive for the production of the polypeptide, wherein the Bacillus cell comprises a nucleic acid construct comprising (i) a consensus promoter obtained from Bacillus amyloliquefaciens alpha-amylase gene (amy Q) or Bacillus subtilis alpha-amylase gene (amy E) having the sequence TTGACA for the “−35” region and TATAAT for the “−10” region operably linked to a single copy of a nucleic acid sequence encoding the polypeptide and (ii) an mRNA processing/stabilizing sequence located downstream of the consensus promoter and upstream of the nucleic acid sequence encoding the polypeptide; and (b) isolating the polypeptide from the cultivation medium.
- 12. The method of claim 11, wherein the mRNA processing/stabilizing sequence is the cryIIIA mRNA processing/stabilizing sequence.
- 13. The method of claim 11, wherein the mRNA processing/stabilizing sequence is the SP82 mRNA processing/stabilizing sequence.
- 14. The method of claim 11, wherein the mRNA processing/stabilizing sequence generates mRNA transcripts essentially of the same size.
- 15. The method of claim 11, wherein the Bacillus cell contains one or more copies of the nucleic acid construct.
- 16. The method of claim 11, wherein the nucleic acid construct further comprises a selectable marker gene.
- 17. The method of claim 11, wherein the Bacillus cell contains no selectable marker gene.
- 18. The method of claim 11, wherein the nucleic acid sequence encodes a polypeptide heterologous to the Bacillus cell.
- 19. The method of claim 11, wherein the polypeptide is a hormone or variant thereof, enzyme, receptor or portion thereof, antibody or portion thereof, or reporter.
- 20. The method of claim 19, wherein the enzyme is an oxidoreductase, transferase, hydrolase, lyase, isomerase, or ligase.
- 21. The method of claim 19, wherein the enzyme is an aminopeptidase, amylase, carbohydrase, carboxypeptidase, catalase, cellulase, chitinase, cutinase, cyclodextrin glycosyltransferase, deoxyribonuclease, esterase, alpha-galactosidase, beta-galactosidase, glucoamylase, alpha-glucosidase, beta-glucosidase, invertase, laccase, lipase, mannosidase, mutanase, oxidase, pectinolytic enzyme, peroxidase, phytase, polyphenoloxidase, proteolytic enzyme, ribonuclease, transglutaminase, or xylanase.
- 22. The method of claim 11, wherein the Bacillus host cell is a Bacillus alkalophilus, Bacillus amyloliquefaciens, Bacillus brevis, Bacillus circulans, Bacillus clausii, Bacillus coagulans, Bacillus firmus, Bacillus lautus, Bacillus lentus, Bacillus licheniformis, Bacillus megaterium, Bacillus pumilus, Bacillus stearothermophilus, Bacillus subtilis, or Bacillus thuringiensis cell.
- 23. The method of claim 11, wherein the Bacillus cell is a Bacillus subtilis cell.
