MICROBIOCIDAL PYRAZOLE DERIVATIVES

Abstract
Compounds of the formula (I) wherein the substituents are as defined in claim 1, are useful as a pesticides.
Description

The present invention relates to microbiocidal pyrazole derivatives, e.g. as active ingredients, which have microbiocidal activity, in particular fungicidal activity. The invention also relates to preparation of these pyrazole derivatives, to pyrazole derivatives used as intermediates in the preparation of these pyrazole derivatives, to preparation of these intermediates, to agrochemical compositions which comprise at least one of the pyrazole derivatives, to preparation of these compositions and to use of the pyrazole derivatives or compositions in agriculture or horticulture for controlling or preventing infestation of plants, harvested food crops, seeds or non-living materials by phytopathogenic microorganisms, preferably fungi.


Certain compounds for use as fungicides are described in WO 2007/014290, WO 2008/013622, WO 2008/013925, WO 2008/091580, WO 2008/091594 and WO 2009/055514.

For the avoidance of doubt, by the expression “providing that at least one of R3, R4, R5, R6, R7 or R8 is, C1-C4alkylsulfanyl, C3-C5cycloalkylsulfanyl, C1-C4haloalkysulfanyl, C1-C4alkylsulfinyl, C3-C5cycloalkylsulfinyl, C1-C4haloalkylsulfinyl, C1-C4alkylsulfonyl, C3-C5cycloalkylsulfonyl, C1-C4haloalkylsulfonyl, —S(O)q(NQ)C1-C4alkyl, —S(O)q(NQ)C3-C5cycloalkyl, —S(O)q(NQ)C1-C4haloalkyl, —SO2(NQ)C1-C4alkyl, —SO2(NQ)C1-C4cycloalkyl, —SO2(NQ)C1-C4haloalkyl, wherein q is 0 or 1 and Q is hydrogen, C1-C4alkyl or C1-C4alkylcarbonyl, it is intended herein that at least one of R3, R4, R5, R6, R7 or R8 is not hydrogen.


Where substituents are indicated as being optionally substituted, this means that they may or may not carry one or more identical or different substituents, e.g. one to five substituents, e.g. one to three substituents. Normally not more than three such optional substituents are present at the same time. Where a group is indicated as being substituted, e.g. alkyl, unless stated otherwise this includes those groups that are part of other groups, e.g. the alkyl in alkylthio.


The term “halogen” refers to fluorine, chlorine, bromine or iodine, preferably fluorine, chlorine or bromine.


Alkyl substituents may be straight-chained or branched. Alkyl on its own or as part of another substituent is, depending upon the number of carbon atoms mentioned, for example, methyl, ethyl, n-propyl, n-butyl, n-pentyl, n-hexyl and the isomers thereof, for example, iso-propyl, iso-butyl, sec-butyl, tert-butyl, iso-amyl or pivaloyl.


Alkenyl substituents can be in the form of straight or branched chains, and the alkenyl moieties, where appropriate, can be of either the (E)- or (Z)-configuration. Examples are vinyl and allyl. The alkenyl groups are preferably C2-C6, more preferably C2-C4 and most preferably C2-C3 alkenyl groups.


Alkynyl substituents can be in the form of straight or branched chains. Examples are ethynyl and propargyl. The alkynyl groups are preferably C2-C6, more preferably C2-C4 and most preferably C2-C3 alkynyl groups.


Haloalkyl groups may contain one or more identical or different halogen atoms and, for example, may stand for CH2Cl, CHCl2, CCl3, CH2F, CHF2, CF3, CF3CH2, CH3CF2, CF3CF2 or CCl3CCl2.


Haloalkenyl groups are alkenyl groups, respectively, which are substituted with one or more of the same or different halogen atoms and are, for example, 2,2-difluorovinyl or 1,2-dichloro-2-fluoro-vinyl.


Haloalkynyl groups are alkynyl groups, respectively, which are substituted with one or more of the same or different halogen atoms and are, for example, 1-chloro-prop-2-ynyl.


Alkoxy means a radical —OR, where R is alkyl, e.g. as defined above. Alkoxy groups include, but are not limited to, methoxy, ethoxy, 1-methylethoxy, propoxy, butoxy, 1-methylpropoxy and 2-methylpropoxy.


Cyano means a —CN group.


Amino means an NH2 group.


Hydroxyl or hydroxy stands for a —OH group.


Aryl means a ring system which may be mono-, bi- or tricyclic. Examples of such rings include phenyl, naphthalenyl, anthracenyl, indenyl or phenanthrenyl. A preferred aryl group is phenyl.


Heteroaryl stands for aromatic ring systems comprising mono-, bi- or tricyclic systems wherein at least one oxygen, nitrogen or sulfur atom is present as a ring member. Monocyclic and bicyclic aromatic ring systems are preferred, monocyclic ring systems are more preferred. For example, monocyclic heteoraryl may be a 5- to 7-membered aromatic ring containing one to three heteroatoms selected from oxygen, nitrogen and sulfur, more preferably selected from nitrogen and sulfur. Bicyclic heteroaryl may be a 9- to 11-membered bicyclic ring containing one to five heteroatoms, preferably one to three heteroatoms, selected from oxygen, nitrogen and sulfur. Examples are furyl, thienyl, pyrrolyl, imidazolyl, pyrazolyl, thiazolyl, isothiazolyl, oxazolyl, isoxazolyl, oxadiazolyl, thiadiazolyl, triazolyl, tetrazolyl, pyridyl, pyridazinyl, pyrimidinyl, pyrazinyl, triazinyl, tetrazinyl, indolyl, benzothiophenyl, benzofuranyl, benzimidazolyl, indazolyl, benzotriazolyl, benzothiazolyl, benzoxazolyl, imiazothiazoyl, quinolinyl, quinoxalinyl, isoquinolinyl, phthalazinyl, quinoxalinyl, quinazolinyl, cinnolinyl and naphthyridinyl, preferably pyridyl, pyrazinyl, pyridazinyl, pyrimidinyl, pyrrolyl, pyrazolyl, imidazolyl, triazolyl, furanyl, thienyl thiazolyl or thiadiazolyl. Heteroaryl rings do not contain adjacent oxygen ring atoms, adjacent sulfur ring atoms or adjacent oxygen and sulfur ring atoms. A link to a heteroaryl group can be via a carbon atom or via a nitrogen atom.


Heterocyclyl is defined to include heteroaryl and in addition their saturated or partially unsaturated analogues. The different rings of bi- or tricyclic heterocyclic ring systems may be linked via one atom belonging to two different rings (spiro), via two adjacent ring atoms belonging to two different rings (annelated) or via two different, not adjacent ring atoms belonging to two different rings (bridged).


The presence of one or more possible asymmetric carbon atoms in a compound of formula I means that the compounds may occur in optically isomeric forms, i.e. enantiomeric or diastereomeric forms. Also atropisomers may occur as a result of restricted rotation about a single bond. Formula I is intended to include all those possible isomeric forms and mixtures thereof. The present invention includes all those possible isomeric forms and mixtures thereof for a compound of formula I. Likewise, formula I is intended to include all possible tautomers. The present invention includes all possible tautomeric forms for a compound of formula I.


In each case, the compounds of formula I according to the invention are in free form, in oxidized form as a N-oxide or in salt form, e.g. an agronomically usable salt form.


N-oxides are oxidized forms of tertiary amines or oxidized forms of nitrogen containing heteroaromatic compounds. They are described for instance in the book “Heterocyclic N-oxides” by A. Albini and S. Pietra, CRC Press, Boca Raton 1991.


The following list provides definitions, including preferred definitions, for substituents G1, G2, T, Y1, Y2, n, p, R1, R2, R3, R4, R5, R6, R7, R8, R9, R10, R11, R12, R13, R14 and R15 with reference to compounds of formula I and other compounds of the invention carrying the same substituents. Preferably, in compounds according to formula I-a, R14 and R15 are independently C1-C4alkyl, C1-C4haloalkyl or halogen.

Surprisingly, it has now been found that the novel compounds of formula I have, for practical purposes, a very advantageous level of biological activity for protecting plants against diseases that are caused by fungi.


The compounds of formula I can be used in the agricultural sector and related fields of use e.g. as active ingredients for controlling plant pests or on non-living materials for control of spoilage microorganisms or organisms potentially harmful to man. The novel compounds are distinguished by excellent activity at low rates of application, by being well tolerated by plants and by being environmentally safe. They have very useful curative, preventive and systemic properties and may be used for protecting numerous cultivated plants. The compounds of formula I can be used to inhibit or destroy the pests that occur on plants or parts of plants (fruit, blossoms, leaves, stems, tubers, roots) of different crops of useful plants, while at the same time protecting also those parts of the plants that grow later e.g. from phytopathogenic microorganisms.


It is also possible to use compounds of formula I as dressing agents for the treatment of plant propagation material, e.g., seed, such as fruits, tubers or grains, or plant cuttings (for example rice), for the protection against fungal infections as well as against phytopathogenic fungi occurring in the soil. The propagation material can be treated with a composition comprising a compound of formula I before planting: seed, for example, can be dressed before being sown. The active ingredients according to the invention can also be applied to grains (coating), either by impregnating the seeds in a liquid formulation or by coating them with a solid formulation. The composition can also be applied to the planting site when the propagation material is being planted, for example, to the seed furrow during sowing. The invention relates also to such methods of treating plant propagation material and to the plant propagation material so treated.


Furthermore the compounds according to present invention can be used for controlling fungi in related areas, for example in the protection of technical materials, including wood and wood related technical products, in food storage, in hygiene management.


In addition, the invention could be used to protect non-living materials from fungal attack, e.g. lumber, wall boards and paint.


The compounds of formula I are, for example, effective against the phytopathogenic fungi of the following classes: Fungi imperfecti (e.g. Alternaria spp.), Basidiomycetes (e.g. Corticium spp., Ceratobasidium spp., Waitea spp., Thanatephorus spp., Rhizoctonia spp., Hemileia spp., Puccinia spp., Phakopsora spp., Ustilago spp., Tilletia spp.), Ascomycetes (e.g. Venturia spp., Blumeria spp., Erysiphe spp., Podosphaera spp., Uncinula spp., Monilinia spp., Sclerotinia spp., Colletotrichum spp., Glomerella spp., Fusarium spp., Gibberella spp., Monographella spp., Phaeosphaeria spp., Mycosphaerella spp., Cercospora spp., Pyrenophora spp., Rhynchosporium spp., Magnaporthe spp., Gaeumannomyces spp., Oculimacula spp., Ramularia spp., Botryotinia spp.) and Oomycetes (e.g. Phytophthora spp., Pythium spp., Plasmopara spp., Peronospora spp., Pseudoperonospora spp. Bremia spp). Outstanding activity is observed against downy mildew (e.g. Plasmopara viticola) and late blight (e.g. Phytophthora infestans). Furthermore, the novel compounds of formula I are effective against phytopathogenic gram negative and gram positive bacteria (e.g. Xanthomonas spp, Pseudomonas spp, Erwinia amylovora, Ralstonia spp.) and viruses (e.g. tobacco mosaic virus).


Within the scope of present invention, target crops and/or useful plants to be protected typically comprise the following species of plants: cereal (wheat, barley, rye, oat, rice, maize, sorghum and related species); beet (sugar beet and fodder beet); pomes, drupes and soft fruit (apples, pears, plums, peaches, almonds, cherries, strawberries, raspberries and blackberries); leguminous plants (beans, lentils, peas, soybeans); oil plants (rape, mustard, poppy, olives, sunflowers, coconut, castor oil plants, cocoa beans, groundnuts); cucumber plants (pumpkins, cucumbers, melons); fibre plants (cotton, flax, hemp, jute); citrus fruit (oranges, lemons, grapefruit, mandarins); vegetables (spinach, lettuce, asparagus, cabbages, carrots, onions, tomatoes, potatoes, paprika); lauraceae (avocado, cinnamomum, camphor) or plants such as tobacco, nuts, coffee, eggplants, sugar cane, tea, pepper, vines, hops, bananas and natural rubber plants, as well as turf and ornamentals.


The useful plants and/or target crops in accordance with the invention include conventional as well as genetically enhanced or engineered varieties such as, for example, insect resistant (e.g. Bt. and VIP varieties) as well as disease resistant, herbicide tolerant (e.g. glyphosate- and glufosinate-resistant maize varieties commercially available under the trade names RoundupReady® and LibertyLink®) and nematode tolerant varieties. By way of example, suitable genetically enhanced or engineered crop varieties include the Stoneville 5599BR cotton and Stoneville 4892BR cotton varieties.


The term “useful plants” and/or “target crops” is to be understood as including also useful plants that have been rendered tolerant to herbicides like bromoxynil or classes of herbicides (such as, for example, HPPD inhibitors, ALS inhibitors, for example primisulfuron, prosulfuron and trifloxysulfuron, EPSPS (5-enol-pyrovyl-shikimate-3-phosphate-synthase) inhibitors, GS (glutamine synthetase) inhibitors or PPO (protoporphyrinogen-oxidase) inhibitors) as a result of conventional methods of breeding or genetic engineering. An example of a crop that has been rendered tolerant to imidazolinones, e.g. imazamox, by conventional methods of breeding (mutagenesis) is Clearfield® summer rape (Canola). Examples of crops that have been rendered tolerant to herbicides or classes of herbicides by genetic engineering methods include glyphosate- and glufosinate-resistant maize varieties commercially available under the trade names RoundupReady®, Herculex I® and LibertyLink®.


The term “useful plants” and/or “target crops” is to be understood as including also useful plants which have been so transformed by the use of recombinant DNA techniques that they are capable of synthesising one or more selectively acting toxins, such as are known, for example, from toxin-producing bacteria, especially those of the genus Bacillus.


The term “useful plants” and/or “target crops” is to be understood as including also useful plants which have been so transformed by the use of recombinant DNA techniques that they are capable of synthesising antipathogenic substances having a selective action, such as, for example, the so-called “pathogenesis-related proteins” (PRPs, see e.g. EP-A-0 392 225). Examples of such antipathogenic substances and transgenic plants capable of synthesising such antipathogenic substances are known, for example, from EP-A-0 392 225, WO 95/33818, and EP-A-0 353 191. The methods of producing such transgenic plants are generally known to the person skilled in the art and are described, for example, in the publications mentioned above.


The term “locus” of a plant as used herein is intended to embrace the place on which the plants are growing, where the plant propagation materials of the plants are sown or where the plant propagation materials of the plants will be placed into the soil. An example for such a locus is a field, on which crop plants are growing.


The term “plant propagation material” is understood to denote generative parts of the plant, such as seeds, which can be used for the multiplication of the latter, and vegetative material, such as cuttings or tubers, for example potatoes. There may be mentioned for example seeds (in the strict sense), roots, fruits, tubers, bulbs, rhizomes and parts of plants. Germinated plants and young plants which are to be transplanted after germination or after emergence from the soil, may also be mentioned. These young plants may be protected before transplantation by a total or partial treatment by immersion. Preferably “plant propagation material” is understood to denote seeds.


The compounds of formula I may be used in unmodified form or, preferably, together with the adjuvants conventionally employed in the art of formulation. To this end they may be conveniently formulated in known manner to emulsifiable concentrates, coatable pastes, directly sprayable or dilutable solutions or suspensions, dilute emulsions, wettable powders, soluble powders, dusts, granulates, and also encapsulations e.g. in polymeric substances. As with the type of the compositions, the methods of application, such as spraying, atomising, dusting, scattering, coating or pouring, are chosen in accordance with the intended objectives and the prevailing circumstances. The compositions may also contain further adjuvants such as stabilizers, antifoams, viscosity regulators, binders or tackifiers as well as fertilizers, micronutrient donors or other formulations for obtaining special effects.


Suitable carriers and adjuvants, e.g. for agricultural use, can be solid or liquid and are substances useful in formulation technology, e.g. natural or regenerated mineral substances, solvents, dispersants, wetting agents, tackifiers, thickeners, binders or fertilizers. Such carriers are for example described in WO 97/33890.


The compounds of formula I are normally used in the form of compositions and can be applied to the crop area or plant to be treated, simultaneously or in succession with further compounds. These further compounds can be e.g. fertilizers or micronutrient donors or other preparations, which influence the growth of plants. They can also be selective herbicides or non-selective herbicides as well as insecticides, fungicides, bactericides, nematicides, molluscicides or mixtures of several of these preparations, if desired together with further carriers, surfactants or application promoting adjuvants customarily employed in the art of formulation.


The compounds of formula I may be used in the form of (fungicidal) compositions for controlling or protecting against phytopathogenic microorganisms, comprising as active ingredient at least one compound of formula I or of at least one preferred individual compound as above-defined, in free form or in agrochemically usable salt form, and at least one of the above-mentioned adjuvants.


The invention provides a composition, preferably a fungicidal composition, comprising at least one compound formula I an agriculturally acceptable carrier and optionally an adjuvant. An agricultural acceptable carrier is for example a carrier that is suitable for agricultural use. Agricultural carriers are well known in the art. Preferably said composition may comprise at least one or more pesticidally active compounds, for example an additional fungicidal active ingredient in addition to the compound of formula I.


The compound of formula (I) may be the sole active ingredient of a composition or it may be admixed with one or more additional active ingredients such as a pesticide, fungicide, synergist, herbicide or plant growth regulator where appropriate. An additional active ingredient may, in some cases, result in unexpected synergistic activities. Another aspect of invention is related to the use of a compound of formula I or of a preferred individual compound as above-defined, of a composition comprising at least one compound of formula I or at least one preferred individual compound as above-defined, or of a fungicidal mixture comprising at least one compound of formula I or at least one preferred individual compound as above-defined, in admixture with other fungicides, as described above, for controlling or preventing infestation of plants, e.g. useful plants such as crop plants, propagation material thereof, e.g. seeds, harvested crops, e.g. harvested food crops, or non-living materials by phytopathogenic microorganisms, preferably fungal organisms.


A further aspect of invention is related to a method of controlling or preventing an infestation of plants, e.g. useful plants such as crop plants, propagation material thereof, e.g. seeds, harvested crops, e.g. harvested food crops, or of non-living materials by phytopathogenic or spoilage microorganisms or organisms potentially harmful to man, especially fungal organisms, which comprises the application of a compound of formula I or of a preferred individual compound as above-defined as active ingredient to the plants, to parts of the plants or to the locus thereof, to the propagation material thereof, or to any part of the non-living materials.


Controlling or preventing means reducing infestation by phytopathogenic or spoilage microorganisms or organisms potentially harmful to man, especially fungal organisms, to such a level that an improvement is demonstrated.


A preferred method of controlling or preventing an infestation of crop plants by phytopathogenic microorganisms, especially fungal organisms, which comprises the application of a compound of formula I, or an agrochemical composition which contains at least one of said compounds, is foliar application. The frequency of application and the rate of application will depend on the risk of infestation by the corresponding pathogen. However, the compounds of formula I can also penetrate the plant through the roots via the soil (systemic action) by drenching the locus of the plant with a liquid formulation, or by applying the compounds in solid form to the soil, e.g. in granular form (soil application). In crops of water rice such granulates can be applied to the flooded rice field. The compounds of formula I may also be applied to seeds (coating) by impregnating the seeds or tubers either with a liquid formulation of the fungicide or coating them with a solid formulation.


A formulation, e.g. a composition containing the compound of formula I, and, if desired, a solid or liquid adjuvant or monomers for encapsulating the compound of formula I, may be prepared in a known manner, typically by intimately mixing and/or grinding the compound with extenders, for example solvents, solid carriers and, optionally, surface active compounds (surfactants).


The agrochemical formulations and/or compositions will usually contain from 0.1 to 99% by weight, preferably from 0.1 to 95% by weight, of the compound of formula 1, 99.9 to 1% by weight, preferably 99.8 to 5% by weight, of a solid or liquid adjuvant, and from 0 to 25% by weight, preferably from 0.1 to 25% by weight, of a surfactant.


Advantageous rates of application are normally from 5 g to 2 kg of active ingredient (a.i.) per hectare (ha), preferably from 10 g to 1 kg a.i./ha, most preferably from 20 g to 600 g a.i./ha. When used as seed drenching agent, convenient dosages are from 10 mg to 1 g of active substance per kg of seeds.


Whereas it is preferred to formulate commercial products as concentrates, the end user will normally use dilute formulations.


The following non-limiting example illustrates the above-described invention in more detail.







EXAMPLE 1
This example illustrates the preparation of 1-[4-[4-[5-(2,6-difluorophenyl)-4,5-dihydroisoxazol-3-yl]thiazol-2-yl]-3-methylsulfanyl-1-piperidyl]-2-[5-methyl-3-(trifluoromethyl)pyrazol-1-yl]ethanone (compound I.az.007)
a) Preparation of tert-butyl 4-(trifluoromethylsulfonyloxy)-3,6-dihydro-2H-pyridine-1-carboxylate

To a solution of N-isopropylpropan-2-amine (16.9 mL, 0.12 mol) in tetrahydrofuran (240 mL) added n-butyllithium (7.45 g, 0.12 mol) (2M Solution in tetrahydrofuran) at −10° C., stirred for 30 min. The resulted lithium di-isopropylamine solution was added drop wise to a solution of tert-butyl 4-oxopiperidine-1-carboxylate (20.0 g, 0.10 mol) in tetrahydrofuran (100 mL) at −78° C. After stirring for 2 h at the same temperature, a solution of 1,1,1-trifluoro-N-phenyl-N-(trifluoromethylsulfonyl)methanesulfonamide (39.4 g, 0.11 mol) in tetrahydrofuran (780 mL) was added to the reaction mixture and stirred for 5 h at −70° C. Reaction mixture was concentrated under reduced pressure. The obtained crude product was purified by combi flash (eluting with 3% ethylacetate:n-hexane) to get 11 g of tert-butyl 4-(trifluoromethylsulfonyloxy)-3,6-dihydro-2H-pyridine-1-carboxylate. MS: m/z=332 (M+1).


b) Preparation of tert-butyl 4-cyano-3,6-dihydro-2H-pyridine-1-carboxylate

To a solution of tert-butyl 4-(trifluoromethylsulfonyloxy)-3,6-dihydro-2H-pyridine-1-carboxylate (6.0 g, 0.018 mol) in dimethyl sulphoxide (60 mL), palladium(0) tetrakis(triphenylphosphine) (0.84 g, 0.007 mmol) and zinc cyanide (3.81 g, 0.032 mol) was added. The reaction mixture was heated to 100° C. for 2 hr. The reaction mixture was diluted with water extracted with ethyl acetate. The organic layer was washed with water, followed by brine further dried over sodium sulphate and evaporated. The residue is purified by combi flash (eluting with 5% ethylacetate:n-hexane) to get 3.5 g of tert-butyl 4-cyano-3,6-dihydro-2H-pyridine-1-carboxylate. MS: m/z=209 (M+1).


c) Preparation of tert-butyl 4-cyano-3-methylsulfanyl-piperidine-1-carboxylate

To a solution of tert-butyl 4-cyano-3,6-dihydro-2H-pyridine-1-carboxylate (0.75 g, 0.0036 mol) in methanol (22 mL) added methanethiol sodium salt (0.29 g, 0.004 mol) at 0° C. and stirred at room temperature for 3 hrs. Methanol was evaporated under reduced pressure and diluted with ethyl acetate. Organic layer was washed with water, brine dried over sodium sulphate and evaporated. The residue was purified by combiflash (eluting with 10% ethylacetate:n-hexane) to give 393 mg of tert-butyl 4-cyano-3-methylsulfanyl-piperidine-1-carboxylate. MS: m/z=157 (M-100).


d) Preparation of 3-methylsulfanylpiperidine-4-carbonitrile hydrochloride

To a solution of tert-butyl 4-cyano-3-methylsulfanyl-piperidine-1-carboxylate (1.00 g, 0.003 mol) in methanol (4 mL) added 4 M hydrochloric acid in dioxane (8 mL) at 0° C. The reaction mixture was stirred at room temperature for 2 h. After completion of reaction (analysed by TLC) solvent was completely evaporated to afford 3-methylsulfanylpiperidine-4-carbonitrile hydrochloride salt that was used as such for the next step.


e) Preparation of 3-methylsulfanyl-1-[2-[5-methyl-3-(trifluoromethyl)pyrazol-1-yl]acetyl]piperidine-4-carbonitrile

To a solution of 2-[5-methyl-3-(trifluoromethyl)pyrazol-1-yl]acetic acid (2.75 g, 0.013 mol) in N,N-dimethylforamide (22 mL), 1-[bis(dimethylamino)methylene]-1H-1,2,3-triazolo[4,5-b]pyridiniumare 3-oxid hexafluorophosphate (5.27 g, 0.014 mol), 3-methylsulfanylpiperidine-4-carbonitrile hydrochloride (2.55 g, 0.013 mol) and triethyl amine (5.52 mL, 0.04 mol) at 0° C. was added and allowed to stirred at room temperature for 3 hrs. After completion of reaction, the reaction mixture was diluted with water extracted with ethyl acetate. The organic layer was washed with water, brine and dried over sodium sulphate. Organic layer was concentrated to dryness. The residue was washed with diethyl ether afforded 3.5 g of 3-methylsulfanyl-1-[2-[5-methyl-3-(trifluoromethyl)pyrazol-1-yl]acetyl]piperidine-4-carbonitrile.


MS: m/z=347 (M+1).


f) Preparation of 3-methylsulfanyl-1-[2-[5-methyl-3-(trifluoromethyl)pyrazol-1-yl]acetyl]piperidine-4-carbothioamide

To a solution of 3-methylsulfanyl-1-[2-[5-methyl-3-(trifluoromethyl)pyrazol-1-yl]acetyl]piperidine-4-carbonitrile (1.2 g, 0.003 mol) in N,N-dimethylformamide (7 mL) ammonium sulfide (1.9 g, 0.028 mmol) was added at room temperature and stirred for 24 hr. Reaction mixture was quenched with ice and extracted with ethyl acetate. The organic layer was washed with water, brine, dried over sodium sulphate and concentrated. The crude product was purified by combiflash (eluting with 30% ethyl acetate: dichloromethane) afforded 800 mg of 3-methylsulfanyl-1-[2-[5-methyl-3-(trifluoromethyl)pyrazol-1-yl]acetyl]piperidine-4-carbothioamide. MS: m/z=381 (M+1).


g) Preparation of (1E/Z)-3-chloro-N-hydroxy-2-oxo-propanimidoyl chloride

To a solution of 1,3-dichloropropan-2-one (3.0 g, 0.023 mol) in 2M hydrogen chloride in diethyl ether (15 mL) added tert-butyl nitrite (1.65 g, 0.016 mol) at 10° C. and stirred for 16 h at room temperature. Solvent was evaporated the obtained viscous material was triturated with 10% diethyl ether/n-hexane to get (1E/Z)-3-chloro-N-hydroxy-2-oxo-propanimidoyl chloride as light brown solid and that was used as such in next step.


h) Preparation of 2-chloro-1-[5-(2,6-difluorophenyl)-4,5-dihydroisoxazol-3-yl]ethanone

To a solution of (1E/Z)-3-chloro-N-hydroxy-2-oxo-propanimidoyl chloride (1.2 g, 0.007 mol) and 1,3-difluoro-2-vinyl-benzene (1.00 g, 0.007 mol) in acetonitrile (15 mL), sodium bicarbonate (1.19 g, 0.014 mol) was added in portion wise at 10° C. and stirred for 5 h at room temperature. Reaction mixture was diluted with ethyl acetate washed with water, brine dried over sodium sulphate. Organic layer was concentrated and residue was purified by combiflash (eluting with 10% ethyl acetate: n-hexane) afforded 2-chloro-1-[5-(2,6-difluorophenyl)-4,5-dihydroisoxazol-3-yl]ethanone. MS: m/z=260 (M+1).


i) Preparation of 1-[4-[4-[5-(2,6-difluorophenyl)-4,5-dihydroisoxazol-3-yl]thiazol-2-yl]-3-methylsulfanyl-1-piperidyl]-2-[5-methyl-3-(trifluoromethyl)pyrazol-1-yl]ethanone (compound I.az.007)

A solution of 3-methylsulfanyl-1-[2-[5-methyl-3-(trifluoromethyl)pyrazol-1-yl]acetyl]piperidine-4-carbothioamide (0.50 g, 0.001 mol) and 2-chloro-1-[5-(2,6-difluorophenyl)-4,5-dihydroisoxazol-3-yl]ethanone (0.30 g, 0.001 mol) in ethanol (15 mL) was heated to reflux for 5 hrs. Ethanol was evaporated, diluted with ethyl acetate, washed with water, brine, dried over sodium sulphate. Organic layer was concentrated and residue was purified by combiflash (eluting with 20-40% ethyl acetate: n-hexane) afforded 1-[4-[4-[5-(2,6-difluorophenyl)-4,5-dihydroisoxazol-3-yl]thiazol-2-yl]-3-methylsulfanyl-1-piperidyl]-2-[5-methyl-3-(trifluoromethyl)pyrazol-1-yl]ethanone (compound I.az.007) as mixture of diastereomer (280 mg). 1H-NMR (400 MHz, CDCl3): δ=1.80-2.40 (m, 8H), 3.30-3.45 (m, 2H), 3.60-3.90 (m, 4H), 4.10-4.20 (m, 1H), 4.50-4.70 (m, 1H), 4.90-5.10 (m, 1H), 5.15 (s, 1H), 6.00-6.15 (m, 1H), 6.33 (d, J=3.6 Hz, 1H), 6.92 (t, J=8.4 Hz, 2H), 7.30-7.40 (m, 1H), 7.68 (d, J=4 Hz, 1H).


MS: m/z=586.1 (M+1).


EXAMPLE 2
This example illustrates the preparation of 1-[4-[4-[5-(2,6-difluorophenyl)-4,5-dihydroisoxazol-3-yl]thiazol-2-yl]-3-methylsulfinyl-1-piperidyl]-2-[5-methyl-3-(trifluoromethyl)pyrazol-1-yl]ethanone (compound I.az.008) and 1-[4-[4-[5-(2,6-difluorophenyl)-4,5-dihydroisoxazol-3-yl]thiazol-2-yl]-3-methylsulfonyl-1-piperidyl]-2-[5-methyl-3-(trifluoromethyl)pyrazol-1-yl]ethanone (compound I.az.009)

To a solution of 1-[4-[4-[5-(2,6-difluorophenyl)-4,5-dihydroisoxazol-3-yl]thiazol-2-yl]-3-methylsulfanyl-1-piperidyl]-2-[5-methyl-3-(trifluoromethyl)pyrazol-1-yl]ethanone (compound I.az.007, 0.2 g, 0.003 mol) in chloroform (2 mL) sodium tungstate dihydrate (0.005 g, 0.02 mmol) and tetrabutylammonium hydrogen sulphate (0.01 g, 0.03 mmol) were added at −5° C. hydrogen peroxide (0.01 g, 0.4 mmol, 50% aqueous solution) was added and resulting reaction mixture was stirred for 20 min at −5° C. Reaction mixture was quenched with saturated sodiumthiosulphite solution and extracted with ethyl acetate. The organic layer was washed with water, brine, dried over sodium sulphate. The organic layer was concentrated. The crude product was purified by combiflash (eluting with 30-70% ethyl acetate/n-hexane and finally eluted with ethyl acetate for 10 min) to afford 30 mg of 1-[4-[4-[5-(2,6-difluorophenyl)-4,5-dihydroisoxazol-3-yl]thiazol-2-yl]-3-methylsulfinyl-1-piperidyl]-2-[5-methyl-3-(trifluoromethyl)pyrazol-1-yl]ethanone (compound I.az.008) as mixture of diastereomer and 38 mg of 1-[4-[4-[5-(2,6-difluorophenyl)-4,5-dihydroisoxazol-3-yl]thiazol-2-yl]-3-methylsulfonyl-1-piperidyl]-2-[5-methyl-3-(trifluoromethyl)pyrazol-1-yl]ethanone (compound I.az.009) as mixture of diastereomer.

This example illustrates the preparation of 2-[3,5-bis(difluoromethyl)pyrazol-1-yl]-1-[4-[4-[5-(2,6-difluorophenyl)-4,5-dihydroisoxazol-3-yl]thiazol-2-yl]-3-methylsulfanyl-1-piperidyl]ethanone (compound I.az.223)
a) Preparation of 1-[2-[3,5-bis(difluoromethyl)pyrazol-1-yl]acetyl]-3-methylsulfanyl-piperidine-4-carbonitrile

To a solution of 2-[3,5-bis(difluoromethyl)pyrazol-1-yl]acetic acid (1.10 g, 4.90 mmol) in N, N-dimethylforamide (9 mL) 1-[bis(dimethylamino)methylene]-1H-1,2,3-triazolo[4,5-b]pyridiniumare 3-oxid hexafluorophosphate (1.9 g, 5.10 mmol), 3-methylsulfanylpiperidine-4-carbonitrile hydrochloride (0.94 g, 4.90 mmol) and triethyl amine (2.1 mL, 15.0 mmol) was added at 0° C. and allowed to stirred at room temperature for 3 hrs. After completion of reaction, the reaction mixture was diluted with water extracted with ethyl acetate. The organic layer was washed with water, brine and dried over sodium sulphate. Organic layer was concentrated to dryness. The residue was washed with diethyl ether afforded 1.1 g of 1-[2-[3,5-bis(difluoromethyl)pyrazol-1-yl]acetyl]-3-methylsulfanyl-piperidine-4-carbonitrile. MS: m/z=365 (M+1).


b) Preparation of 1-[2-[3,5-bis(difluoromethyl)pyrazol-1-yl]acetyl]-3-methylsulfanyl-piperidine-4-carbothioamide

To a solution of 1-[2-[3,5-bis(difluoromethyl)pyrazol-1-yl]acetyl]-3-methylsulfanyl-piperidine-4-carbonitrile (1.0 g, 0.0027 mol) in N,N-dimethylformamide (6 mL) added ammonium sulfide (1.49 g, 0.021 mol) at room temperature and stirred for 24 hr. Reaction mixture was quenched with ice and extracted with ethyl acetate. The organic layer was washed with water, brine, dried over sodium sulphate and concentrated. The crude product was purified by combiflash (eluting with 30% ethyl acetate: dichloromethane) afforded 900 mg of 1-[2-10 [3,5-bis(difluoromethyl)pyrazol-1-yl]acetyl]-3-methylsulfanyl-piperidine-4-carbothioamide.


MS: m/z=399 (M+1).


c) Preparation of 2-[3,5-bis(difluoromethyl)pyrazol-1-yl]-1-[4-[4-[5-(2,6-difluorophenyl)-4,5-dihydroisoxazol-3-yl]thiazol-2-yl]-3-methylsulfanyl-1-piperidyl]ethanone (compound I.az.223)

A solution of 1-[2-[3,5-bis(difluoromethyl)pyrazol-1-yl]acetyl]-3-methylsulfanyl-piperidine-4-carbothioamide (0.20 g, 0.50 mmol) and 2-chloro-1-[5-(2,6-difluorophenyl)-4,5-dihydroisoxazol-3-yl]ethanone (0.10 g, 0.6 mmol) in ethanol (6 mL) was heated to reflux for 5 hrs. Ethanol was evaporated, diluted with ethyl acetate, washed with water, brine, dried over sodium sulphate. Organic layer was concentrated and residue was purified by combiflash (eluting with 20-40% Ethyl acetate: n-hexane) afforded 1-[4-[4-[5-(2,6-difluorophenyl)-4,5-dihydroisoxazol-3-yl]thiazol-2-yl]-3-methylsulfanyl-1-piperidyl]-2-[5-methyl-3-(trifluoromethyl)pyrazol-1-yl]ethanone (compound I.az.223) as mixture of diastereomer (64 mg). 1H-NMR (400 MHz, CDCl3): δ=1.90-2.40 (m, 6H), 2.80-3.20 (m, 1H), 3.20-3.42 (m, 2H), 3.60-3.95 (m, 3H), 4.20-4.40 (m, 1H), 5.20-5.30 (m, 2H), 6.00-6.20 (m, 1H), 6.50-7.10 (m, 5H), 7.30-7.40 (m, 1H), 7.70 (m, 1H)


MS: m/z=604.3 (M+1).

BIOLOGICAL EXAMPLES

Phytophthora infestans/Tomato/Leaf Disc Preventative (Tomato Late Blight)

Tomato leaf disks are placed on water agar in multiwell plates (24-well format) and sprayed with the formulated test compound diluted in water. The leaf disks are inoculated with a spore suspension of the fungus 1 day after application. The inoculated leaf disks are incubated at 16° C. and 75% rh under a light regime of 24 h darkness followed by 12 h light/12 h darkness in a climate cabinet and the activity of a compound is assessed as percent disease control compared to untreated when an appropriate level of disease damage appears in untreated check leaf disks (5-7 days after application).


Compounds I.az.007, I.az.008, I.az.009, I.az.223 at 200 ppm give at least 80% disease control in this test when compared to untreated control leaf disks under the same conditions, which show extensive disease development.

2-week old potato plants cv. Bintje are sprayed in a spray chamber with the formulated test compound diluted in water. Compounds I.az.007, I.az.223 at 200 ppm give at least 80% disease control in this test when compared to untreated control leaf disks under the same conditions, which show extensive disease development.

Grape vine leaf disks are placed on water agar in multiwell plates (24-well format) and sprayed with the formulated test compound diluted in water. The leaf disks are inoculated with a spore suspension of the fungus 1 day after application. The inoculated leaf disks are incubated at 19° C. and 80% rh under a light regime of 12 h light/12 h darkness in a climate cabinet and the activity of a compound is assessed as percent disease control compared to untreated when an appropriate level of disease damage appears in untreated check leaf disks (6-8 days after application).


Compounds I.az.007, I.az.008, I.az.009, I.az.223 at 200 ppm give at least 80% disease control in this test when compared to untreated control leaf disks under the same conditions, which show extensive disease development.

Pythium ultimum/Liquid Culture (Seedling Damping Off)

Mycelia fragments and oospores of a newly grown liquid culture of the fungus are directly mixed into nutrient broth (PDB potato dextrose broth). After placing a (DMSO) solution of test compound into a microtiter plate (96-well format), the nutrient broth containing the fungal mycelia/spore mixture is added. The test plates are incubated at 24° C. and the inhibition of growth is determined photometrically 2-3 days after application.


Compounds I.az.007, I.az.008, I.az.009, I.az.223 at 200 ppm give at least 80% disease control in this test when compared to untreated control leaf disks under the same conditions, which show extensive disease development.

Claims
  • 1. A compound of formula I:

A compound according to claim 1, wherein one of R3 and R5 is C1-C4alkylsulfanyl, C3-C5cycloalkylsulfanyl, C1-C4haloalkysulfanyl, C1-C4alkylsulfinyl, C3-C5cycloalkylsulfinyl, C1-C4haloalkylsulfinyl, C1-C4alkylsulfonyl, C3-C5cycloalkylsulfonyl, C1-C4haloalkylsulfonyl and R4, R6, R7 and R8 are hydrogen.
  • 3. A compound according to claim 1, one of R3 and R5 is C1-C4alkylsulfanyl, C1-C4alkylsulfinyl, C1-C4alkylsulfonyl and R4, R6, R7 and R8 are hydrogen.
  • 4. A compound according to claim 1, wherein the compound of formula I is a compound of formula I-a
  • 5. A compound according to claim 4, wherein Z is selected from Z-1, Z-4, Z-8, Z-10 and Z-12.
  • 6. A compound according to claim 4, wherein R14 and R15 are independently C1-C4alkyl, C1-C4haloalkyl or halogen.

A method of controlling or preventing an infestation of plants, propagation material thereof, harvested crops or of non-living materials by phytopathogenic or spoilage microorganisms or organisms potentially harmful to man, which comprises the application of a compound as defined in claim 1, to the plant, to parts of the plants or to the locus thereof, to propagation material thereof or to any part of the non-living materials.
