Claims
- 1. A rapamycin 42-ester with a dicarboxylic acid conjugate to a fluorescein derivative.
- 2. The conjugate of claim 1 wherein the dicarboxylic acid is succinic acid.
- 3. The conjugate of claim 2 wherein the derivative of fluorescein is 5-glycinylfluoresceinarnine, 5-aminomethylfluorescein, or 4′-amninomethylfluorescein.
- 4. The conjugate of claim 1 wherein the dicarboxylic acid is adipic acid.
- 5. The conjugate of claim 4 wherein the derivative of fluorescein is 5-glycinylfluoresceinamine, 5-aminomethylfluorescein, or 4′ -aminomethylfluorescein.
- 6. In an immunoassay method for determining levels of rapamycin or a derivative thereof, the improvement comprises using the conjugate of claim 1 as a detector molecule.
- 7. A test kit for measuring levels of rapamycin or a derivative thereof comprising the rapamycin conjugate of claim 1 and an antibody capable of specifically binding to rapamycin or a derivative thereof.
- 8. A secorapamycin 41-ester with a dicarboxylic acid conjugate to a fluorescein derivative.
- 9. The conjugate of claim 8 wherein the dicarboxylic acid is succinic acid.
- 10. The conjugate of claim 9 wherein the fluorescein derivative is 5-glycinylfluoresceinamine.
- 11. In an immunoassay method for determining levels of rapamycin or a derivative thereof, the improvement comprises using the conjugate of claim 8 as a detector molecule.
- 12. A test kit for measuring levels of rapamycin or a derivative thereof comprising the rapamycin conjugate of claim 8 and an antibody capable of specifically binding to rapamycin or a derivative thereof.
- 13. A monoclonal antibody specifically binding to a ring opened rapamycin which is designated as RAP42-OVAF2#1MoAb.
- 14. The monoclonal antibody of claim 13, wherein the ring opened rapamycin is secompamycin.
- 15. A test kit for determining levels of a ring opened rapamycin or a derivative thereof, the improvement comprises employing the antibody of claim 13.
- 16. A method of separating a mixture of a ring opened rapamycin or a derivative thereof from rapamycin or a derivative thereof, which comprises treating the mixture with the antibody of claim 13 bound to a solid matrix, and eluting the unbound rapamycin from the bound open chain rapamycin-antibody-solid matrix complex.
- 17. A method of separating a mixture of a ring opened rapamycin or a derivative thereof from rapamycin or a derivative thereof, which comprises treating the mixture with the antibody of claim 13, capturing the ring opened rapamycin-antibody complex with a solid support, and eluting the unbound rapamycin from the open chain rapamycin-antibody-solid matrix complex.
- 18. A method of separating a mixture of a ring opened rapamycin or derivative thereof from rapamycin or a derivative thereof, which comprises treating the mixture with the antibody of claim 13, followed by anti-mouse immunoglobulin, and removing the precipitated ring opened rapamycin-antibody-anti-mouse immunoglobulin complex.
- 19. The method according to claim 18, wherein the anti-mouse immunoglobulin is rabbit, mouse, rat, sheep, or goat antimouse immunoglobulin.
- 20. A method of separating a mixture of a ring opened rapamycin or a derivative thereof from rapamycin or derivative thereof, which comprises treating the mixture with the antibody of claim 13, followed by protein A or G, and removing the precipitated ring opened rapamycin-antibody-protein A or G complex.
- 21. A monoclonal antibody specifically binding to rapamycin which is designated as 34-294-163MoAb.
- 22. A hybridoma cell line capable of producing antibodies specific for rapamycin which is designated as 34-294-163hc.
